Summary: miR-346 induces rapid and extensive DNA damage in prostate cancer cells through transcriptional hyperactivation, R-loop formation, and replication stress. It disrupts the interaction between NORAD and PUM2, leading to PUM2 stabilization and increased turnover of DNA damage response transcripts. miR-346 downregulates target transcripts preferentially at binding sites of highly transcriptionally active transcription factors. Targeting miR-346 may enhance the sensitivity of prostate cancer cells to DNA-damaging drugs and induce tumor regression as a monotherapy.

Summary: This study utilized high-throughput sequencing to analyze the transcriptome of tiger pufferfish gonads, identifying numerous miRNAs and lncRNAs potentially involved in regulating sex-related gene expression. Summary: Long non-coding RNAs (lncRNAs) are a newly identified class of RNA molecules at least 200 nucleotides in length that are not translated into proteins. The lncRNA prostate cancer-associated transcript 6 (PCAT6) is upregulated in various human malignancies and is closely correlated with aggressive clinicopathological characteristics and poor prognosis in cancer patients, indicating it is an oncogenic lncRNA that could serve as a new prognostic biomarker and therapeutic target for malignancies. The molecular mechanisms, regulatory functions, and potential clinical applications of PCAT6 in cancer are discussed.

This review emphasizes the recent developments and advances in ncRNA biology relevant to glioma tumorigenesis and therapy response, highlighting the potential of ncRNAs as biomarkers and therapeutic agents in glioma.
