Tau oligomers accumulation sensitizes prostate cancer cells to docetaxel treatment

Summary: Human tau is a multifunctional protein involved in microtubule turnover, and tau oligomers are pathologic forms associated with protein accumulation diseases. In prostate cancer cells, autophagy inhibition led to tau oligomers accumulation and increased cytotoxic effect by docetaxel, indicating a potential role of autophagy in cancer growth and chemotherapy response. Therapeutic strategies targeting tau oligomers formation, such as autophagy inhibition, could be effective in cancer therapy.

Summary: In the last decade, the human helicase protein DDX3X has emerged as a promising target for anticancer drug development. A novel inhibitor, BA103, has shown broad-spectrum anticancer activity by targeting DDX3X's helicase binding site. BA103 exhibited good tolerability, pharmacokinetic properties, and potential for inhibiting tumor growth and migration, especially in glioblastoma models. This study demonstrates the feasibility and promise of DDX3X-targeted small molecules as potential drugs for treating various cancers, including glioblastoma.

Comparative Analysis of Dasatinib Effect between 2D and 3D Tumor Cell Cultures

Summary: Three-dimensional cell culture methods, particularly the 3D multicellular tumor spheroids model, enhance the predictive relevance of in vitro tumor models by better resembling tumor complexity and drug resistance. Recent advancements in 3D printing techniques and suitable biomaterials have provided new opportunities for developing reproducible and high-throughput 3D tissue cultures. In this study, the comparison of DU145 and U87 cancer cells cultured in the 3D spheroids model and the 3D bioprinting model showed that bioprinted cells demonstrated similar drug resistance to spheroids, suggesting a potential mechanism of drug resistance. The 3D bioprinted model utilizing alginate/gelatin hydrogel was proven to be a feasible, reproducible, and scalable model for drug screening when spheroid growth is required.

Summary: This study aimed to explore the expression levels and functional relationships between miR-182-5p and Cyld-Foxo1 in hepatic tissues from a diet-induced NAFLD/HCC mouse model. The results showed that miR-182-5p was upregulated during NAFLD progression and in tumors, and it inhibited the expression of tumor-suppressor genes Cyld and Foxo1 in HepG2 cells. Analysis of human HCC samples confirmed these findings and demonstrated the potential of miR-182-5p as a diagnostic biomarker or therapeutic target.
