The essential attributes of a solvent system for separation polar compounds on CCC are polarity, selectively and performance. Here, hydrophilic organic/salt-containing aqueous two-phase system (HO/S TPS) was evaluated as an alternative solvent system for CCC separation of polar compounds. Polarity measurements based on Rohrschneider-Snyder parameter was developed as quantitative assessing the polarity of HO/S TPS and comparing with an organic/aqueous system. All investigated 1-butanol/ethanol/saturated ammonium sulfate solution/water (BEAsWat) and 1-butanol/ethanol/saturated dipotassium hydrogen phosphate solution/water (BEDhpWat) systems with polarity values of organic phase from 4.5 to 6.8, were more polar than chloroform/methanol/water (1/1/1). The considerable water content of BEAsWat and BEDhpWat (0/1/1/1/) was 45.4 and 42.6% (w%) of hydrophilic organic phase, and 66.4 and 51.2% (w%) of salt-containing aqueous phase, respectively, closed to conventional aqueous two-phase system. Therefore, the polarity of HO/S TPS is in the middle of organic/aqueous and aqueous two-phase system. The LogK(C) values of twenty four polar compounds as model mixture confirmed that the polarities of HO/S TPSs were matched to that of the polar compounds and shown to be a very selective technique capable of separating positional isomers. Moreover, BEAsWat and BEDhpWat systems can be easily retained in CCC column with suitable elution mode. The hydrodynamic behavior reversion of HO/S TPS on hydrodynamic CCC was observed and was tentatively explained based on the density difference. Finally, caffeoylquinic acid isomers and dihydroxybenzoic acid isomers were successfully separated with HO/S TPS on CCC, respectively. Those results demonstrate that HO/S TPS on CCC is a performant and stable way to separate polar compounds from natural products. (C) 2016 Elsevier B.V. All rights reserved.

Summary: This study demonstrates that shading can alleviate the detrimental effects of Huanglongbing (HLB) on citrus trees. Through metabolomics and physiological approaches, the underlying metabolic mechanisms related to shade-induced morpho-physiological changes in citrus were investigated. Moderate shading prevents excessive starch accumulation, increases foliar sucrose levels, and promotes plant growth and fruit yield.

Effects of Different Rootstocks on the Metabolites of Huanglongbing-Affected Sweet Orange Juices Using a Novel Combined Strategy of Untargeted Metabolomics and Machine Learning

Xin Liu, Zhixin Wang, Frederick G. Gmitter Jr, Jude W. Grosser, Yu Wang

Summary: In this study, a combined strategy of untargeted metabolomics and machine learning was used to investigate the effects of rootstocks on the metabolites of HLB-affected sweet orange juices. A total of 2531 ion features were detected using high-resolution mass spectrometry, and 54 metabolites were identified as differential markers based on the random forest algorithm. The results revealed that rootstocks might regulate the physiological response against HLB by affecting glutamate, aspartate, proline, and arginine metabolism, as well as phenylalanine and phenylpropanoid metabolism. These findings provide new insights on the effects of rootstocks on the metabolites of HLB-affected sweet orange juices.

Integration of Volatilomics and Metabolomics Unveils Key Flavor-Related Biological Pathways in Different Carambola Cultivars

Jingwen Li, Zhixin Wang, Jonathan Crane, Yu Wang

Summary: This study investigated the contribution of volatile and non-volatile metabolites to the flavor variation of five carambola cultivars using a combination strategy of GC-MS/O-based volatilomics and LC-MS-based metabolomics. Several significant flavor-related pathways were identified, including amino acid biosynthesis or metabolism, terpenoids, fatty acids, sugar and organic acid, and flavonoids. The results showed that flavor traits of different carambola cultivars differed due to the up- or downregulation of metabolites in the flavor-related pathways. This study provides a valuable reference for breeders and researchers interested in the regulation of flavor.

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Jian Sun, Zheheng Zheng, Zixiao Jia, Jiabin Wang, Xucong Lin

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.
