Transcriptomic signature of high dietary organic selenium supplementation in sheep: A nutrigenomic insight using a custom microarray platform and gene set enrichment analysis

The objective of this study was to investigate the effect of a high dietary Se supplementation on the whole transcriptome of sheep. A custom sheep whole-transcriptome microarray, with more than 23,000 unique transcripts, was designed and then used to profile the global gene expression of sheep after feeding a high dietary supplementation of organic Se. Lactating crossbred ewes (n = 10; 3 to 4 yr of age and 55 to 65 kg BW) at late lactation (100 +/- 8 d in milk) were acclimated to indoor individual pen feeding of a basal control diet (0.40 mg Se/d, sodium selenite) for 4 wk. Sheep were then kept on a diet with an extra (high) supplementation of organic Se (1.45 mg Se/d as Sel-Plex; Alltech Biotechnology Pty Ltd, Dandenong, Victoria, Australia) for 40 d. Whole blood was collected at 2 time points (last day of the acclimatization period [T0] and after 40 d of the organic Se supplementation [T40]), and then total RNA was isolated and labeled for the subsequent microarray analysis. Significance Analysis of Microarrays, using the t-statistic, of the microarray data (T40 versus T0) evidenced the up-and downregulation of 942 and 244 transcripts (false discovery rate < 0.05), respectively. Seven genes showed the same trend of expression (up- or downregulation) when tested by quantitative real-time PCR (qPCR) in a cross-validation step. The microarray showed significant upregulation of the following selenoproteins at T40: selenium binding protein 1 (SELENBP1), selenoprotein W1 (SEPW1), glutathione peroxidase 3 (GPX3), and septin 8 (SEPT8). And the expression trends for SEPW1 and SEPT8 were validated using qPCR. Functional annotation of the differentially expressed genes showed the enrichment of several immune system-related biological processes (lymphocyte activation, cytokine binding, leukocyte activation, T cell differentiation, and B cell activation) and pathways (cytokine and interleukin signaling). Moreover, Gene Set Enrichment Analysis evidenced the enrichment of B and T cell receptors signaling pathways, with an enrichment score of 0.63 and 0.59, respectively. Overall, from a global gene expression (whole-transcriptome) point of view, short-term supplementation of a high dietary organic Se to Se-nondeficient sheep results in a transcriptomic signature that mainly reflects an induced immune system and a modulation of transcription effect. Also, the present study provides a custom whole-transcriptome microarray platform that can be used in further global gene expression studies in the ovine species.

Evaluation of Commercial Meat Products of Red Chicken Reared under LED Lights

Summary: This study investigated the effects of three different light-color temperatures of LEDs (Neutral, Cool, and Warm) on the attributes of chicken breast meat. The results showed that the LEDs caused changes in the physical and chemical characteristics of the meat. Specifically, significant variations were observed in the fatty acid profiles and accumulation of volatile compounds in the meat. These findings highlight the potential impact of LED lighting on the quality of chicken breast meat.

Qualitative attributes of meat from Teramana goat kids, an Italian native breed of the Abruzzo region

Qualitative Attributes of Commercial Pig Meat from an Italian Native Breed: The Nero d'Abruzzo

Summary: This study characterized the qualitative properties of commercial meat obtained from the Nero d'Abruzzo pig breed. The results showed that the Nero d'Abruzzo meat had higher fat content, higher concentration of alpha-linolenic acid, and higher Coenzyme Q10 content compared to meat from hybrid pigs. The Nero d'Abruzzo meat also exhibited better oxidative stability and reduced accumulation of hexanal.

Evaluation of Chemical-Nutritional Characteristics of Whey and Ricotta Obtained by Ewes Fed Red Grape Pomace Dietary Supplementation

Summary: This study investigated the effects of red grape pomace dietary supplementation on the chemical quality of whey and Ricotta obtained from ewes. The results showed that the supplementation did not affect the lipid content of whey and Ricotta, but significant variations were observed in color. The fatty acid profile of whey and Ricotta was modified, while the protein profile of whey protein remained unchanged. Overall, the inclusion of red grape pomace in the ewes' diet did not have negative effects on the characteristics and quality of dairy by-products.

Impact of different shades of light-emitting diode on fecal microbiota and gut health in broiler chickens

Nutritional Properties of Milk from Dairy Ewes Fed with a Diet Containing Grape Pomace

Seasonal and Feeding System Effects on Qualitative Parameters of Bovine Milk Produced in the Abruzzo Region (Italy)

Summary: This study aimed to investigate the variations in cow milk composition based on different breeding systems and seasonal changes. The results showed that milk samples from outdoor grazing had lower concentrations of saturated fatty acids and higher concentrations of rumenic, vaccenic, and oleic acids, as well as a greater presence of specific caseins. In contrast, intensive farming showed the opposite results. The concentration of retinol significantly increased during summer in both breeding systems.

Determination of hair cortisol in horses: comparison of immunoassay vs LC-HRMS/MS

Summary: Comparing the hair cortisol concentrations (HCC) of 47 horses under different managements, this study found that liquid chromatography coupled to hybrid high-resolution mass spectrometry (LC-HRMS/MS) provided more accurate results compared to the enzyme-linked immunosorbent assay (ELISA), which may overestimate the HCC. This finding is of significance in the development of accurate and reliable methods for monitoring stress in animals.

New Insight into the Quality Traits of Milk and Cheese from Teramana Goats, a Native Italian Breed

Summary: In this study, milk and cheese from the native Teramana goat breed were analysed and compared with those from the intensive Saanen breed. The Teramana breed showed higher levels of certain fatty acids, especially linoleic acid, in its milk and cheese. The Teramana cheeses also exhibited better oxidative stability during ripening due to lower levels of ketones and esters and higher levels of carboxylic acids. These findings highlight the importance of preserving and promoting native goat breeds.

Comparative Proteomics Analysis of Pig Muscle Exudate through Label-Free Liquid Chromatography-Mass Spectrometry

Summary: This study compared the proteomic differences between Nero d'Abruzzo and commercial-hybrid pigs, revealing significant differences in the protein expression of a large pool of proteins. Certain proteins, such as fatty acid synthase, catalase, and glutathione peroxidase, were found to be differentially expressed and may contribute to the phenotypic differences between the pigs. This work has identified potential proteins that could be useful for new applications in pig breeding programs.

Characterization of Phenolic Profile in Milk Obtained by Ewes Fed Grape Pomace: Reflection on Antioxidant and Anti-Inflammatory Status

Summary: The aim of this study was to evaluate the effect of grape pomace (GP) in the diet of dairy ewes on the content of phenolic compounds (PCs) in milk and its anti-inflammatory and antioxidative status. The results showed that the use of GP increased the concentration of phenolic compounds in milk, but had minimal impact on its antioxidant and inflammatory status.

Comparative Label-Free Liquid Chromatography-Mass Spectrometry Milk Proteomic Profiles Highlight Putative Differences between the Autochthon Teramana and Saanen Goat Breeds

Grape Pomace in Ewes Diet Affects Metagenomic Profile, Volatile Compounds and Biogenic Amines Contents of Ripened Cheese

Summary: The objective of this research was to evaluate the development of volatile organic compounds (VOCs) and the accumulation of biogenic amines (BAs) in cheese and their relationship with microbial population composition. The results showed that diet had an impact on the microbial composition and compound levels in the cheese, and there were significant positive correlations between certain bacterial families and compound formation.

Metagenomic Characterization and Volatile Compounds Determination in Rumen from Saanen Goat Kids Fed Olive Leaves

Summary: The aim of this study was to examine the effects of feeding olive leaves on the rumen microbiota and volatile organic compounds (VOCs) in Saanen goat kids. The results showed that feeding olive leaves influenced the microbial community in the rumen, increasing the abundance of cellulolytic bacteria. In addition, the analysis of VOCs in the rumen highlighted differences related to the diet and the presence of compounds indicative of health status.
