Identifying Molecular Changes in Early Cervical Cancer Samples of Patients That Developed Metastasis

The study aimed to identify biomarkers in early-stage primary cervical cancer that recurred after surgery. Several genes and immune cells were found to be differentially expressed in samples from patients who developed distant recurrence.
Cervical cancer is one of the most common cancers in women worldwide. Patients diagnosed with early-stage cervical cancer have a good prognosis, however, 10-20% suffer from local or distant recurrent disease after primary treatment. Treatment options for recurrent cervical cancer are limited. Therefore, it is crucial to identify factors that can predict patients with an increased risk of recurrence to optimize treatment to prevent the recurrence of cervical cancer. We aimed to identify biomarkers in early-stage primary cervical cancer which recurred after surgery. Formalin-Fixed, Paraffin-Embedded surgical specimens of 34 patients with early-stage cervical cancer (FIGO 2009 stage 1B1) and 7 healthy controls were analyzed. Targeted gene expression profiling using the PanCancer IO 360 panel of NanoString Technology was performed. The findings were confirmed by performing immunohistochemistry stainings. Various genes, namely GLS, CD36, WNT5a, HRAS, DDB2, PIK3R2, and CDH2 were found to be differentially highly expressed in primary cervical cancer samples of patients who developed distant recurrence. In addition, The relative infiltration score of CD8+ T cells, CD80+CD86+ macrophages, CD163+MRC1+ macrophages, and FOXP3+IL2RA+ regulatory T cells were significantly higher in this group of samples. In contrast, no significant differences in gene expression and relative immune infiltration were found in samples of patients who developed local recurrence. The infiltration of CD8 and FOXP3 cells were validated by immunohistochemistry using all samples included in the study. We identified molecular alterations in primary cervical cancer samples from patients who developed recurrent disease. These findings can be utilized towards developing a molecular signature for the early detection of patients with a high risk to develop metastasis.

Summary: Lung tissue from COVID-19 patients shows histomorphological features similar to chronic lung allograft disease, suggesting activation of autoimmune-related pathways. Analysis of mRNA expression in post-mortem lung tissue reveals significant up-regulation of mast cell-related genes in COVID-19. Genes associated with fibrosis and thrombosis are also up-regulated, indicating that mast cells may play a crucial role in the pathogenesis of COVID-19.

Unresectable Ovarian Cancer Requires a Structured Plan of Action: A Prospective Cohort Study

The median overall survival was 16 months, with 15% of patients alive at 24 months of follow-up.

Summary: This study investigated the impact of a single cycle of FOLFIRINOX in combination with a granulocyte colony-stimulating factor on the blood immune transcriptome of patients with pancreatic ductal adenocarcinoma. A novel eight-gene FFX-Delta GEP score was developed to predict the lack of FOLFIRINOX response after one cycle. The findings highlight the potential of using the peripheral immune transcriptome as an early circulating biomarker for predicting treatment outcomes.

Serous endometrial intra-epithelial carcinoma: an observational study

Summary: This study evaluated the surgical management of patients with serous endometrial intra-epithelial carcinoma and its impact on oncologic outcomes and complications. The results showed that median progression-free survival reached nearly 3 years and no recurrences have been reported in patients with pure serous endometrial intra-epithelial carcinoma. This suggests that full surgical staging might lead to overtreatment.

The efficacy of topical imiquimod in high-grade cervical intraepithelial neoplasia: A systematic review and meta-analysis

Summary: The aim of this study was to determine the efficacy of topical imiquimod in the treatment of high-grade CIN. The results showed a higher histological regression rate in the imiquimod treatment group compared to the placebo group, but lower than the surgical treatment group. Additionally, there was a difference in hr-HPV clearance rate between the imiquimod and surgical treatment groups.

Quantitative Proteomic Analysis of MCM3 in ThinPrep Samples of Patients with Cervical Preinvasive Cancer

Summary: By using a targeted proteomics approach, it was found that the concentration levels of MCM3 and EVPL proteins can discriminate between non-dysplastic and dysplastic samples, improving the accuracy of cervical cancer screening. This suggests that MCM3 and EVPL may be promising candidate protein biomarkers for population-based cervical cancer screening.

Tumor lineage-specific immune response in brain metastatic disease: opportunities for targeted immunotherapy regimen?

Summary: This study aimed to compare the brain immune response to brain metastasis between lung adenocarcinoma and breast cancer. The results showed a more active immune response in lung adenocarcinoma brain metastasis, with higher expression levels of VISTA and IDO1. Therefore, targeted immune therapy should be considered for patients with lung adenocarcinoma brain metastasis.

Plasma Device Functions and Tissue Effects in the Female Pelvis-A Systematic Review

Summary: This review discusses the historical and future applications of plasma in gynaecology and evaluates the depths of (non-)thermal effects in female pelvic tissues. The study found that the depth of thermal effects for plasma-assisted electrosurgical devices relied on current dispersion in tissue and was less than 2.4 mm, while the depth for electrically neutral argon plasma remained superficial (<1.0 mm). The depth and uniformity of cold atmospheric plasma effects require further investigation.

Patient-Reported Mobility, Physical Activity, and Bicycle Use after Vulvar Carcinoma Surgery

Summary: For vulvar cancer patients, walking and bicycling can cause discomfort and pain, leading to a decrease in mobility, physical activity, social contacts, self-reliance, and quality of life. We evaluated these problems using questionnaires and found that patients reported lower health, mobility, and physical activity compared to baseline values. Many experienced pain during bicycling and were unable to do it. This highlights the need for interventions to reduce discomfort and help patients regain their mobility and self-reliance.

Cytoreductive Surgery with the PlasmaJet Improved Quality-of-Life for Advanced Stage Ovarian Cancer Patients

Summary: This study aimed to determine whether the use of the PlasmaJet Surgical device during surgery has an effect on the quality-of-life of patients with advanced ovarian cancer. The results showed that using the PlasmaJet device during surgery leads to a higher quality-of-life compared to surgery with electrocoagulation alone. The differences in quality-of-life mainly relate to physical and role functioning, fatigue, and pain.

Summary: This retrospective cohort study analyzed the relationship between ultrasound features and the management of placenta accreta spectrum (PAS). The results showed significant correlations between certain ultrasound features and the choice of surgical approach. Placental bulge and higher grades of intracervical hypervascularity were associated with cesarean hysterectomy, while Grade 1 intracervical hypervascularity was associated with one-step conservative surgery (OSCS).
