In this study, an AI-based analysis pipeline was used to investigate the genetic factors associated with the size and function of the ascending aorta (AAo). The results showed that 78 loci were causally associated with aortic aneurysm development, but not with other vascular diseases.
Background Alterations in the anatomic and biomechanical properties of the ascending aorta (AAo) can give rise to various vascular pathologies. The aim of the current study is to gain additional insights in the biology of the AAo size and function. Methods We developed an AI based analysis pipeline for the segmentation of the AAo, and the extraction of AAO parameters. We then performed genome-wide association studies of AAo maximum area, AAo minimum area and AAo distensibility in up to 37,910 individuals from the UK Biobank. Variants that were significantly associated with AAo phenotypes were used as instrumental variables in Mendelian randomization analyses to investigate potential causal relationships with coronary artery disease, myocardial infarction, stroke and aneurysms. Findings Genome-wide association studies revealed a total of 107 SNPs in 78 loci. We annotated 101 candidate genes involved in various biological processes, including connective tissue development (THSD4 and COL6A3). Mendelian randomization analyses showed a causal association with aneurysm development, but not with other vascular diseases. Interpretation We identified 78 loci that provide insights into mechanisms underlying AAo size and function in the general population and provide genetic evidence for their role in aortic aneurysm development. Summary: This study aimed to analyze the association between clonal hematopoiesis of indeterminate potential (CHIP) and incident heart failure (HF) in a European population cohort. Results showed that CHIP mutations were correlated with age, smoking, hypertension, and cardiovascular biomarkers, but the frequency of CHIP was similar between individuals with incident HF and control participants. However, among people younger than 65 years, CHIP mutations were more frequently detected in the case cohort and significantly associated with new-onset HF.

The results showed that female gender, multivessel coronary artery disease, and micro-vascular damage were important predictors of adverse diastolic remodeling. Summary: Using multi-ancestry genome-wide association studies, this research identified 61 loci related to spatial QRS-T angle and 11 loci related to frontal QRS-T angle. These loci also showed shared genetic influences with classical ECG traits and were associated with cardiovascular diseases. The findings provide insights into the biology of QRS-T angles and their relationships with cardiovascular traits and diseases, which can inform future research and risk prediction.

Summary: In both CAD and non-CAD patients, EAT density, but not EAT volume, showed significant associations with PCAT(MA). Men had higher PCAT(MA) and EAT volume independently of disease status, but similar or slightly lower EAT density compared to women. Personalized interpretation and thresholding may be needed due to differences in trends and relations of PCAT(MA) and EAT by sex.

Summary: This study identifies new genetic variants associated with resting heart rate (RHR) and demonstrates that higher genetically predicted RHR is associated with a decreased risk of atrial fibrillation and ischemic stroke. Genome-wide analysis reveals multiple genetic variants in cardiomyocyte-related genes and provides insights into their electrocardiogram (ECG) signature. Mendelian randomization analyses indicate that higher genetically predicted RHR increases the risk of dilated cardiomyopathy, but reduces the risk of atrial fibrillation, ischemic stroke, and cardio-embolic stroke.

Selecting cardiac magnetic resonance images suitable for annotation of pulmonary arteries using an active-learning based deep learning model

Summary: The increasing and aging patient population presents challenges for healthcare professionals. Automation of medical imaging diagnostics using deep learning has the potential to improve patient care and reduce manpower requirements. However, labeled datasets are often laborious and costly to obtain in the medical field. Active learning, particularly dynamic querying, provides a method to optimize the selection of unlabeled samples for model improvement while minimizing the use of labeled images. This approach shows a faster learning curve and lower dataset size requirements compared to fully-supervised training methods.

Summary: This study identified 17 genetic variants associated with HRV traits, including 8 previously unreported loci. Additionally, lower HRV was associated with increased mortality risk. However, genetically predicted HRV was not associated with mortality.
