Summary: Currently, extracellular vesicles (EVs) are considered promising for cell-free therapies, but their effectiveness is limited by rapid clearance by the mononuclear phagocyte system (MPS). Strategies to avoid EV elimination are necessary for their clinical application. On the other hand, dysfunctional MPS is associated with immune-related pathologies, which could potentially be reversed by EVs through modulating macrophage phenotype or regulating antigen presentation by dendritic cells. Targeting EVs to MPS macrophages for replication and repackaging of their molecules into new vesicle subtype can enable specific targeting to desired cells. Further research on MPS-EV interactions is needed for the therapeutic use of EVs.

Summary: This study investigated the release of extracellular traps (ETs) by immune cells of Atlantic salmon, revealing significant stimulation of ET release by calcium ionophore and the fish pathogenic bacterium A. salmonicida. Microscopic observations showed bacteria associated with ETs, with reduced bacterial colonies recovered after co-incubation with neutrophils induced to release ETs. Interestingly, spontaneous ET release was inversely associated with fish mass, indicating a potential role in innate immunity and host-pathogen interaction in protecting against infectious diseases in cultured stocks.

Summary: This study investigates the effects of various methods for neutrophil characterization on neutrophil activation and degranulation levels. By comparing different methods, it is found that staining whole blood at low temperature and removing unbound antibodies prior to fixation and red blood cell lysis can minimize neutrophil activation, thus improving the consistency and reproducibility of neutrophil characterization.

Summary: Piscirickettsiosis outbreaks caused by Piscirickettsia salmonis in Chilean salmon aquaculture result in significant financial losses. P. salmonis secretes non-replicating and highly immunogenic outer membrane vesicles (OMVs). This study found that P. salmonis OMVs induce an immune response in Atlantic salmon, with the liver being the most immune-induced organ.
