This literature review discusses methods of modifying alginates and the importance of utilizing both natural and synthetic peptides for functionalization. It emphasizes the need for successful functionalization to ensure desired transformations while preventing degradation of alginates.
Modified alginates have a wide range of applications, including in the manufacture of dressings and scaffolds used for regenerative medicine, in systems for selective drug delivery, and as hydrogel materials. This literature review discusses the methods used to modify alginates and obtain materials with new or improved functional properties. It discusses the diverse biological and functional activity of alginates. It presents methods of modification that utilize both natural and synthetic peptides, and describes their influence on the biological properties of the alginates. The success of functionalization depends on the reaction conditions being sufficient to guarantee the desired transformations and provide modified alginates with new desirable properties, but mild enough to prevent degradation of the alginates. This review is a literature description of efficient methods of alginate functionalization using biologically active ligands. Particular attention was paid to methods of alginate functionalization with peptides, because the combination of the properties of alginates and peptides leads to the obtaining of conjugates with properties resulting from both components as well as a completely new, different functionality.

Additionally, this peptide was found to inhibit insulin hot spot aggregation, indicating its universal potential in inhibiting the aggregation process of hormones regulating carbohydrate metabolism. Ongoing research is focusing on the extensive use of this cyclic fragment as a peptide inhibitor in the polypeptide/protein aggregation process.

Dawid Maliszewski, Agnieszka Wrobel, Beata Kolesinska, Justyna Fraczyk, Danuta Drozdowska

Summary: A series of new analogs of nitrogen mustards containing the 1,3,5-triazine ring substituted with dipeptide residue were synthesized and evaluated for the inhibition of both acetylcholinesterase (AChE) and beta-secretase (BACE1) enzymes. The most active compounds against both AChE and BACE1 were identified as compounds A and 4a, with considerable inhibitory concentrations.

Synthesis and Properties of Targeted Radioisotope Carriers Based on Poly(Acrylic Acid) Nanogels

Malgorzata Matusiak, Beata P. Rurarz, Slawomir Kadlubowski, Marian Wolszczak, Urszula Karczmarczyk, Michal Maurin, Beata Kolesinska, Piotr Ulanski

Summary: Radiation crosslinking was used to create nanocarriers based on poly(acrylic acid) for targeted delivery of radioactive isotopes. The internally crosslinked hydrophilic macromolecules-nanogels were conjugated with bombesin-derivative-oligopeptide and tetraazacyclododecane-1,4,7,10-tetraacetate for selective radioligand transport. The efficiency of radiolabeling nanocarriers with Lu-177 was chromatographically tested, showing promise for theranostic applications.

Synthesis and Hemostatic Activity of New Amide Derivatives

Lukasz Banach, Lukasz Janczewski, Jakub Kajdanek, Katarzyna Milowska, Joanna Kolodziejczyk-Czepas, Grzegorz Galita, Wioletta Rozpedek-Kaminska, Ewa Kucharska, Ireneusz Majsterek, Beata Kolesinska

Summary: Eight dipeptides containing antifibrinolytic agents were successfully synthesized using a three-step process. Hemolysis tests and clotting tests showed that these compounds had no significant effects on blood components and clotting time. The clot formation and fibrinolysis assay revealed that two of the compounds exhibited high activity in the plasma coagulation cascade and fibrinolytic efficiency. The cytotoxicity and genotoxicity evaluation demonstrated that the synthesized dipeptides caused minimal damage to cells and DNA.

Isothiocyanates (ITCs) 1-(Isothiocyanatomethyl)-4-phenylbenzene and 1-Isothiocyanato-3,5-bis(trifluoromethyl)benzene-Aldehyde Dehydrogenase (ALDH) Inhibitors, Decreases Cisplatin Tolerance and Migratory Ability of NSCLC

Jolanta Kryczka, Jakub Kryczka, Lukasz Janczewski, Anna Gajda, Andrzej Fraczyk, Joanna Boncela, Beata Kolesinska, Ewa Brzezianska-Lasota

Summary: One of the main challenges in the treatment of non-small-cell lung cancer is the development of resistance to cisplatin-based chemotherapy. This study investigates the potential of isothiocyanates (ITCs) and the ALDH inhibitor disulfiram (DSF) as adjuvants to cisplatin chemotherapy. The results show that certain ITCs and DSF can reduce the cisplatin tolerance of resistant lung cancer cells, suggesting their potential in overcoming drug resistance.

Summary: New scaffold materials composed of biodegradable components are of great interest in regenerative medicine. In this study, peptide-polysaccharide conjugates were analyzed as scaffolds for regenerative medicine. The conjugates containing calcium alginate and the ternary nonwoven material were found to be the most supportive of muscle tissue regeneration. Peptides 1 and 2 exhibited the best biological properties and showed high antibacterial activity against Staphylococcus aureus and Klebsiella pneumoniae. These findings suggest that polysaccharide conjugates with peptides 1 and 2 have potential applications in regenerative medicine.
