Loss of miR-182 affects B-cell extrafollicular antibody response

MicroRNAs have been shown to play a role in B-cell differentiation and activation. Here, we found miR-182 to be highly induced in activated B cells. However, mice lacking miR-182 have normal B-cell and T-cell development. Interestingly, mutant mice exhibited a defective antibody response at early time-points in the immunization regimen when challenged with a T-cell-dependent antigen. Germinal centres were formed but the generation of extrafollicular plasma cells was defective in the spleens of immunized miR-182-deficient mice. Mutant mice were also not able to respond to a T-cell-independent type 2 antigen, which typically elicited an extrafollicular B-cell response. Taken together, the data indicated that miR-182 plays a critical role in driving extrafollicular B-cell antibody responses.

Summary: Cataract surgery can improve visual acuity for patients with retinitis pigmentosa during long-term follow-up. However, the surgery should not be performed on patients with invisible preoperative macular ellipsoid zone. Further studies are needed to address the issue of excessive light exposure to the degenerated retina in RP patients after cataract removal.

Mechanosensitive channel Piezo1 is an essential regulator in cell cycle progression of optic nerve head astrocytes

Summary: Optic nerve head (ONH) astrocytes play a crucial role in supporting neuronal axons through their mechanical sensitivity, mediated by the ion channel Piezo1. Knockdown or overexpression of Piezo1 in vitro revealed its necessity but insufficiency in ONH astrocyte proliferation. Loss of Piezo1 led to cell cycle arrest, possibly through decreased YAP nuclear localization and downregulation of cell cycle-associated factors. Gene ontology analysis highlighted the impact of Piezo1 on cell division processes. These findings establish Piezo1 as an essential regulator of cell cycle progression in ONH astrocytes.

SLAMF7/STAT6 Pathway Inhibits Innate Immune Response in Late-Stage Human Acanthamoeba Keratitis: A Comparative Transcriptome Analysis

Summary: Acanthamoeba keratitis is an eye infection caused by the protozoan Acanthamoeba. Transcriptome analysis revealed differentially expressed genes and enriched signaling pathways related to leukocyte migration, T-cell activation, extracellular matrix, immune receptor activity, and cytokine binding in cornea tissues of AK patients. The immune infiltration profiles showed minimal differences between AK and normal controls, except for a higher proportion of M0 macrophages and CD8 T cells in AK cornea tissues.

Immunomodulatory properties of mesenchymal stem cells: A potential therapeutic strategy for allergic rhinitis

Summary: Allergic rhinitis is a common chronic inflammatory disorder that significantly impacts patients' health and quality of life. Current treatments for allergic rhinitis are limited in their ability to restore immune balance and target specific allergens. Mesenchymal stem cells (MSCs) offer a potential therapeutic strategy for allergic rhinitis due to their immune privilege and strong immunomodulatory effects. Recent studies have investigated the use of MSCs in animal models of allergic rhinitis and have shown promising results. This review discusses the immunomodulatory effects of MSCs on allergic airway inflammation, highlights recent research on the role of MSCs in modulating immune cells, and explores the clinical potential of MSC-based therapy for allergic rhinitis.

Mutational investigation of 17 causative genes in a cohort of 113 families with nonsyndromic early-onset high myopia in northwestern China

Summary: We investigated genetic variations in a cohort of 113 families with nonsyndromic early-onset high myopia from northwestern China and identified sixteen potentially pathogenic variants in eight of seventeen causative genes for high myopia. These newly identified genetic variations expand the genetic and clinical spectra of high myopia and provide evidence for the contribution of ARR3, NDUFAF7, TNFRSF21, and ZNF644 genes to hereditable high myopia.

REG1A protects retinal photoreceptors from blue light damage

Summary: With the increasing use of artificial light and optoelectronic products, light damage (LD) to the human retina has become a global issue. This study investigated the mechanism of how blue light leads to photoreceptor death and found that regenerating islet-derived 1 alpha (REG1A) prevents LD-induced apoptosis in photoreceptor cells by increasing the expression of the anti-apoptotic gene Bcl2 and decreasing the expression of the pro-apoptotic gene Bax. REG1A restores mitochondrial function and cell apoptosis after LD-induced damage, suggesting its potential application in the prevention and treatment of retinal vision loss.

Acute onset of decreased vision after violently vomiting: A case report

Retinal organoid and gene editing for basic and translational research

Summary: The advanced technologies of retinal organoids (ROs) and gene editing have revolutionized the field of mammalian retina research. ROs can be generated using various induction methods to mimic in vivo retinal development and closely resemble the molecular and cellular profiles of the retina. Gene editing techniques, including CRISPR-Cas9 editing and its derivatives, have further enhanced the possibilities in studying retinal development, pathogenesis, and therapeutics. This review discusses recent advances in ROs, gene editing methodologies, delivery vectors, and their implications in retinal studies.

CRX haploinsufficiency compromises photoreceptor precursor translocation and differentiation in human retinal organoids

Summary: We created monoallelic CRX by replacing one allele with tdTomato in human embryonic stem cells (hESCs) and studied the pathogenesis in hESCs-derived retinal organoids. We found that gene haploinsufficiency is the mechanism for the dominant pathogenicity of CRX, and observed the translocation of monoallelic CRX+ cells in early retinal differentiation.

Bietti's crystalline dystrophy: genotyping and deep qualitative and quantitative phenotyping in preparation for clinical trials

Summary: This five-year cohort study explores the relationship between color vision deficiency and myopia progression in Chinese primary school children. The results suggest that children with color-vision deficiency have a lower incidence and slower progression of myopia.
