Hydroxytyrosol modulates Par j 1-induced IL-10 production by PBMCs in healthy subjects

Several papers have demonstrated the importance of substances from natural sources which can exert powerful anti-inflammatory effects. In this respect, hydroxytyrosol, one of the major elements of the phenolic components of olive oil, has been extensively studied for its anti-inflammatory activities and safety profile. In this report, we demonstrate that the co-stimulation of human PBMCs from healthy subjects with the Par j 1 allergen and hydroxytyrosol induced a statistically significant increase in the amount of Par j 1-induced IL-10, demonstrating that hydroxytyrosol can modulate an allergen-specific immune response potentiating a suppressive immune response towards an allergen. Our work opens the way to further studies to elaborate the possibility of using hydroxytyrosol as a nutrient for allergy prevention. (C) 2016 Elsevier GmbH. All rights reserved.

Summary: This study aimed to define an in vitro model to test the effect of primed hAMSCs in reducing immune and hepatic cell damage caused by IRI. The results showed that IFN-gamma pre-treatment and 3D culture significantly reduced inflammation induced by M1-differentiated macrophages, and primed hAMSCs inhibited parenchymal apoptosis at early reperfusion timepoints by blocking the activation of caspase 3/7.

PM2.5 elemental composition in indoor residential environments and co-exposure effects on respiratory health in an industrial area

Summary: This study aimed to identify and characterize indoor sources of particulate matter (PM) in domestic environments. The study evaluated 74 inhabited apartments in urban and rural areas of Gela, Italy for the elemental composition of PM2.5. The results showed that indoor air in the apartments was affected by both outdoor pollution and specific indoor emission sources, such as soil, industrial sources, heavy oil combustion, cigarette smoke, and use of vacuum dust cleaners. The study also found that cigarette smoke and heavy oil combustion increased health risks.

Impact of the flame retardant 2,2'4,4'-tetrabromodiphenyl ether (PBDE-47) in THP-1 macrophage-like cell function via small extracellular vesicles

Summary: This study investigates the effects of PBDE-47 treatment on the miRNA cargo of sEVs and their downstream effects on bystander macrophages. The sEVs purified from PBDE-47 treated cells were found to modulate the expression of 12 miRNAs in resting THP-1 cells. Furthermore, PBDE-47 treatment may impair immune activity and down-regulate differentiation markers in macrophages. These findings suggest that PBDE-47 perturbs cellular regulatory pathways and affects macrophage differentiation markers by altering miRNA cargo.

Relationship among Body Composition, Adipocytokines, and Irisin on Exercise Capacity and Quality of Life in COPD: A Pilot Study

Summary: Adipose tissue is an endocrine organ that affects the severity of COPD. The role of adipocytokines and myokines in COPD is still controversial. This study evaluated the impact of body composition, leptin, adiponectin, haptoglobin, and irisin on exercise capacity, respiratory function, and QoL in COPD patients. The results showed increased levels of leptin, total adiponectin, and haptoglobin in COPD patients, along with decreased 6MWT and physical functioning scores. There were significant relationships between leptin/fat mass and 6MWT/nutritional status. Overall, this pilot study further supports the influence of body composition and adipocytokines on exercise capacity, respiratory function, and QoL in COPD.

Summary: Ischemia/reperfusion injury (IRI) is a harmful process that occurs when blood flow is interrupted and restored, causing cell death and tissue dysfunction. Current therapies are not effective, creating a need for new approaches. Emerging evidence shows that mesenchymal stem/stromal cells (MSCs) can reduce inflammation and promote tissue repair through paracrine effects. This review explores the potential use of MSCs and their secretome for treating IRI in various tissues/organs.

Summary: This study investigated the effects of priming on the miRNA expression of MSCs and their exosomes. The results showed that different priming conditions led to distinct miRNA expression patterns in both MSCs and exosomes, but there were common deregulated miRNAs related to tissue repair/regeneration. The findings suggest that both 3D culture and IFN-gamma treatment can enhance the therapeutic potential of MSCs by modulating miRNA expression. Overall, this study provides valuable insights into the molecular mechanisms underlying the miRNA-mediated beneficial effects of MSCs.

Studies have shown that COVID-19 patients with ARDS treated with ECMO have a similar survival rate to those with non-COVID-19-related ARDS. This study examined the changes in the transcriptomic BCR repertoire in COVID-19 patients at different stages using single-cell V(D)J and gene expression sequencing.

Neurological Screening in Elderly Liver Transplantation Candidates: A Single Center Experience

Summary: This study aimed to determine whether cerebral small vessels disease (cSVD) predicts acute neurological complications (NCs) in liver transplant patients aged 65 years or older. The results showed that more severe cSVD was associated with seizures, longer hospital stay, and disability. Hepatic encephalopathy and ascites were predictors of NCs after liver transplantation.
