Proteomic fertility markers in ram sperm

This study compared the proteomic profiles of ram sperm with contrasting fertility phenotypes, suggesting potential protein markers for evaluation of semen quality and estimation of ram sperm fertilizing capacity. The differential abundances of sperm proteins between high and low-fertility rams indicate functional implications of the sperm proteome on male fertility.
Precise estimation of ram fertility is important for sheep farming to sustain reproduction efficiency and profitability of production. There, however, is no conventional method to accurately predict ram fertility. The objective of this study, therefore, was to ascertain proteomic profiles of ram sperm having contrasting fertility phenotypes. Mature rams (n = 66) having greater pregnancy rates than average (89.4 +/- 7.2%) were assigned into relatively-greater fertility (GF; n = 31; 94.5 +/- 2.8%) whereas those with less-than-average pregnancy rates were assigned into a lesserfertility (LF; n = 25; 83.1 +/- 5.73%; P = 0.028) group. Sperm samples from the outlier greatestand least-fertility rams (n = 6, pregnancy rate; 98.4 +/- 1.8% and 76.1 +/- 3.9%) were used for proteomics assessments utilizing Label-free LC-MS/MS. A total of 997 proteins were identified, and among these, 840 were shared by both groups, and 57 and 93 were unique to GF and LF, respectively. Furthermore, 190 differentially abundant proteins were identified; the abundance of 124 was larger in GF while 66 was larger in LF rams. The GF ram sperm had 79 GO/pathway terms in ten major biological networks while there were 47 GO/pathway terms in six biological networks in sperm of LF rams. Accordingly, differential abundances of sperm proteins between sperm of GF and LF rams were indicative of functional implications of sperm proteome on male fertility. The results of this study emphasize there are potential protein markers for evaluation of semen quality and estimation of ram sperm fertilizing capacity.

Summary: The study aimed to establish a suitable ram semen extender model by examining different combinations of glycerol concentrations, trehalose, and boron to prevent sperm deterioration and investigate gene expressions. Results showed that supplementation of 0.25 mm boron improved sperm parameters and acrosome integrity, while high boron concentrations had adverse effects on sperm parameters and antioxidant gene expression after thawing. The use of sufficient boron can decrease cryodamages during sperm cryopreservation and tissue engineering.

Summary: This study evaluated the biochemical, pharmacological, and toxicological aspects of Capparis ovata water extract (COWE). The results showed that COWE is rich in minerals, unsaturated fatty acids, and flavonoids/phenolics, and has high radical-scavenging ability. It significantly inhibits inflammatory responses and modulates immune responses through gene expression regulation.

Summary: In this study, the therapeutic effect of a compound called OPCA on multiple sclerosis (MS) was investigated using a preclinical model. The results demonstrated that OPCA protected the mouse brain, maintained blood-brain barrier integrity, and reduced inflammation. Furthermore, OPCA increased the expression of genes involved in myelin development and structure, and reduced the release of pro-inflammatory cytokines. Therefore, OPCA is a promising and appealing candidate for treating human MS.

Summary: In this study, a novel strain of Pseudomonas mandelii, named KGI_MA19, was identified and characterized from sediment samples of King George Island in Antarctica. This psychrotolerant and halotolerant strain exhibited the ability to use disaccharides as a carbon source, denitrification ability, and multiple antibiotic resistance. Comparative proteomic analyses revealed the adaptive mechanisms of P. mandelii KGI_MA19 in the cold environment.

Does using microfluidic sperm sorting chips in bovine IVEP affect blastocyst development?

Summary: This study determined the effect of microfluidic sperm sorting chip on embryo development and quality in the sperm treatment step in in vitro embryo production in cattle. The study found that using the microfluidic sperm sorting chip increased the rate of reaching the blastocyst, embryo development, and quality in the cattle. It also reduced the possibility of apoptosis in developing blastocysts.

Effect of putrescine supplementation to in vitro maturation medium on embryo development and quality in cattle

Summary: This study investigated the effect of putrescine supplementation in the maturation medium during in vitro embryo production in cattle. The results showed that putrescine supplementation increased the degree of cumulus expansion, the rate of resumption of meiosis, and the rate of reaching the blastocyst stage in oocytes. The addition of putrescine also led to better cell development in embryos.

Development of an ID-LC-MS/MS method using targeted proteomics for quantifying cardiac troponin I in human serum

Summary: This study aimed to develop a method traceable to the International System of Units for accurately measuring cardiac cTnI levels in human serum. The method involved immobilizing anti-cTnI antibodies onto magnetic nanoparticles, isolating cTnI from serum, and quantifying it using isotope dilution liquid chromatography-tandem mass spectrometry. The developed method showed high analytical performance in accurately quantifying cTnI and has the potential to harmonize cTnI results between clinical laboratories.

Summary: This study investigates the role of TNF-related apoptosis-inducing ligand (TRAIL) in macrophage polarization. It shows that TRAIL promotes the polarization of human macrophages towards a proinflammatory M1 phenotype via both DR4 and DR5, suggesting that targeting DRs can enhance the anti-tumorigenic response of macrophages in the tumor microenvironment by increasing M1 polarization.
