Prevention of allergic conjunctivitis in mice by a rice-based edible vaccine containing modified Japanese cedar pollen allergens

Background/aims To determine whether oral immunotherapy with transgenic rice seeds expressing hypoallergenic modified antigens suppresses cedar pollen-induced allergic conjunctivitis by eliciting immune tolerance in mice. Methods BALB/c mice were fed once a day for 20 days with 220 mg of transgenic rice expressing modified Japanese cedar pollen allergens Cry j 1 and Cry j 2 or with non-transgenic rice seeds as a control. They were then sensitised with two intraperitoneal injections of Japanese cedar pollen in alum before challenge twice with pollen in eye drops. Twenty-four hours after the second challenge, the conjunctiva, spleen, and blood were isolated for histological analysis, cytokine production assays, and measurement of serum immunoglobulin E concentrations, respectively. Results The numbers of eosinophils and total inflammatory cells in the conjunctiva were significantly lower in mice fed the transgenic rice than in those fed non-transgenic rice. The clinical score evaluated at 15 min after antigen challenge was also significantly lower in mice fed the transgenic rice than in those fed non-transgenic rice. The serum concentrations of both total and allergen-specific immunoglobulin E were also significantly lower in mice fed the transgenic rice. Oral vaccination with transgenic rice resulted in significant down-regulation of the allergen-induced production of interleukin (IL)-2, IL-4, IL-5, IL-12p70, interferon-gamma, and IL-17A by splenocytes. Conclusions Oral immunotherapy with transgenic rice expressing modified Japanese cedar pollen allergens suppressed pollen-induced experimental allergic conjunctivitis in mice by eliciting immune tolerance. This novel prophylactic approach is potentially safe and effective for allergen-specific oral immunotherapy in allergic conjunctivitis.

Clinical profile and visual outcome of intraocular inflammation associated with cat-scratch disease in Japanese patients

Summary: In this study, evaluation of intraocular inflammation in Japanese patients with cat-scratch disease revealed that white retinal or retinochoroidal foci were the most common manifestations in the posterior segment of the eye, suggesting a possible diagnosis of the disease. Symptoms such as high fever, decreased visual acuity, retinitis, and macular star may also be associated with ocular complications of cat-scratch disease.

Biologics for allergy: therapeutic potential for ocular allergic diseases and adverse effects on the eye

Summary: The use of biologics targeting specific antibodies and receptors has significantly improved the treatment outcomes in allergic diseases. However, these drugs have not been approved for ocular allergic diseases. While some studies have reported beneficial effects, adverse ocular symptoms have also been observed. Different biologics may have specific effects on different ocular allergies, and allergists must fully understand the effects of each drug on the eye. This review outlines the potential therapeutic and adverse effects of various biologics on allergic diseases of the eye.

Pivotal Role of Corneal Fibroblasts in Progression to Corneal Ulcer in Bacterial Keratitis

Summary: The shape and transparency of the cornea are crucial for vision clarity, but its susceptibility to pathogenic microorganisms can lead to bacterial keratitis and potentially corneal ulcers. Timely diagnosis and treatment are essential for successful management of bacterial keratitis, as progression to corneal ulcer can cause collagen degradation and serious complications. In corneal ulcer, corneal fibroblasts play a central role in collagen destruction, with a cascade of reactions involving uPA, plasmin, MMPs, interleukin-8, and ICAM-1 contributing to tissue damage and inflammation.

Purification of membrane vesicles from Gram-positive bacteria using flow cytometry, after iodixanol density-gradient ultracentrifugation

Summary: In this study, high-performance flow cytometry was used to analyze membrane vesicles (MVs) from Gram-positive bacteria, Staphylococcus aureus and Bacillus subtilis, showing that flow cytometric sorting provided higher purity and uniformity compared to gradient isolation alone. The MV purification method using flow cytometry should be useful for applications requiring very high purity of MV samples such as proteomic, metagenomic, or lipidomic studies.
