Role of Extracellular Vesicles on Cancer Lymphangiogenesis and Lymph Node Metastasis

Studies have shown that various cells in the tumor microenvironment release extracellular vesicles (EVs) encapsulating different molecules, which can be taken up by lymph endothelial cells (LECs) to regulate tumor lymphangiogenesis. EVs may serve as biomarkers for tumor lymph node metastasis and hold potential for anti-tumor lymphangiogenesis therapy.
Lymph node metastasis (LNM) of tumors is an established indicator of poor prognosis in patients. Tumor-associated lymphangiogenesis is a key step in LNM and has gained much attention. However, currently, there is no anti-tumor lymphangiogenesis drug used in clinical practice. Recently, studies on extracellular vesicles (EVs) have shown that different types of cells in the tumor microenvironment can release EVs that encapsulate a variety of molecules, including proteins, nucleic acids, and metabolites. Lymph endothelial cells (LECs) regulate tumor lymphangiogenesis through the uptake of EVs packed with different biologically active contents. In this review, we will discuss the possible mechanisms by which EVs participate in the regulation of tumor-associated lymphangiogenesis and LNM, summarize the potential value of EVs that can be used as biomarkers for the determination of tumor LNM, and indicate the potential anti-tumor lymphangiogenesis therapy.

Small extracellular vesicles containing miR-192/215 mediate hypoxia-induced cancer-associated fibroblast development in head and neck squamous cell carcinoma

Summary: This study revealed the mechanisms by which hypoxic cancer cells induce CAF-like differentiation through secreting sEVs and TGF-beta, with miRNAs delivered by sEVs playing a crucial role in this process. Elevated levels of miR-192/215 in HNSCC tissue-derived sEVs indicated a hypoxic and aggressive cancer stroma, and miR-215 in tumor tissue-derived sEVs was associated with poor overall survival in HNSCC patients. The findings suggest that sEVs serve as a messenger to transfer miRNAs from cancer cells to fibroblasts, contributing to the remodeling of the hypoxic tumor microenvironment and indicating the potential of cancer tissue-derived sEVs as biomarkers.

Oral Tongue Cancer in a Patient with Fanconi Anemia: A Case Report and Literature Review

Summary: This case report describes a patient with Fanconi anemia who developed oral tongue cancer and experienced severe bone marrow failure after receiving cisplatin/radiation treatment. Genetic testing revealed novel mutations in the FANCA gene inherited from the patient's parents. Despite successful symptom relief with reduced-dose radiotherapy, the patient eventually died of disease progression.
