Summary: Epigenetic mechanisms, particularly DNA methylation, are considered to be important in autoimmune and chronic inflammatory diseases, connecting environmental stimuli with genetic susceptibility. The causal relationship between DNA methylation and gene silencing is well established, but there is controversy in comparing DNA methylation results from different studies. Intriguing evidence comes from comparing the epigenome in clinically discordant monozygotic twins.

Summary: This study investigated the functional consequences of the single-nucleotide polymorphism rs4648889 in a putative enhancer upstream of the RUNX3 promoter associated with susceptibility to ankylosing spondylitis (AS). The results indicated that IRF5 plays a crucial role in the functions of CD8+ lymphocytes, suggesting new therapeutic targets for managing AS.

Perspectives on the Genetic Associations of Ankylosing Spondylitis

Summary: Ankylosing spondylitis is a common form of inflammatory spinal arthritis with a complex polygenic etiology. Genetic associations with AS have been identified in regions involved in antigen presentation, Th17 responses, and macrophages/T-cells. Many associations are found in intergenic regions, reflecting functional polymorphisms regulating gene expression. Understanding these associations may lead to better insights into the pathogenic pathways of AS and potential targets for drug therapy.

Summary: The efficacy and safety of the Moderna-1273 mRNA vaccine were evaluated in patients with immune-mediated diseases under different treatments. The study found that mycophenolate treatment may impair vaccine immunogenicity, and higher doses were associated with a lower humoral response. These findings suggest the need to consider the effects of different immunotherapies when selecting a vaccination regimen.
