Drosha, DGCR8, and Dicer mRNAs are down-regulated in human cells infected with dengue virus 4, and play a role in viral pathogenesis

Dengue virus (DENV) and its four serotypes (DENV1-4) belong to the Flavivirus genus of the Flaviviridae family. DENV infection is a life-threatening disease, which results in up to 20,000 deaths each year. Viruses have been shown to encode trans-regulatory small RNAs, or microRNAs (miRNAs), which bind to messenger RNA and negatively regulate host or viral gene expression. During DENV infections, miRNAs interact with proteins in the RNAi pathway, and are processed by ribonucleases such as Dicer and Drosha. This study aims to investigate Drosha, DGCR8, and Dicer expression levels in human A-549 cells following DENV4 infection. DENV4 infected A-549 cells were collected daily for 5 days, and RNA was extracted to quantify viral load. Gene expression of Drosha, Dicer, and DGCR8 was determined using quantitative PCR (RT-qPCR). We found that DENV4 infection exhibited the highest viral load 3 days post-infection. Dicer, Drosha, and DGCR8 showed reduced expression following DENV4 infection as compared with negative controls. In addition, we hypothesize that reduced expression of DGCR8 may not only be related to miRNA biogenesis, but also other small RNAs. This study may change our understanding regarding the relationship between host cells and the dengue virus.

Summary: This study characterized and analyzed the genetic diversity of Mycobacterium tuberculosis Central Asian Strain (CAS) Lineage 3 (L3) genotype in northern regions of Brazil. The results confirmed the presence of a novel genotype (CAS/SIT2545) belonging to L3. Further analysis showed genetic differences between the Brazilian strains and global strains. This highlights the importance of studying the genetic diversity of Mycobacterium tuberculosis in northern regions of Brazil.

Sequencing and description of the complete mitochondrial genome of Limatus durhamii (Diptera: Culicidae)

Summary: This study reports the first complete sequencing of the mitochondrial genome of Limatus durhamii Theobald, 1901. The phylogeny reconstruction based on the concatenation of all 13 protein-coding genes further validates the taxonomic classification of this genus. These findings are important for the taxonomic and evolutionary studies of this genus and the Culicidae family as a whole.

The immune response involves the expression of various markers, cytokines, and Treg cells.

Summary: Viruses such as Saint Louis encephalitis virus, Venezuelan equine encephalitis virus, Eastern equine encephalitis virus, Western equine encephalitis virus, Dengue virus, Zika virus, Chikungunya virus, Mayaro virus, and West Nile virus can cause neurological conditions of clinical and epidemiological importance. This study aimed to determine the number of arboviruses with neuroinvasive potential isolated in Brazil from 1954 to 2022. A total of 1,347 arbovirus samples were isolated from mice, 5,065 human samples were isolated exclusively by cell culture, and 676 viruses were isolated from mosquitoes. The detection of circulating arboviruses with neuroinvasive potential justifies the need for continued active epidemiological surveillance in Brazil.

Summary: In this study, histopathological analysis of liver samples from 70 patients who died due to infections and complications of yellow fever, dengue fever, and chikungunya fever was conducted. The results showed significant differences in histopathological alterations between the control and infection groups, with a predominance of changes in the midzonal area in all three cases analyzed. Hepatic involvement in yellow fever cases showed more severe histopathological changes.

Summary: The largest outbreak of sylvatic yellow fever virus (YFV) in Brazil between 2016-2018 was reported, and entomo-virological surveillance was considered important for monitoring YFV. Mosquitoes were collected from six Brazilian states and tested for YFV using RT-qPCR. YFV infection was detected in various mosquito species, including Ae. albopictus, indicating the potential for urban YFV re-emergence. The study highlights the need to strengthen YFV surveillance, vaccination coverage, and vector control measures in Brazil.

Emergence of a New Strain of DENV-2 in South America: Introduction of the Cosmopolitan Genotype through the Brazilian-Peruvian Border

Summary: In this study, 163 human serum samples from the DENV outbreak in Acre, Northern Brazil between 2020 and 2021 were tested for DENV genotypes by RT-qPCR. Of these samples, 139 were positive for DENV-2, and 5 were positive for DENV-1. Sequencing of 5 DENV-2-positive samples from early 2021 revealed clustering with previously recorded DENV-2 cosmopolitan genotype sequences in South America. These findings suggest a possible route of introduction of the DENV-2 cosmopolitan genotype into Brazil from Peru, followed by dispersal to the Midwest region.

Summary: This study identified six antigenic variants in rabies surveillance in northern and northeastern Brazil through phylogenetic analysis. The discovery of the AgV6 variant in northern Brazil for the first time provides valuable information for rabies surveillance in the country. Further genomic surveillance studies are needed to understand the transmission dynamics and geographic distribution changes of each variant.

Replication of Rocio virus in primary cultures of mouse neural cells

Summary: This study demonstrated the efficient replication of the Rocio virus in primary cultured neural cells from mice. The infection was confirmed through immunocytochemistry and viral replication was quantified. The results suggest that mouse neural cell primary cultures could be an alternative system for studying Flavivirus infection in the central nervous system.
