Summary: This study found that the use of MAGL inhibitor AM9928 can effectively block the adhesion and migration of TNBC cells, reduce the secretion of pro-inflammatory factors by cells, and inhibit the activation of HBMECs and cytokine secretion caused by TNBC. Animal experiments showed that AM9928 can inhibit the growth of mammary tumors in the mammary fat pads and reduce the permeability changes of the blood brain barrier, thus reducing the metastasis of TNBC in the brain. These findings suggest that MAGL inhibitors have the potential to be a novel strategy for TNBC treatment.

Summary: Breast cancer stem cells (BCSCs) are a small population of tumor cells with high metastatic potential. The molecule biglycan (BGN) has been identified as a potential molecular target in BCSCs that regulates their aggressive characteristics. Removing BGN reduces BCSC proportions and tumorsphere formation, as well as decreases BCSC-associated migration, invasion, and metabolism. These findings suggest that targeting BGN could be an important therapeutic strategy for treating metastatic breast cancer.

Summary: The study reveals that GBP5 is overexpressed in TNBC tissues, correlates with cellular migration ability, and serves as a poor prognostic marker. Co-expression analysis suggests that upregulation of GBP5 may be associated with the activation of interferon gamma and NF-kappa B signaling cascades, and is significantly positively correlated with PD-L1 expression in TNBC tissues.

Summary: The NF-kappa B signaling pathway is overactivated in tumor cells, releasing inflammatory factors and promoting tumor immunosuppression and metastasis. In breast cancer cells, tetrahydrocurcumin (THC) induces apoptosis, inhibits proliferation and migration via the CYP1A1/NF-kappa B signaling pathway, and shows significant tumor growth inhibition and anti-pulmonary metastasis effects in a mouse model.

Summary: ROC1 is up-regulated in BCa tissues and cell lines, with high levels correlated with higher tumor grade, metastasis, and poor prognosis. It promotes BCa invasion and migration by activating NF-κB signaling through enhancing p-IκBα ubiquitination, leading to p65 nuclear translocation and transcription of metastasis-related genes.

Summary: In this study, mutant RANKL was used as a vaccine for immunotherapy of bone metastatic breast cancer, and it was found that anti-RANKL antibodies can inhibit intraosseous tumor growth, reduce osteolytic lesions, and improve overall survival and skeletal metastasis in tumor-bearing mice. The induction of anti-RANKL antibodies by RANKL(M) led to decreased GDF-15 production, deactivating the NF-κB signaling pathway and ultimately inhibiting the metastasis of breast cancer cells to bone.

Summary: The study identified MEST as a key regulator of invasion and metastasis in lung cancer, promoting cellular metastasis by activating the NF-kappa B signaling pathway. MEST interacts with VCP to coordinate the pathway, and high expressions of MEST and VCP are associated with poor survival in lung cancer patients. Targeting the MEST/VCP/I kappa B alpha/NF-kappa B signaling pathway may be a promising strategy for treating lung cancer.

Summary: The role of long non-coding RNAs (lncRNAs) in cancer metastasis, particularly in the context of NF-kappa B signaling and epithelial-mesenchymal transition (EMT), is a hot topic of research. NKILA has emerged as a well-studied lncRNA that regulates NF-kappa B and inhibits EMT-induced metastasis. lncRNAs like FOXD2-AS1, KRT19P3, MALAT1, SNHG15, CRNDE, and AC007271.3 are also implicated in the regulation of NF-kappa B signaling and cancer metastasis.

Summary: The study found that in breast cancer metastasis, OR5B21 showed increased transcript abundance in distant metastatic sites, and its knockdown significantly reduced invasion and migration of cancer cells. OR5B21 expression was associated with epithelial to mesenchymal transition through the STAT3/NF-kappa B/CEBPb signaling axis.

Summary: TRAF6 has emerged as a key regulator of breast cancer (BCa), and the inhibition of TRAF6 shows promise in the treatment of metastatic BCa. However, the low number of studies and scarcity of evidence from animal and human studies may limit the translation of these findings into clinical practice.

Summary: Tumor-derived extracellular vesicles play critical roles in generating a pre-metastatic microenvironment for organotropic metastasis. This study discovers the crucial role of large oncosomes in bone-tropic metastasis of hepatocellular carcinoma by engineering an osteoclastic pre-metastatic niche. The interaction between VAMP-associated protein A on large oncosomes and osteoclasts mediates the formation of the microenvironment and contributes to HCC bone metastasis.
