This study systematically investigates the functionalization of silica gel using thiol-ene click reaction, comparing the effects of MPS and MPTMS chemistry on stationary phase synthesis. The results indicate that MPS has faster reaction kinetics and forms a more homogeneous, thinner monolayer on the silica surface.
The present work systematically investigates a new strategy for the functionalization of silica gel using alkyl silatrane chemistry instead of alkylsilanes for synthesis of chromatographic stationary phases. In this work, silica was chemically modified for further functionalization by a thiol-ene click reaction. Thus, 3-mercaptopropylsilatrane (MPS) was used which is capable to form self-assembled monolayers (SAM) on top of silanol surfaces in a controlled manner as previously shown for silicon wafers. The utility of this chemistry for stationary phase synthesis in liquid chromatography was not evaluated yet. Hence, silica surface modifications using MPS were studied in comparison to established 3-mercaptopropyltrimethoxysilane (MPTMS) chemistry. First, the employed elemental analysis method was validated and it showed excellent intra-day and inter-day precisions (typically less than 5% RSD). It could be shown that the reaction kinetics of MPS was roughly 35-times faster than with MPTMS. After 30 min reaction time with MPS, the thiol content reached 74% of the maximal coverage. Due to controlled chemistry with MPS, which does not lead to oligomeric siloxane network at the silica surface, the ligand coverage was lower. However, multiple silanization cycles with MPS led to a dense surface coverage (around 4 mu mol m(-2)). Si-29 cross polarization/magic angle spinning (CP/MAS) solid-state NMR revealed distinct T-1/T-2/T-3 ratios for MPS and MPTMS materials with up to 80% T-3 (indicative for trifunctional siloxane linkage) for MPS and around 20% T3 for MPTMS. This indicates a more homogeneous, thinner monolayer film of MPS on the silica surface, as compared to an irregular thick oligomeric siloxane network with MPTMS. Bonding of quinine carbamate as chiral selector afforded an efficient chiral stationary phase (CSP) for chromatographic enantiomer separation. Separation factors were comparable to MPTMS-bonded CSP, however, chromatographic efficiency was much better for the MPS-bonded CSP. H/u curves indicated a reduced mass transfer resistance by roughly factor 3 for MPS- compared to MPTMS-bonded CSP. This confirms better chromatographic performance of surfaces with homogeneous monolayer compared to network structures on the silica surface which suffer from poor stationary phase mass transfer. Summary: The chromatographic behavior of mixed-mode and hydrophilic interaction liquid chromatography (HILIC) was assessed in this study for the separation of metabolites from glycolysis and pentose phosphate pathways. Optimization of chromatographic conditions allowed for selective targeted analysis of glycolytic phosphorylated carbohydrate metabolites using liquid chromatography with tandem mass spectrometry (MS/MS). The separation of pentose phosphate isomers was achieved using mixed-mode HILIC/SAX and HILIC BEH Amide columns. HILIC-MS/MS showed challenges in separating hexose monophosphate metabolites, but the BEH Amide column provided the best results for this separation.

Summary: In this work, new polymeric chiral stationary phases were prepared and compared with brush-type chiral stationary phases. The polymeric CSPs showed similar enantioselectivity but lower chromatographic efficiencies compared to the brush phases. Oxidation of the polymeric CSPs reduced retention times and allowed for lower counterion concentrations in liquid chromatography-electrospray ionization-mass spectrometry applications. In addition, cryogenic cooling of the column enabled baseline separation of estradiol.

Profiling of branched chain and straight chain saturated fatty acids by ultra-high performance liquid chromatography-mass spectrometry

Summary: In this study, seven commercially available UHPLC columns were systematically evaluated for their retention and isomer selectivity of branched chain fatty acids (BCFAs). It was found that IG-U column showed the best isomer selectivity for short and medium-chain BCFA isomers, while RP C18 column had good selectivity for long-chain BCFA isomers. Furthermore, an untargeted isomer-selective BCFA profiling method was developed using UHPLC-ESI-MS/MS without pre-column derivatization.

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles
