Summary: We evaluated the effects of a dimethyl sulfoxide (Me2SO)-free cryopreservation solution on human adipose-derived mesenchymal stromal cells (hADSCs). The results showed that the addition of 3% trehalose and 5% dextran to lactated Ringer's solution (LR) improved cell viability after thawing. Additionally, the optimal concentration of propylene glycol (PG) to maintain cell characteristics was found to be 2.5% to 5%.

Summary: The study found that SVF cells can enhance bone healing, with cryopreserved cells showing almost equal potential as fresh cells. SVF cells can be used to improve nonunion bone fracture healing as an alternative to other mesenchymal stem cells, and their effect can be maintained under cryopreservation.

Summary: The study demonstrated that a high percentage of serum (80%) is necessary for optimal cryopreservation with 10% DMSO. Thawed cells at passages from P2 to P4, even after seven years, could still be considered for therapeutic application and in vitro study as they retain stem potential post-cryopreservation.

Summary: Adipose tissue senescence is linked to obesity and aging-related diseases, and adipose-derived stem cells are used in regenerative medicine. Cryopreservation allows for greater flexibility in tissue use. Trehalose, a sugar synthesized by lower organisms, has been tested as a cryoprotective agent, showing comparable effectiveness to DMSO when introduced into cells using appropriate methods.

Summary: Autophagy is a cellular process that maintains cell homeostasis by activating a specific pathway and providing necessary factors in stressful and physiological situations. In a 3D hydrogel, adipose-derived mesenchymal stromal cells (ASCs) showed increased expression of autophagic markers and features. Autophagy plays a crucial role in ASCs chondrogenic differentiation by providing energy and necessary factors.

Summary: Adipose tissue relies on the regulation of multipotent mesenchymal stromal cells (MSCs), which serve as both precursors for new adipocytes and key paracrine regulators of adipose tissue homeostasis. The activity of MSCs is tightly controlled, regulating aspects such as proliferation, differentiation, and signal molecule production through modulation of hormonal sensitivity.

Summary: In this study, a DMSO analogue called l-methionine sulfoxide (Met(O)-OH) was investigated as a cryoprotectant for cell cryopreservation. The results showed that Met(O)-OH could stably bind the surface of ice crystals and form stable hydrogen bonds with ice. Additionally, Met(O)-OH significantly reduced cell damage caused by osmotic shock and had high post-thaw survival efficiency for L929 cells.

Summary: This review highlights the potential of mesenchymal stromal cells (MSCs) in treating osteoarthritis (OA), focusing on their chondrogenic and immunomodulatory potential. The review provides an overview of the effects of MSCs in preclinical and clinical studies of OA treatment, comparing different MSC sources. Furthermore, the review summarizes the current evidence for the assessment of cartilage in OA using magnetic resonance imaging (MRI) and second-look arthroscopy after MSC therapy.

Summary: Adipose-derived mesenchymal stromal cells (ASCs) hold great potential for cellular therapies and can differentiate into endothelial cells (ECs) and other angiogenic cell types. Nanofibrillated cellulose (NFC) hydrogel is a suitable matrix for 3D cell culture of ASCs and can stimulate angiogenesis. This study recommends the use of NFC hydrogel as an animal-free biomaterial scaffold for therapeutic angiogenesis and other cell therapy purposes.

Summary: This study investigates the use of adipose-derived mesenchymal stromal/stem cells (AD-MSCs) in autologous fat transfer (AFT) for tissue reconstruction. The results show that AD-MSCs can improve graft persistence and reduce tissue resorption after AFT. The implanted AD-MSCs can persist and differentiate into mature adipocytes, leading to better outcomes in terms of graft weight and reduced cyst formation. This suggests a long-term protective and anti-inflammatory/proangiogenic role of AD-MSCs in autologous AFT tissue resorption.

Summary: MSCs from liver and adipose tissue showed similar proliferation and migration properties, but had differences in immunomodulatory and pro-angiogenic abilities. Selecting cell-based therapy according to the specific characteristics of different cell sources may be important for treating certain diseases.

Summary: In this study, we primed human MSC derived from adipose tissues using toll-like receptor agonists and investigated their interactions with prostate cancer cells. The results showed that primed MSC significantly accelerated tumor growth and induced changes in the tumor microenvironment, including altered modulation of angiogenesis and immune response processes.

Summary: Cell-based therapies using PDLSC may be a promising alternative to MSC(M) and MSC(AT) for periodontal regeneration, as PDLSC showed enhanced proliferative and osteogenic potential. PDLSC expressed osteogenic markers and had higher calcium content and upregulated osteogenic/periodontal genes compared to MSC(M) and MSC(AT). CD146 was specifically expressed by PDLSC, which also had a higher proliferative capacity. However, alkaline phosphatase activity did not increase in PDLSC.

Summary: This study developed an efficient method for liquid storage of in vitro-derived porcine blastocysts at ambient temperature for 24 hr. The results showed that storing the blastocysts in a Cell-S solution at 25 degrees Celsius maintained their survival and development, and supplementation with curcumin improved their development rate.

Summary: This study successfully created and characterized Gal KO adult pig bone marrow MSCs, showing similar characteristics and gene upregulation to WT MSCs. While the colony-forming efficiency of Gal KO apBM-MSCs was slightly lower than that of WT apBM-MSCs, there were no significant differences in differentiation and gene expression between the two. This suggests that Gal KO apBM-MSCs may be a safer and more effective therapeutic option.

Summary: This study aimed to develop a simple method for short-term preservation of in vitro-produced porcine zygotes at 25°C for up to 2 days. The results showed that the Cell-W solution was the most efficient for 24-hour storage of porcine zygotes at 25°C, with no apparent effects on blastocyst quality. However, as storage duration increased, blastocyst formation rates significantly decreased.
