Potential therapeutic targets of Klebsiella pneumoniae: a multi-omics review perspective

The prolonged use of antibiotics has led to the development of multidrug resistance in many organisms, causing a global health crisis. Klebsiella pneumoniae is a causative organism for various infections and recently, both immunocompromised and healthy individuals are equally susceptible to K. pneumoniae infections due to the emergence of hypervirulent strains. This review discusses the pathophysiology of K. pneumoniae, including its virulence determinants, and explores potential drug targets for the discovery of novel drugs and effective treatment regimens.
The multidrug resistance developed in many organisms due to the prolonged use of antibiotics has been an increasing global health crisis. Klebsiella pneumoniae is a causal organism for various infections, including respiratory, urinary tract and biliary diseases. Initially, immunocompromised individuals are primarily affected by K. pneumoniae. Due to the emergence of hypervirulent strains recently, both healthy and immunocompetent individuals are equally susceptible to K. pneumoniae infections. The infections caused by multidrug-resistant and hypervirulent K. pneumoniae strains are complicated to treat, illustrating an urgent need to develop novel and more practical approaches to combat the pathogen. We focused on the previously performed high-throughput analyses by other groups to discover several novel enzymes that may be considered attractive drug targets of K. pneumoniae. These targets qualify most of the selection criteria for drug targeting, including an absence of its homolog's gene in the host. The capsule, lipopolysaccharide, fimbriae, siderophores and essential virulence factors facilitate the pathogen entry, infection and survival inside the host. This review discusses K. pneumoniae pathophysiology, including its virulence determinants and further the potential drug targets that might facilitate the discovery of novel drugs and effective treatment regimens shortly.

Ejection Fraction estimation using deep semantic segmentation neural network

Summary: This paper proposes an automated Ejection Fraction estimation system from 2D echocardiography images using deep semantic segmentation neural networks. Two parallel pipelines of deep semantic segmentation neural network models have been proposed for efficient left ventricle segmentation, and three different neural networks, UNet, ResUNet, and Deep ResUNet, have been implemented. The most accurate model achieved high Dice scores for left ventricle segmentation in both systolic and diastolic states. The proposed system can remove the eyeball estimation practice and reduce inter-observer variability.

Summary: Spleen tyrosine kinase (SYK) is a non-receptor tyrosine kinase that plays a crucial role in signal transduction in different cell types. In this study, two natural compounds, Isolysergic acid and Michelanugine, were identified as potential SYK inhibitors with strong affinity and specificity for the SYK binding pocket. These compounds have the potential to be developed into promising SYK inhibitors for the treatment of autoimmune disorders, cancer, and inflammatory diseases.

Counteraction of the cetyltrimethylammonium bromide-induced protein aggregation by heparin: Potential impact on protein aggregation and neurodegenerative diseases using biophysical approaches

Summary: This study investigates the effect of heparin on CTAB-induced aggregation of bovine serum albumin (BSA) using biophysical techniques. The results show that heparin completely inhibits CTAB-induced protein aggregation and reduces the apparent rate constant. CTAB acts as a protein-aggregating agent, but heparin prevents the perturbation of secondary and tertiary structure induced by CTAB and the formation of amyloid-like structures. ITC measurements reveal that CTAB and heparin form a complex that does not bind with protein. The CTAB-heparin complex may play a role in inhibiting protein aggregation and could serve as a basis for developing therapeutic strategies for various neurodegenerative complications.

Summary: Protein kinases (PKs) are extensively studied drug targets, particularly in the field of oncology and neurodegeneration. Tau protein hyperphosphorylation, a post-translational modification observed in Alzheimer's disease (AD), highlights the involvement of PKs in neurodegeneration. The efficacy of protein kinase inhibitors (PKIs) and protein phosphatase 2 A (PP2A) activators in AD has been investigated in preclinical and clinical studies, but the outcomes have been variable. This review examines the results of these studies and evaluates the potential of targeting tau hyperphosphorylation as a disease modifying therapy.

Summary: In this study, ZnFe2O4 nanocomposites were successfully synthesized and used for dye removal from aqueous solutions. The results showed that pH, adsorbent dosage, contact time, and initial concentration of dyes had an effect on the adsorption performance. The nanocomposites were found to be reusable to some extent.

Integrated spectroscopic and MD simulation approach to decipher the effect of pH on the structure function of Staphylococcus aureus thymidine kinase

Summary: Staphylococcus aureus is a significant human pathogen that is becoming increasingly resistant to antibiotics. This study focuses on thymidine kinase (TK), an enzyme essential for DNA synthesis and repair, and investigates how different pH levels affect its structure and function. The findings indicate that pH variation disrupts the secondary and tertiary structures of TK, leading to aggregate formation at pH 5.0. The enzyme displays maximum activity within the physiological pH range, suggesting a connection between structural changes and enzymatic activity.
