Summary: This study aims to investigate the feasibility of awake surgery in glioblastoma patients with severe aphasia using a patient-tailored approach. Despite preoperative severe aphasia, functional boundaries for critical language areas could be identified in each patient, and postoperatively, all patients had stable or improved language outcome.

Summary: Preoperative clinical MRI protocols for gliomas still rely on conventional structural MRI, which does not provide information on tumor genotype and has limitations in the delineation of diffuse gliomas. The GliMR COST action aims to raise awareness about advanced MRI techniques in gliomas and their potential clinical translation. This review summarizes current methods, limitations, and applications of advanced MRI for the preoperative assessment of glioma, and evaluates the level of clinical validation of different techniques.

Summary: Antibody-drug conjugates (ADCs) have shown efficacy in various cancers, but their activity in the central nervous system (CNS) may be limited by the blood-brain barrier. This review summarizes the available clinical data on ADCs against primary and secondary brain tumors and ongoing trials. It also discusses the physical, biological, and molecular determinants of ADCs' CNS activity and potential strategies to improve drug delivery to brain tumors.

Summary: This study aimed to evaluate the effectiveness of levetiracetam combined with valproic acid and clobazam compared with other triple therapy combinations in patients with glioma. The results showed that there were no significant differences in treatment failure rate, uncontrolled seizure rate, adverse effects rate, and recurrent seizure rate between levetiracetam combined with valproic acid and clobazam and other triple therapy combinations.

Summary: This study found that simultaneous inactivation of Nbs1 and p53 in mice leads to the development of high-grade gliomas with radiation-induced glioma features. This animal model could be useful for preclinical studies to improve the prognosis of these deadly tumours, and it highlights the singularity of NBS1 among the other DNA damage response proteins in the etiology of brain tumors.

The study found that MGMT promoter methylation and RTK1 classifier assignment were prognostic factors, while NF1 mutation may predict response to bevacizumab treatment.

The molecular diagnostic yield of frame-based stereotactic biopsies in the age of precision neuro-oncology: a cross-sectional study

Summary: This study investigated the success rate of diagnosis after frame-based stereotactic biopsies of intracranial lesions. The results showed that a conclusive diagnosis was possible in 94.5% of cases, diagnosis with missing molecular genetics data was established in 2.0% of cases, and an inconclusive diagnosis was met in 3.5% of cases. The study concluded that although frame-based stereotactic biopsies provide limited tissue samples, they have high diagnostic yields in histopathology and molecular genetics.

Summary: The study aimed to determine whether simultaneous inactivation of Nbs1 and p53 in neural progenitors triggers brain tumorigenesis, and if so, in which category this tumor can be classified. The results showed that Nbs1/P53-deficient mice develop high-grade gliomas occurring in the olfactory bulbs and in the cortex along the rostral migratory stream. Molecular analysis revealed striking similarities between these tumors and pediatric human high-grade gliomas, as well as radiation-induced gliomas.
