Proof-of-Concept Study of Multifunctional Hybrid Nanoparticle System Combined with NIR Laser Irradiation for the Treatment of Melanoma

The global impact of cancer emphasizes the importance of developing innovative, effective and minimally invasive therapies. In the context of superficial cancers, the development of a multifunctional nanoparticle-based system and its in vitro and in vivo safety and efficacy characterization are, herein, proposed as a proof-of-concept. This multifunctional system consists of gold nanoparticles coated with hyaluronic and oleic acids, and functionalized with epidermal growth factor for greater specificity towards cutaneous melanoma cells. This nanoparticle system is activated by a near-infrared laser. The characterization of this nanoparticle system included several phases, with in vitro assays being firstly performed to assess the safety of gold nanoparticles without laser irradiation. Then, hairless immunocompromised mice were selected for a xenograft model upon inoculation of A375 human melanoma cells. Treatment with near-infrared laser irradiation for five minutes combined with in situ administration of the nanoparticles showed a tumor volume reduction of approximately 80% and, in some cases, led to the formation of several necrotic foci, observed histologically. No significant skin erythema at the irradiation zone was verified, nor other harmful effects on the excised organs. In conclusion, these assays suggest that this system is safe and shows promising results for the treatment of superficial melanoma.

Prediction of transition to psychosis from an at-risk mental state using structural neuroimaging, genetic, and environmental data

Summary: This study aimed to predict the transition to psychosis from an At Risk Mental State (ARMS) by using a combination of machine learning, structural magnetic resonance imaging (sMRI), genome-wide genotypes, and environmental risk factors as predictors. The results suggested that none of the modalities alone, namely neuroimaging, genetic data, or environmental data, could predict psychosis from an ARMS statistically better than chance, indicating the need to re-evaluate the value of sMRI data and genome-wide genotypes in predicting psychosis risk.

A Universal Pharmacological-Based List of Drugs with Anticholinergic Activity

Summary: This study aimed to provide a comprehensive list of drugs with documented affinity for muscarinic receptors, in order to address the gaps in anticholinergic burden tools. A literature review and database search were conducted to identify these tools and evaluate the drugs included in them. A universal list of 133 drugs with anticholinergic activity, categorized by their affinity for different subtypes of muscarinic receptors and their ability to cross the blood-brain barrier, was developed. The proportion of drugs with confirmed antagonism varied among the tools, highlighting the need for further validation in different clinical settings.

Summary: Hepatocellular carcinoma (HCC) is a concerning and increasingly prevalent tumor. Traditional pharmacological approaches have limitations and side effects, necessitating the development of new molecules. This study shows that LPAR6 plays a role in CDAA diet-induced HCC in mice, and XAA can counteract diet-induced effects on liver health. These findings are supported by experiments in HCC cells with altered LPAR6 function.

Edible Seaweeds Extracts: Characterization and Functional Properties for Health Conditions

Summary: In this study, extracts of the edible seaweeds Arame, Nori, and Fucus were compared to explore their health properties. The results showed that the fractionation of the extracts led to a reduction in bioactivity, indicating a loss of synergistic effects. Arame extract exhibited the highest antioxidant activity, while Nori showed the highest potential for acetylcholinesterase inhibition. These findings suggest a promising potential for the use of these seaweed extracts, especially Arame and Nori, in improving health and managing diseases associated with oxidative stress and neurodegeneration.

Structural Optimization of Antimycobacterial Azaaurones Towards Improved Solubility and Metabolic Stability

Summary: This study successfully improved the metabolic stability and solubility of N-acetyl azaaurones by replacing the N-acetyl moiety with a N-carbamoyl group. The new N-carbamoyl azaaurones exhibited potent antimycobacterial activity against drug-susceptible and drug-resistant M. tuberculosis isolates, with improved microsomal stability and kinetic solubility. These findings highlight the potential of azaaurones as antimycobacterial agents.

Summary: This study evaluated the role of hsa-miRNA-21-5p in the development of hepatocellular carcinoma (HCC) in non-alcoholic fatty liver disease (NAFLD) patients. The expression of hsa-miRNA-21-5p was found to increase with NAFLD severity and was associated with hepatic lipotoxicity. Inhibition of hsa-miRNA-21-5p was shown to prevent the progression of NAFLD to NASH and HCC.

Make It Less Complex: Autoencoder for Speckle Noise Removal-Application to Breast and Lung Ultrasound

Summary: This study proposed a simplified Convolutional Neural Network Autoencoder (CNN-AE) to remove noise from breast and lung ultrasound images. The results showed that the model outperformed traditional filters in denoising. Furthermore, in the classification of malignant breast lesions, the use of the denoised images by the model reduced misclassifications. Therefore, this research is of great importance for practical clinical applications.
