Summary: This study suggests that NOTCH mutations (NOTCHmut) may serve as an indicator for poor prognosis of acute myeloid leukemia (AML). NOTCHmut is associated with low platelet count and coexistence with other mutations. The presence of NOTCHmut is correlated with worse overall survival and relapse-free survival, especially within the intermediate-risk group according to the European Leukemia Net 2017 classification.

Summary: The study confirms the extremely poor outcome of multiple myeloma patients with a double hit situation of 17p deletion and TP53 mutations, as well as the poor outcome associated with 17p deletion alone. These findings highlight the importance of these genetic features as prognostic indicators for poor outcome in multiple myeloma patients.

The effect of this treatment also varies among patients with different MGMT methylation, IDH1 mutation, and Ki67 expression levels.

Summary: Dynamic amino acid positron emission tomography (PET) is essential in neuro-oncology, particularly for predicting isocitrate dehydrogenase (IDH) mutations in newly diagnosed gliomas. This study compared different models for predicting IDH mutations using F-18-FDOPA PET images of gliomas, and found that a semiquantitative (SQ) model had comparable performance to more complex models, supporting its routine clinical application.

Summary: The study evaluated the value of FET-PET parameters in diagnosing pseudoprogression related to lomustine-temozolomide chemoradiation, finding that TBRmean had the highest accuracy in identifying pseudoprogression. Furthermore, integrating relative changes of TBRmean improved accuracy, and combining static and dynamic parameters increased specificity to 100%.

Summary: Clinical and staging chest CT characteristics play an important role in predicting brain metastasis in patients with newly diagnosed NSCLC, with distinct predictors identified for resectable and unresectable stages. Factors such as age, sex, extrathoracic metastasis, lung cancer subtypes, and EGFR mutations were associated with brain metastasis in these patients.

Summary: The SOLO1 study demonstrated that maintenance therapy with olaparib can effectively prolong progression-free survival in Chinese patients with newly diagnosed advanced ovarian cancer, who have a BRCA mutation and are in complete or partial response after platinum-based chemotherapy. Moreover, the treatment showed good safety profile, making it an effective option for patients in this population.

Summary: In this study, FLT3 (ITD-TKD) mutations were investigated in 100 newly adult Syrian patients with AML-Normal karyotype (NK). The prevalence of FLT3-ITD mutation was found to be 24%, with various duplication and insertion mutations. Significant differences were observed in lactate dehydrogenase level, overall survival, relapse, and event free survival between patients with and without FLT3-ITD mutation.

Summary: In the absence of head-to-head trials, a population-adjusted indirect treatment comparison was conducted to evaluate the efficacy and safety of maintenance olaparib and bevacizumab in patients with advanced ovarian cancer and a BRCA mutation. Findings suggest that adding bevacizumab to olaparib could improve progression-free survival, and maintenance olaparib alone or in combination with bevacizumab is a recommended treatment option for newly diagnosed patients with advanced ovarian cancer and a BRCA mutation.

Summary: This study explored health workforce responses to COVID-19 through analyzing job advertisements, finding that nurses were the most advertised position in each country and frontline workers were in higher demand compared to other roles. Job advertisements served as a valuable source of information on how workforces adapt to a pandemic, emphasizing the crucial role of frontline workers, especially nurses, in the initial phases of response.

Summary: Weight loss in people with type 2 diabetes can lead to improved glycemic control, quality of life, and comorbidities. Achieving diabetes remission can reduce the risk of complications and lessen the burden on healthcare systems. However, maintaining weight loss in the long term is challenging. Strict glycemic control has its benefits, but its impact on cardiovascular and all-cause mortality is still uncertain. New antidiabetic agents like GLP-1 receptor agonists and SGLT2 inhibitors offer additional benefits of weight loss and reduced cardiorenal risk. The debate on prioritizing weight loss or glycemic control in recent type 2 diabetes patients is discussed in this article, suggesting a combined strategy with early and aggressive use of GLP-1 receptor agonists and SGLT2 inhibitors for maximum benefits and decreased complication risk. A personalized approach considering specific patient characteristics can improve diabetes care.

Summary: Evidence from the BRIDGES study suggests that germline protein truncating variants (PTVs) in FANCM are associated with increased risk of ER-negative and triple-negative breast cancer (TNBC), particularly for those with a family history. This study further investigates the association between FANCM missense variants (MVs) and breast cancer risk using the BRIDGES study, analyzing a total of 689 MVs. The results indicate that FANCM MVs may be low/moderate risk factors for ER-negative and TNBC subtypes of breast cancer.

Summary: With the legalization of social egg freezing in Singapore starting from 2023, prospective patients are required to undergo mandatory pre-procedure counseling in order to make an informed choice. However, there are currently no clear guidelines on what this counseling should entail, potentially leading patients to have unrealistic expectations about the procedure. To address this, a systematic protocol and methodology tailored to Singapore's sociocultural context has been developed to provide comprehensive counseling and ensure that women considering elective egg freezing are well-informed about success rates and potential outcomes.

Summary: To aid in the accurate removal of cancer cells during breast-conserving surgery, researchers have developed a handheld optical coherence tomography (OCT) probe and attenuation imaging method, which allows real-time imaging of the surgical cavity. Through experimental validation, this method has been shown to provide accurate and repeatable imaging, and offers an additional tissue contrast mechanism to assist in the identification and removal of cancer cells during breast-conserving surgery.

Summary: Optical palpation is a new emerging technique that uses an optical imaging system to generate images of mechanical contrast in tissue. A camera-based optical palpation system, called stereoscopic optical palpation (SOP), is proposed in this paper to directly map the strain of a compliant layer placed on top of the tissue. With SOP, a strain sensitivity of 4.2 m is achieved, which is a 24-fold improvement over previous camera-based optical palpation techniques. SOP has the potential for rapid and reliable imaging, and could be used for tumor margin assessment in breast-conserving surgery.
