Highlighting the gaps in hazard and risk assessment of unregulated Endocrine Active Substances in surface waters: retinoids as a European case study

The current regulatory hazard and risk assessment of endocrine-active substances are limited and do not fully cover the complexity of the endocrine system. The importance of the retinoid signaling pathway in environmental risks and the need for further research to understand its impacts are highlighted. Focusing on the retinoid signaling pathway can improve the scope and relevance of risk assessment for endocrine disruptors.
Regulatory hazard and risk assessment of endocrine-active substances currently specifies four modes of action: interference with sex hormone (oestrogen, androgen) pathways, steroidogenesis, and thyroid hormone signalling. This does not encompass the full complexity of the endocrine system and its extended interfaces with environmental pollutants that can potentially disrupt the carefully maintained balance. Here we take the retinoid signalling pathway as a European case study for both, under- and unregulated endocrine pathways and outline the different levels of interference, discuss their adversity, and indicate crosstalk to other signalling pathways. Retinoid compounds already exist in drinking water sources, occur naturally in cyanobacterial blooms and/or enter surface waters via wastewater discharge, where they pose a potential hazard to the environment and human health - a situation that can be expected to worsen due to water shortages induced by climate-change and population growth. We briefly review relevant aspects of current endocrine disruptor (ED) testing for regulatory purposes and then expand upon the needs for inclusion of disruption of retinoid signalling in (ED) regulatory safety assessment contributing to adverse health outcomes that include cognitive function and neurological disease. An overview of developmental effects of retinoid signalling disruption across species highlights critical processes and potential crosstalk with other signalling pathways. A focused weight of evidence-based evaluation of the biologically plausible associations between neurological disorders and altered retinoid signalling highlights the evidence gaps. We show that monitoring only a limited number of anthropogenic priority chemicals in water is insufficient to address the environmental risks of retinoid signalling disruption. To comprehensively assess impacts on the endpoints, processes, and pathways of the endocrine system that are most vulnerable to chemical interference we need further investigation of the true mixture composition in environmental matrices. On a weight of evidence-basis this information can then be integrated into a reliable, inclusive, quantitative approach that ultimately accommodates all the critical pathways. By focusing on the retinoid signalling pathway, we intend to improve the scope and relevance of an integrated approach for the risk assessment of endocrine disruptors.

Summary: A hepatitis E virus (HEV) genotype 3-specific monoclonal antibody (mAb) was successfully generated by immunising BALB/c mice with a carboxy-terminal part of its capsid protein expressed in Escherichia coli. The mAb, named G117-AA4, showed strong reactivity with homologous and heterologous HEV-3 capsid proteins, as well as with capsid protein derivatives from other genotypes and related viruses. This novel mAb may prove valuable for further studies on HEV infections and diagnostic purposes.

Summary: This study introduced and validated an androgen receptor (AR) transactivation assay for the detection of AR agonists and antagonists using a stably transfected human prostate cancer cell line. Integration of Epigenetic Mechanisms into Non-Genotoxic Carcinogenicity Hazard Assessment: Focus on DNA Methylation and Histone Modifications

Summary: Epigenetics plays a crucial role in cellular functions and differentiation through mechanisms involving histone and DNA modifications as well as non-coding RNAs. Both epigenetic anomalies and DNA mutations contribute to cellular dysfunctions, including carcinogenesis, in cancers regardless of the initial carcinogenic events' nature. As part of non-genotoxic mechanisms of action, epigenetic assays integrated into testing frameworks can aid in identifying chemical carcinogens and improving public health protection.

Mechanistic Interrogation of Cell Transformation In Vitro: The Transformics Assay as an Exemplar of Oncotransformation

Summary: In this study, the Transformics Assay was used to test the lowest transforming concentration of the carcinogenic chemical B(a)P in order to identify molecular signatures and mechanisms of chemical-induced transformation. The results showed that the activation of AhR led to a cascade of cellular events and modifications in the microenvironment, with the immune and inflammatory responses playing a major role in cell response. Pathways and processes related to cell cycle regulation, cytoskeletal adhesion and remodeling, cell differentiation and transformation were also observed.

Summary: New Approach Methodologies (NAMs) include in vitro, in silico, or chemistry-based methods that can provide information for chemical safety assessment. The European Union's current chemical legislation has limitations on the widespread use of NAMs, although some are considered suitable for regulatory purposes. A workshop identified key points to increase the use and regulatory acceptance of NAMs, such as changes in regulatory frameworks, education, training, stakeholder engagement, and addressing gaps in the scientific basis of NAMs.

Target Mechanisms of the Cyanotoxin Cylindrospermopsin in Immortalized Human Airway Epithelial Cells

Summary: Cylindrospermopsin (CYN) is a cyanobacterial toxin that has delayed effects on animals and humans. This study investigated the effects of CYN on protein expression in human bronchial epithelial cells using a proteomic approach. The results showed that CYN affected the expression levels of proteins involved in various cellular functions, including protein stability regulation, cell adhesion and integration, cell proliferation, cell cycle regulation, and cytokinesis. In addition, CYN reduced cell adhesion, attenuated cell proliferation, delayed mitosis completion, and induced genomic instability.

The Cell Transformation Assay: A Historical Assessment of Current Knowledge of Applications in an Integrated Approach to Testing and Assessment for Non-Genotoxic Carcinogens

Summary: This article describes the history of the development of cell transformation assays (CTAs), providing an overview from their origin to the new transcriptomic-based CTAs. The application of this knowledge is used to address how different types of CTAs can be included in the integrated approach to testing and assessment (IATA) for non-genotoxic carcinogens. The article also discusses how different CTA models fit into the preceding steps of the IATA, and maps complementary key biomarkers to represent the complexity of (non-genotoxic) carcinogenesis process.
