Summary: This study explored the pharmacological actions and mechanisms of Raddeanin A (RA) in non-small-cell lung cancer (NSCLC) using network pharmacology. The potential targets of RA for NSCLC therapy, such as SRC, MAPK1, and STAT3, were identified. Enrichment analyses indicated the involvement of these targets in regulating cell death, MAPK cascade, Ras signaling pathway, and PI3K/AKT signaling pathway. Experimental results showed that RA effectively inhibited proliferation, induced apoptosis, and induced autophagy in A549 lung cancer cells and downregulated the activity of the PI3K/AKT/mTOR pathway. Overall, this study revealed the antitumor effects and underlying mechanisms of RA on apoptosis and autophagy in A549 cells, suggesting its potential as an effective antineoplastic agent.

Summary: Bavachinin (BVC), a natural small molecule derived from the Chinese herb Fructus Psoraleae, displays multiple pharmacological effects and may serve as a potential drug candidate for the treatment of rheumatoid arthritis (RA). This study identified BVC targets and RA-related targets, and found that BVC primarily acts through the PPARG/PI3K/AKT signaling pathway to inhibit cell proliferation, migration, and inflammatory cytokine production, while promoting cell apoptosis. In vivo experiments showed that BVC alleviated joint injury and inflammation in CIA mice. These findings provide a theoretical foundation for the development of RA therapies.

Summary: In this study, the effective components and underlying mechanisms of Quince (Cydonia oblonga Mill, COM) extract against atherosclerosis were identified. Using UHPLC-Q-TOF-MS/MS, 14 serum components of COM extract were identified. Network pharmacology analysis predicted 573 targets, including 224 atherosclerosis-specific targets. The key targets included GSK3 beta, ESR1, EGFR, and HSP90AA1, and the key signaling pathway was the PI3K-Akt signaling pathway. COM extract exhibited hypolipidemic, anti-oxidative, anti-inflammatory, anti-thrombotic, and vascular endothelium protective effects in a rat model of atherosclerosis. These effects may be related to the inhibition of the EGFR/PI3K/AKT/GSK-3 beta signaling pathway.

Summary: This study revealed the antimetastatic effect of SAG on melanoma and identified the underlying molecular mechanisms through the inhibition of the PI3K-AKT signaling pathway. Results showed that SAG inhibited the proliferation, migration, and invasion of melanoma cells and effectively suppressed tumor growth in a xenograft model. These findings offer a theoretical foundation for the clinical application of SAG in antitumor therapy.

Summary: This study found that hydroxychloroquine (HCQ) can alleviate inflammation and cell apoptosis caused by renal fibrosis by inhibiting the activation of the PI3K/AKT and NF-kB pathways, improving the epithelial-mesenchymal transition (EMT) and extracellular matrix (ECM) deposition in renal tubular epithelial cells. This provides theoretical evidence for the treatment of renal fibrosis with HCQ.

Summary: This study identifies eriodictyol as the most effective anti-gastric cancer flavonoid from Polygoni orientalis Fructus (POF) and investigates its potential targets in gastric cancer. Eriodictyol exhibits inhibitory effects on gastric cancer cell viability, suppresses colony formation, induces cell apoptosis, and inhibits tumor growth in vivo. Network pharmacology analysis suggests that the PI3K/AKT signaling pathway may be a potential target for eriodictyol in gastric cancer.

Summary: This study investigated the effect and mechanism of Zhuanggu Zhitong Capsule (ZGZT) in osteosarcopenia (OS) rats. Results showed that ZGZT could effectively improve the phenotypes of OS by up-regulating the PI3K/Akt/Bcl2 signaling pathway. This suggests that ZGZT may be a new treatment option for OS prevention and treatment.
