Summary: Osteosarcoma is a disease in humans characterized by TP53 mutations. In mice, loss of p53 triggers OS development, and osteoprogenitor-specific p53-deleted mice are widely used to study the process of osteosarcomagenesis. However, the molecular mechanisms underlying the initiation or progression of OS following or parallel to p53 inactivation remain largely unknown. This study reveals a novel tumor suppressive mechanism mediated by C/ebp alpha in p53-deficient osteosarcomagenesis, highlighting the importance of the Runx-Myc oncogenic axis as a therapeutic target for OS.

Summary: This study found that stroma-specific loss of p53 enhances tumorigenesis driven by the oncogene Kras(G12D) in mammary glands. The loss of p53 in the stroma promotes both epithelial and fibroblast proliferation, increases DNA damage and/or replication stress in tumor cells, and decreases apoptosis. Furthermore, it affects the tumor immune landscape and has an impact on patient survival.

Summary: This study investigated the active effects of Radix Bupleuri-Radix Paeoniae Alba (BP) on hepatocellular carcinoma (HCC) and found that the major active ingredients in BP were quercetin, kaempferol, isorhamnetin, stigmasterol, and beta-sitosterol. Molecular docking analysis confirmed the binding activity of these active ingredients with PTEN. In an HCC mouse model, BP exhibited anti-tumor effects and affected the immune microenvironment by increasing CD8+ and IFN-gamma+/CD4+ T cell levels and decreasing PD-1+/CD8+ T cell and Treg cell levels. BP also up-regulated IL-6, IFN-gamma, and TNF-alpha levels while down-regulating IL-10 levels in HCC mice. The effects of BP were dependent on PTEN activation.

The findings show that SlHY5 promotes chlorophyll accumulation in fruits, with a stronger effect on the illuminated side. Transcriptome and metabolome analyses also demonstrate that SlHY5 is involved in the regulation of phenylpropanoids and steroidal glycoalkaloids. These results highlight the importance of manipulating SlHY5 for improving seedling fitness and fruit quality in tomato.

This developmental pathway is conserved in humans, where AKT-dependent splenic IgD+CD27+ B cells are proposed as the equivalent of MZ B cells.

Summary: This study reveals the impact of differential expression of pyruvate kinase isoforms on cancer initiation and progression. The M2 splice isoform of pyruvate kinase (PKM2) supports cancer cell proliferation and survival in prostate cancer, while the M1 isoform (PKM1) has a suppressive effect on tumor development. Alterations in nucleotide levels are observed in tumors with high PKM1 expression, leading to DNA replication stress and senescence which inhibits tumor progression. A small molecule pyruvate kinase activator that mimics the high activity PKM1-like state can suppress the progression of established prostate tumors.

Summary: QKI is found to be reduced in human cervical cancer, and its overexpression inhibits cell proliferation and promotes apoptosis. Through RNA-seq and qRT-PCR, hundreds of QKI-regulated alternative splicing events and differentially expressed genes were identified, closely related to cancer, apoptosis, and transcriptional regulation functions. In conclusion, QKI may impact the occurrence and development of cervical cancer by regulating gene expression through alternative splicing.
