Noncoding RNA transcripts, as important regulators in the tumor microenvironment, play crucial roles in modulating macrophage polarization and influencing tumor progression and clinical outcomes. More research is needed to further understand the mechanisms underlying these processes and the potential therapeutic implications.
Noncoding RNA (ncRNA) transcripts that did not code proteins but regulate their functions were extensively studied for the last two decades and the plethora of discoveries have instigated scientists to investigate their dynamic roles in several diseases especially in cancer. However, there is much more to learn about the role of ncRNAs as drivers of malignant cell evolution in relation to macrophage polarization in the tumor microenvironment. At the initial stage of tumor development, macrophages have an important role in directing Go/No-go decisions to the promotion of tumor growth, immunosuppression, and angiogenesis. Tumor-associated macrophages behave differently as they are predominantly induced to be polarized into M2, a pro-tumorigenic type when recruited with the tumor tissue and thereby favoring the tumorigenesis. Polarization of macrophages into M1 or M2 subtypes plays a vital role in regulating tumor progression, metastasis, and clinical outcome, highlighting the importance of studying the factors driving this process. A substantial number of studies have demonstrated that ncRNAs are involved in the macrophage polarization based on their ability to drive M1 or M2 polarization and in this review we have described their functions and categorized them into oncogenes, tumor suppressors, Juggling tumor suppressors, and Juggling oncogenes.

Non-coding RNAs and ferroptosis: potential implications for cancer therapy

Summary: Ferroptosis is a distinct form of regulated cell death that differs from traditional programmed cell death. It is associated with various cellular signaling pathways and molecules, and alterations in the ferroptosis-regulating network can contribute to the development of diseases, including cancer. Suppression of ferroptosis is common in cancer cells, allowing them to survive and progress. However, cancer cells resistant to common chemotherapeutic drugs are highly susceptible to ferroptosis inducers, suggesting the potential of targeting ferroptosis for cancer treatment. Non-coding RNAs (ncRNAs) play a crucial role in regulating ferroptosis in cancer and hold promise as therapeutic targets.

Translational Modeling Identifies Synergy between Nanoparticle-Delivered miRNA-22 and Standard-of-Care Drugs in Triple-Negative Breast Cancer

Summary: The downregulation of miRNA-22 in TNBC is associated with upregulation of eEF2K protein, and miRNA-22 nanotherapy shows potential as an investigational therapeutic agent. A mechanistic model was developed to evaluate the translational potential of miRNA-22 nanotherapy, revealing dose-response relationship and optimal treatment frequency. Combining miRNA-22 nanotherapy with immune checkpoint inhibitors and standard-of-care drugs showed promising results in improving treatment outcomes for TNBC.

Lnc-uc.147 Is Associated with Disease Stage of Liver, Gastric, and Renal Cancer

Summary: This study aimed to investigate the role of long non-coding RNA lnc-uc.147 in different types of cancer and suggest its functional and regulation aspects. The results showed that deregulated expression of lnc-uc.147 was associated with histologic grade in hepatocellular carcinoma and tumor stage in clear cell renal and gastric adenocarcinoma. Silencing lnc-uc.147 reduced the viability and clonogenic capacity of HepG2 cell lines. Additionally, a relationship between the transcription factor TEAD4 and lnc-uc.147 was suggested in liver and breast cancer cells.

Artificial miRNAs derived from miR-181 family members have potential in cancer therapy due to an altered spectrum of target mRNAs

Summary: miRNAs are noncoding RNAs that regulate genes and play a key role in cell homeostasis. The interaction between miRNAs and their target mRNAs is believed to depend on sequence complementarity, but some evidence suggests that miRNAs can adopt different structures that affect their function. By studying the oncogenic miR-181 family, we propose that the primary sequence and secondary structure of miRNAs may impact the number and range of targeted cellular transcripts. We also highlight that specific changes in miR-181 primary sequences may limit target gene selection and lead to the targeting of new transcripts with enhanced cancer-related functions.

Summary: The causes and consequences of abnormal biogenesis of extracellular vesicles (EVs) in breast cancers (BCs), especially estrogen receptor-positive (ER+) BCs, have been investigated. It was found that physiological doses of estrogen can promote EV secretion from ER+ BC cells by inhibiting miR-149-5p, which affects the regulatory activity of SP1 on the EV biogenesis factor nSMase2. In addition, the downregulation of miR-149-5p promotes the expression of hnRNPA1, responsible for loading let-7's miRNAs into EVs. This estrogen-driven mechanism leads to the elimination of tumor suppressor miRNAs in EVs, affecting tumor-associated macrophages in the microenvironment.

Going circular: history, present, and future of circRNAs in cancer

Summary: To date, thousands of circRNAs, which are highly abundant and conserved single-stranded RNA molecules shaped into ring structures, have been identified. CircRNAs have been shown to regulate gene expression and exhibit tissue- and development-specific expression patterns associated with normal and disease conditions, including cancer. They have drawn attention as potential diagnostic and prognostic biomarkers in cancer due to their stability and resistance to ribonucleases. However, their circular shape limits their annotation and a complete understanding of their functions is still lacking, making their detection and biomedical application challenging.
