The role of vitamin D in breast cancer risk and progression

Summary: The active form of vitamin D-3, 1,25-dihydroxyvitamin D-3 (1,25(OH)(2)D-3), plays a key role in regulating calcium and phosphate levels. Low levels of 25(OH)D, a marker for 1,25(OH)(2)D-3, are associated with an increased risk of breast cancer (BC) development. However, vitamin D-3 supplementation does not show a significant decrease in BC incidence. 1,25(OH)(2)D-3 or its analogues may have more potential anticancer activity when combined with existing cancer therapies.

Summary: Three novel 19-nor-1 alpha,25-dihydroxyvitamin D-3 derivatives modified at C1 and/or C3 with an amino group were synthesized to investigate the impact of substituting one or both hydroxyl groups of the A-ring with amino groups on the affinity for VDR and hDBP, as well as the antiproliferative activity. The A-ring precursors were obtained through a desymmetrization reaction of a prochiral 1,3-diol catalyzed by Pseudomonas cepacia lipase, and the diastereoisomers were identified by NMR spectroscopy.

Structural analysis and biological activities of C25-amino and C25-nitro vitamin D analogs

Summary: This article describes the structural analysis and biological evaluation of two derivatives of 1,25-dihydroxyvitamin D3 with modifications limited to the replacement of the 25-hydroxyl group by a 25-amino or 25-nitro groups. Both compounds are agonists of the vitamin D receptor and mediate biological effects similar to 1,25-dihydroxyvitamin D3. The 25-amino derivative is the most potent one while being less calcemic than 1,25-dihydroxyvitamin D3. The in vivo properties of these compounds make them potentially valuable for therapeutic purposes.

Summary: Neuropilin 2 (NRP2) plays a role in bone homeostasis by regulating osteoblasts and osteoclasts. The expression of NRP2 in osteoblasts is induced by 1,25-dihydroxyvitamin D-3. Deletion of NRP2 in osteoblasts results in reduced bone mass, while deletion in osteoclasts does not have a bone phenotype.

Summary: This study demonstrates that BXL-62 and Gemini-72, two C-20-modified vitamin D analogs, restore the transcriptional activities of VDR variants unresponsive to the natural ligand. The elucidated mechanisms of action highlight the mutual adaptation between the ligand and the VDR ligand-binding pocket.

Local nebulization of 1α,25(OH)2D3 attenuates LPS-induced acute lung inflammation

Summary: Local delivery of vitamin D to the lungs efficiently reduced inflammatory cells induced by LPS in bronchoalveolar lavage and slightly attenuated an increase in CXCL5. In cases of severe vitamin D deficiency, despite not significantly minimizing cellular inflammation, local vitamin D prevented epithelial barrier leakage and damage in the lung. Additional research is needed to determine the long-term beneficial effects of local vitamin D delivery on lung inflammation.
