Summary: Numerous studies have shown that anti-inflammatory agents can have antitumor effects in cancer therapy by directly increasing apoptosis and sensitivity to therapies, and decreasing invasion and metastasis. In this study, ebastine, a H1-histamine receptor antagonist, was screened and found to have antitumor effects in osteosarcoma cells. Ebastine treatment induced cell cycle arrest, inhibited metastasis, and induced apoptosis and autophagy both in vitro and in vivo. Mechanistically, ebastine activated the AMPK/ULK1 signaling pathway, triggering proapoptotic autophagy. IPMK was identified as a positive regulator of AMPK in osteosarcoma cells, and IPMK knockdown reversed the effects of ebastine on autophagy and AMPK/ULK1 signaling. This study suggests that ebastine has potential as a therapeutic candidate for osteosarcoma.

Summary: The study investigated the antileukemia effects of HDAC inhibitor chidamide in AML cells and its synergistic activity with the cytotoxic agent adriamycin. Chidamide suppressed EZH2, H3K27me3, and DNMT3A levels, exerted potential antileukemia activity, and increased sensitivity to adriamycin by disrupting the Smo/Gli-1 pathway and downstream signaling target p-AKT. Inhibition of EZH2 by chidamide or shEZH2 enhanced the antileukemia activity of adriamycin in vitro and in vivo.

Summary: Minimal residual disease (MRD) is the main cause of relapse in solid cancers and leukemias. In chronic myeloid leukemia (CML), autophagy plays a crucial role in maintaining the survival of drug resistant leukemic stem cells. Inhibition of autophagy-initiating kinase ULK1 could lead to stress-induced differentiation of leukemic stem cells, making them sensitive to drug treatment, providing a promising strategy for selective eradication of leukemic stem cells in CML patients.

Summary: This study aims to investigate the autophagy mediating mechanism underlying the neuroprotective effect of acupuncture in traumatic brain injury (TBI) rats. The results showed that acupuncture promoted neuronal autophagy on Day 3 after TBI, but inhibited it on Day 7 and Day 14, alleviating nerve damage. The mTOR/ULK1 pathway may be involved in the regulatory effect of acupuncture on autophagy in different stages of TBI.
