Nanoparticles as Vaccines to Prevent Arbovirus Infection: A Long Road Ahead

Arthropod-borne viruses (arboviruses) are a global public health concern, and vaccination is considered an effective strategy. Nanoparticles have shown potential as new vaccine platforms against medically important arboviruses, particularly Chikungunya virus, Dengue virus, Zika virus, Japanese encephalitis virus, and West Nile virus. Research has demonstrated that nanoparticles can enhance neutralizing antibody production in mice, but further efforts are needed to fully utilize their potential as vaccines for arboviruses.
Arthropod-borne viruses (arboviruses) are a significant public health problem worldwide. Vaccination is considered one of the most effective ways to control arbovirus diseases in the human population. Nanoparticles have been widely explored as new vaccine platforms. Although nanoparticles' potential to act as new vaccines against infectious diseases has been identified, nanotechnology's impact on developing new vaccines to prevent arboviruses is unclear. Thus, we used a comprehensive bibliographic survey to integrate data concerning the use of diverse nanoparticles as vaccines against medically important arboviruses. Our analysis showed that considerable research had been conducted to develop and evaluate nanovaccines against Chikungunya virus, Dengue virus, Zika virus, Japanese encephalitis virus, and West Nile virus. The main findings indicate that nanoparticles have great potential for use as a new vaccine system against arboviruses. Most of the studies showed an increase in neutralizing antibody production after mouse immunization. Nevertheless, even with significant advances in this field, further efforts are necessary to address the nanoparticles' potential to act as a vaccine against these arboviruses. To promote advances in the field, we proposed a roadmap to help researchers better characterize and evaluate nanovaccines against medically important arboviruses.

The results showed that VOCs bind more efficiently to human ACE2, and ACE2 protein variations can weaken the interaction and protein stability with SARS-CoV-2.

Summary: The rapid spread of SARS-CoV-2 led to the COVID-19 pandemic, prompting research into drugs and vaccines to prevent or treat the infection. Despite two years of efforts, there is still much to be understood about the virus' pathology. Transcriptomics and deep learning algorithms were used to identify differentially expressed targets and potential inhibitors for COVID-19 treatment, offering a helpful tool for personalized screening of molecules.

Ultrastructural analysis of monkeypox virus replication in Vero cells

Summary: In May 2022, the first worldwide outbreak of monkeypox virus (MPXV) was reported, showing different clinical aspects compared to previous human monkeypox infections. This study used transmission electron microscopy (TEM) to evaluate the ultrastructural features of MPXV infection in Vero cells. The results revealed various aspects of the MPXV cycle, including the entry of intracellular mature virus particles, reorganization of the endoplasmic reticulum, and distinct regions in the viral factory. The findings also showed viral crescents, different MPXV particle morphotypes, and cellular alterations induced by infection.

Diversity of Surface Fibril Patterns in Mimivirus Isolates

Summary: PCM is a severe mycosis caused by Paracoccidioides brasiliensis, mainly found in tropical countries. Itraconazole has marked antifungal activity against Pb and can activate both innate and acquired immunity. The effect of Itraconazole on Pb infection varies with different doses, with lower doses enhancing immune response and higher doses not requiring immune response.

Seroprevalence of SARS-CoV-2 in hospital workers in the southern region of Minas Gerais state in Brazil: An analysis of the pre-vaccine period

Summary: This study aimed to conduct a seroepidemiological survey of SARS-CoV-2 in three hospitals in the south of Minas Gerais state, Brazil. A total of 859 samples were collected between August and December 2020, and Enzyme-linked immunosorbent assays (ELISA) were performed on participants sera. The results showed that 21.6% of participants had positive serological tests, with higher prevalence in women and healthcare professionals.

Anti-Zika Virus Activity and Isolation of Flavonoids from Ethanol Extracts of Curatella americana L. Leaves

Summary: The ethanolic extract of Curatella americana showed significant in vitro anti-Zika virus activity. It had no cytotoxicity for MRC-5 cells and moderately cytotoxicity for Vero cells. The ethyl acetate fraction of the extract exhibited potent activity against the Zika virus, and several flavonoids isolated from this fraction also showed antiviral activity.

Evaluating Known Zika Virus NS2B-NS3 Protease Inhibitor Scaffolds via In Silico Screening and Biochemical Assays

Summary: The NS2B-NS3 protease (NS2B-NS3pro) is an important target for drug development against the Zika virus (ZIKV). This study utilized virtual screening and computational methods to target two non-peptide small-molecule scaffolds with inhibitory activity against ZIKV NS2B-NS3pro. While initial results were promising, further assays revealed artificial inhibition of the compounds. However, two compounds were found to exhibit antiviral properties in cellular-based assays.

Surface fibrils on the particles of nucleocytoviruses: A review

Summary: The capsid plays a central role in the life cycle of viruses, protecting the viral genome and facilitating host cell interaction and infection. Giant viruses that infect protists have attracted attention for their bacterial-like dimensions and unique features, such as surface fibrils. This mini review compiles important information on the surface fibrils of nucleocytoviruses, including their functions, composition, length, organization, and origins.
