Generation and functional characterization of a single-chain variable fragment (scFv) of the anti-FGF2 3F12E7 monoclonal antibody

Single-chain variable fragments (scFvs) are small-sized artificial constructs composed of the immunoglobulin heavy and light chain variable regions connected by a peptide linker. We have previously described an anti-fibroblast growth factor 2 (FGF2) immunoglobulin G (IgG) monoclonal antibody (mAb), named 3F12E7, with notable antitumor potential revealed by preclinical assays. FGF2 is a known angiogenesis-associated molecule implicated in tumor progression. In this report, we describe a recombinant scFv format for the 3F12E7 mAb. The results demonstrate that the generated 3F12E7 scFv, although prone to aggregation, comprises an active anti-FGF2 product that contains monomers and small oligomers. Functionally, the 3F12E7 scFv preparations specifically recognize FGF2 and inhibit tumor growth similar to the corresponding full-length IgG counterpart in an experimental model. In silico molecular analysis provided insights into the aggregation propensity and the antigen-recognition by scFv units. Antigen-binding determinants were predicted outside the most aggregation-prone hotspots. Overall, our experimental and prediction dataset describes an scFv scaffold for the 3F12E7 mAb and also provides insights to further engineer non-aggregated anti-FGF2 scFv-based tools for therapeutic and research purposes.

Summary: In this study, 13 fungal isolates with hydrolytic and ligninolytic abilities were selected from soil samples from the Atlantic Rain Forest Park incremented with sugar cane biomass, three of which showed xylanolytic activity and eleven showed cellulolytic abilities. Proteolytic enzymes were detected in three fungi, leading to their classification as metalloprotease and serine protease. Additionally, the strain Mucor sp. (SPZPF3_47) was highlighted for its ligninolytic abilities, particularly as this fungal genus had not been well described in the specific conditions of the study.

Probing the Conformational States of Thimet Oligopeptidase in Solution

Summary: This study investigates the conformational change of Thimet oligopeptidase (TOP) using fluorescence techniques and reveals that inhibitor binding induces a hinge-like movement in TOP. The catalytic activity of TOP is closely related to the Donor-Acceptor (D-A) distance, with shorter D-A distances corresponding to higher activities. This study provides insights into the hinge-bending motion of a metallopeptidase in solution, contributing to a better understanding of the mode of action of enzymes like neurolysin and angiotensin-converting enzyme 2 (ACE2).

Summary: Pseudomonas aeruginosa is often associated with life-threatening infections due to its intrinsic and acquired antimicrobial mechanisms, including different types of beta-lactamases. The study found that OXA-488 has a higher catalytic efficiency for benzylpenicillin and imipenem compared to OXA-50, although its carbapenemase activity is considered weak. Additionally, OXA-488 and OXA-494 have increased affinity for penicillins, contributing to improved catalytic efficiency against ampicillin.

Molecular Structure and Antibacterial Activity of Degradation Products from Cephalexin Solutions Submitted to Thermal and Photolytic Stress

Summary: This study investigated the structure and antibacterial activity of cephalexin solutions under heat stress and photolytic irradiation, revealing different chemical species among the degradation byproducts. While photolysis produced oxidized species, thermodegradation resulted in the production of inactive compounds. Microbiological analyses using E. coli showed that samples exposed to photolysis retained bactericidal power, while those exposed to thermal degradation lost their antimicrobial capacity.
