The presence of pharmaceuticals and personal care products (PPCPs) in natural water resources due to incomplete removal in Wastewater Treatment Plants (WWTPs) is a serious environmental concern at present. In this work, the effects of three pharmaceuticals (propranolol, triclosan, and nimesulide) on Gammarus pulex metabolic profiles at different doses and times of exposure have been investigated by liquid chromatography coupled to high-resolution mass spectrometry (LC-HRMS). The complex data sets generated in the different exposure experiments were analyzed with the ROIMCR procedure, based on the selection of the MS regions of interest (ROI) data and on their analysis by the Multivariate Curve-Resolution Alternating Least Squares (MCR-ALS) chemometrics method. This approach, allowed the resolution and identification of the metabolites present in the analyzed samples, as well as the estimation of their concentration changes due to the exposure experiments. ANOVA Simultaneous Component Analysis (ASCA) and Partial Least Squares Discriminant Analysis (PLS-DA) were then conducted to assess the changes in the concentration of the metabolites for the three pharmaceuticals at the different conditions of exposure. The three tested pharmaceuticals changed the concentrations of metabolites, which were related to different KEGG functional classes. These changes summarize the biochemical response of Gammarus pulex to the exposure by the three investigated pharmaceuticals. Possible pathway alterations related to protein synthesis and oxidative stress were observed in the concentration of identified metabolites. (C) 2020 Elsevier Ltd. All rights reserved.

Increasing HNF4α expression through synthetic antisense long non-coding RNA improves HCC cell function, slows down glycolysis, accelerates gluconeogenesis, improves dysregulated cholesterol metabolism, and inhibits migration, invasion, and proliferation of HCC cells.

Their convenience in storage and utilization, as well as their lack of cell therapy concerns, make them a promising approach for angiogenesis induction and other therapeutic functions in ischemic disorders.

Impact of the antibiotic doxycycline on the D. magna reproduction, associated microbiome and antibiotic resistance genes in treated wastewater conditions

Summary: Wastewater Treatment Plant (WWTP) effluents are significant sources of antibiotics, antibiotic resistance genes (ARGs), and resistant bacteria, posing threats to aquatic biota and human health. This study explores the impact of the antibiotic doxycycline on the microbiome of Daphnia magna in lab water and treated wastewater media. The results indicate that doxycycline and the media significantly influence the composition of the Daphnia-associated microbiome, with notable changes in Enterococcus spp. Furthermore, doxycycline reduces the presence of Limnohabitans sp. and impairs Daphnia reproduction in lab water, while treated wastewater increases microbiome diversity, richness, and promotes fecundity. The study also detects various ARG genes, including qnrS1, sul1, and blaTEM, with a higher abundance in treated wastewater compared to lab water.

(-)-Epigallocatechin-3-gallate induced apoptosis by dissociation of c-FLIP/Ku70 complex in gastric cancer cells

Summary: The anti-cancer properties of EGCG involve induction of apoptosis, inhibition of cell proliferation and histone deacetylase. This study examined the role of EGCG in gastric cancer cells by dissociating the c-FLIP/Ku70 complex and found that EGCG induced apoptosis, changed the expression of various proteins involved in cell cycle regulation, and decreased the interaction between c-FLIP and Ku70 while increasing Ku70 nuclear content. By dissociating the c-FLIP/Ku70 complex, EGCG could improve the therapeutic outcome in combination with other cancer therapy protocols.

Therapeutic implications of targeting autophagy and TGF-& beta; crosstalk for the treatment of liver fibrosis

Summary: Liver fibrosis is characterized by excessive deposition and accumulation of extracellular matrix components, mainly collagens, in response to various triggers. Autophagy, a homeostatic system for cell survival, and transforming growth factor-beta 1 (TGF-β1) play important roles in liver fibrosis. Recent studies have shown that TGF-β1 regulates autophagy, which affects pathophysiological aspects of liver fibrosis. This review highlights the cellular and molecular mechanisms of autophagy, its regulation by TGF-β1, and the potential for simultaneous inhibition of these pathways in anti-fibrotic therapy.

Non-target ROIMCR LC-MS analysis of the disruptive effects of TBT over time on the lipidomics of Daphnia magna

Subcutaneous Injection of Allogeneic Adipose-Derived Mesenchymal Stromal Cells in Psoriasis Plaques: Clinical Trial Phase I

Summary: The safety and potential efficacy of injecting allogeneic adipose-derived mesenchymal stromal cells (ADSCs) in psoriatic patients were evaluated in this study. The results showed that after ADSCs injection, the lesions of the patients improved and no major adverse effects were observed. Additionally, the expression levels of pro-inflammatory factors in the patient blood samples were reduced, indicating modulation of inflammation after ADSCs administration. Overall, ADSC injection could be considered as a safe and effective therapeutic approach for psoriatic plaques.

Burn injuries are devastating trauma with long-term medical care and high costs. Post-burn scars are inevitable, making the development of new treatment modalities necessary. This review aims to provide insight into the use of PRP as an adjuvant therapy for burn injuries and subsequent scars.

Summary: Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disease worldwide. However, the lack of reliable in vitro models has limited drug development studies, resulting in no approved medicine for NAFLD treatment. A biomimetic in vitro liver model requires an optimal natural microenvironment with appropriate cellular composition and specific bio-molecules to mimic native tissue conditions. Bioengineered three-dimensional tissues, such as microtissues and organoids, and infusion-based cultivation systems like microfluidics can improve the physiological function of in vitro constructs.
