Tumour inflammation signature and expression of S100A12 and HLA class I improve survival in HPV-negative hypopharyngeal cancer

Summary: The study identified a subset of HPV-negative HPSCC patients with favorable prognosis, showing gene expression signature characterized by overexpression of calgranulins and immune genes. High immune activity was found to be strongly related to survival in HPV-negative HPSCC, indicating a potential benefit of immunotherapy. Low expression of HLA class I and S100A12 could potentially be used to select patients for local surgery.

Single-Cell Deconvolution of Head and Neck Squamous Cell Carcinoma

Summary: Tumors are composed of diverse cells rather than a uniform ball of cells. Recent studies have shown that deconvolution algorithms can be used to computationally estimate cell-type proportions from gene expression data, although their performance in tumor tissues remains unclear.

Proteomic Profiling of Tissue Exosomes Indicates Continuous Release of Malignant Exosomes in Urinary Bladder Cancer Patients, Even with Pathologically Undetectable Tumour

Maria Eldh, Michael Mints, Stefanie Hiltbrunner, Sam Ladjevardi, Farhood Alamdari, Markus Johansson, Tomasz Jakubczyk, Rosanne E. Veerman, Ola Winqvist, Amir Sherif, Susanne Gabrielsson

Summary: The study investigated the release of exosomes from urinary bladder tissue post-TUR-B in UBC patients to find cancer-promoting exosomes originating from both tumour-proximal and tumour-distant sites, highlighting the need for early radical cystectomy. This suggests that the entire bladder tissue may release exosomes contributing to metastasis despite the absence of detectable tumour, supporting the role of exosomes as potential biomarkers and mediators of metastatic spread. Additionally, the enrichment of cancer-related pathways in tissue-derived exosomes from both sites strengthens the hypothesis and emphasizes the necessity for early removal of the bladder in UBC patients.

Summary: Through whole-exome sequencing of primary tumors and relapses in HPV+ TSCC/BOTSCC patients, we identified a specific CDC27 deletion as a potential marker for recurrence and highlighted the significance of mutated genes like BCLAF1 and AQP7 for further investigation.

Suva, Sidharth V. Puram, Itay Tirosh

Summary: This study curated and integrated data from 77 studies to analyze transcriptional intratumor heterogeneity (ITH) patterns across 1,163 tumor samples, identifying 41 consensus meta-programs and mapping them into 11 hallmarks of transcriptional ITH. The meta-programs cover diverse cellular processes and most of them are similar to those found in non-malignant cells.

Cellular states are coupled to genomic and viral heterogeneity in HPV-related oropharyngeal carcinoma

Zevallos, Jessica Silva-Fisher, Itay Tirosh

Summary: Single-cell analysis reveals high levels of inter- and intratumoral heterogeneity in head and neck squamous cell carcinoma (HNSCC), specifically in human papillomavirus (HPV)-positive and HPV-negative oropharynx tumors. Divergent patterns of HPV gene expression within HPV-positive tumors have functional effects on treatment resistance.
