Summary: Tape-strips and biopsies capture overlapping but distinct features of the AD molecular signature. Tape-strips are more sensitive to dysregulation of innate immunity and Th2/Th17-associated genes, while biopsies are more sensitive to Th22 and dermal cytokines. Both techniques detect epidermal barrier abnormalities, with tape-strips better representing terminal differentiation defects and biopsies better detecting epidermal hyperplasia changes.

Summary: Atopic dermatitis is an inflammatory disorder characterized by loss of epidermal barrier function and Th2 immune responses. The heterogeneity and complexity of the disease limit the rational use of current therapies, but the development of advanced therapeutic agents and nanocarriers shows promise. However, there are challenges such as the understanding of disease mechanisms, adverse effects of current therapies, and inadequacy of animal models. Despite these challenges, nanocarriers have shown excellent preclinical results in the treatment of AD.

Summary: Topical agents currently used for atopic dermatitis treatment may be effective for mild or mild-moderate cases, but more complex or unresponsive cases often require additional systemic therapies. The most commonly used compounds are topical corticosteroids and calcineurin inhibitors, although they may have side effects, low response rates, and poor patient compliance. Therefore, the development of new drugs with higher efficacy and safety is necessary.

Sampling the stratum corneum with non-invasive tape strips may be useful for identifying biomarkers associated with disease severity, food allergy, and filaggrin mutations.

Topical bismuth oxide-manganese composite nanospheres alleviate atopic dermatitis-like inflammation

Summary: The study found that Bi2-xMnxO3 nanospheres (BM) were effective in alleviating skin inflammation in AD-like C57BL/6 mice. BM treatment reduced the level of inflammatory factors and increased the expression of barrier proteins in the skin. Furthermore, BM reduced the expression of phosphorylated STAT6, which resulted in decreased expression of GATA3 and Th2 cells, leading to a reduction in IL-4 expression.

Cutaneous body image in psoriasis: The role of attachment style and alexithymia

The study found that, independent of disease severity and depressive symptoms, patients with an insecure-avoidant attachment style and higher levels of alexithymia reported greater cutaneous body dissatisfaction.

Atopic Dermatitis and Ulcerative Colitis Successfully Treated with Upadacitinib

Summary: JAK inhibitors have shown efficacy in treating immune-related diseases such as ulcerative colitis and atopic dermatitis. This case report demonstrates the successful management of a patient with overlapping conditions of ulcerative colitis and atopic dermatitis using upadacitinib, resulting in complete and persistent clinical remission. These findings suggest that upadacitinib may be an optimal treatment strategy for individuals with coexisting Th1 and Th2-mediated conditions.

The molecular findings support the clinical analysis that apremilast may enhance the repigmentation effects of NB-UVB in generalized vitiligo.
