12-O-Tetradecanoylphorbol-13-acetate (TPA) is the most widely used diacylglycerol (DAG) mimetic agent and inducer of protein kinase C (PKC)-mediated cellular response in biomedical studies. TPA has been proposed as a pluripotent cell differentiation factor, but results obtained have been inconsistent. In the present study we show that TPA can be applied as a cardiomyogenesis-promoting factor for the differentiation of mouse embryonic stem (mES) cells in vitro. The mechanism of TPA action is mediated by the induction of extracellular signal-regulated kinase (ERK) activity and the subsequent phosphorylation of GATA4 transcription factor. Interestingly, general mitogens (FGF, EGF, VEGF and serum) or canonical WNT signalling did not mimic the effect of TPA. Moreover, on the basis of our results, we postulate that a TPA-sensitive population of cardiac progenitor cells exists at a certain time point (after days 6-8 of the differentiation protocol) and that the proposed treatment can be used to increase the multiplication of ES cell-derived cardiomyocytes.

Cross-Linked Gelatine by Modified Dextran as a Potential Bioink Prepared by a Simple and Non-Toxic Process

Summary: In this study, a novel cytocompatible material suitable for extrusion-based 3D printing was designed. Hydrogels were prepared by cross-linking gelatins with dextran polyaldehyde, exhibiting desirable rheological behavior and cell-friendly environment. The hydrogels showed strong shear-thinning behavior, quick gelation, and formed self-supported porous structures. Additionally, the hydrogels were non-cytotoxic and allowed homogeneous cell distribution within the printed scaffolds.

Summary: The study found that HA-atRA upregulated gene expression similarly to atRA, but HA-atRA activated the expression of cholesterol metabolism genes. The results showed that HA-atRA removed cholesterol from the cells and induced cholesterol synthesis to replenish it. Additionally, HA-C18:1 also removed cholesterol from the cells, indicating that the cholesterol removal stemmed from the amphiphilic nature of these derivatives.

Summary: We report a new synthetic strategy for the flexible preparation of forskolin-like molecules. Stereoselective Michael addition and ring-opening metathesis paired with ring closure were key steps in achieving the concise assembly of the tricyclic core. Late-stage functionalization sequences transformed the tricyclic intermediates into a set of different forskolin-like molecules.

Single-cell RNA sequencing analysis of T helper cell differentiation and heterogeneity

Summary: Single-cell transcriptomics is a powerful tool for investigating the biological characteristics of cells. In this study, human T cells were analyzed using single-cell transcriptomic approach with support of labelled antibodies and comprehensive bioinformatics analysis. The results revealed differences in expression profiles among different T cell subpopulations, providing insights into the importance of each step of the analysis and aiding in a more practical understanding of the results.

Summary: On-chip vascular microfluidic models using PDMS have been widely used to study cardiovascular diseases. However, the hydrophobic surface of PDMS needs to be modified for biological applications. This study introduces a high-frequency low-pressure air-plasma surface modification method that improves the surface properties of seamless channels within a PDMS microfluidic chip, enabling efficient adhesion and long-term culture of vascular endothelial cells under flow conditions.

Magneto-responsive hyaluronan hydrogel for hyperthermia and bioprinting: Magnetic, rheological properties and biocompatibility

Summary: In this study, a magneto-responsive hydrogel based on hyaluronan was fabricated, which exhibited good mechanical properties and thermosensitivity. It was found that the hydrogel could rapidly heat under exposure to low amplitude radio frequency alternating magnetic field. In addition, the hydrogel showed cytocompatibility and 3D printability, making it suitable for biomedical applications.

A forskolin-mediated increase in cAMP promotes T helper cell differentiation into the Th1 and Th2 subsets rather than into the Th17 subset

Summary: The adenylyl cyclase (AC) signaling pathway is suggested to play a key role in regulating immune system functions. This study demonstrates that cyclic adenosine mono-phosphate (cAMP) affects the differentiation of T helper (Th) cells, enhancing Th1 cell phenotype, supporting Th2 differentiation but negatively affecting cell metabolism, and suppressing Th17 immunophenotype while upregulating CXCL13.
