Summary: Osteosarcoma, the most common primary bone malignancy in children and adolescents, has a high mortality rate due to its rapid growth and metastatic potential. The JAK2/STAT3 signaling pathway, critical for osteosarcoma occurrence and development, is a potential target for treatment. Telocinobufagin (TCB), a natural product isolated from toad cake, was identified as a potent candidate with anti-osteosarcoma effects. TCB inhibited osteosarcoma cell growth, migration, invasion, and induced cancer cell apoptosis by specifically inhibiting the JAK2/STAT3 signaling pathway. In an osteosarcoma xenograft animal model, TCB significantly suppressed tumor growth and metastasis. Moreover, TCB showed strong inhibitory effects in other cancer types such as lung cancer, liver cancer, colon cancer, breast cancer, and gastric cancer.

Summary: Folate-modified liposomes loaded with resveratrol were prepared and investigated for their anti-osteosarcoma effects in vitro and in vivo. The results showed that folate-modified liposomes significantly increased the uptake of resveratrol by osteosarcoma cells and inhibited tumor proliferation, migration, and induced apoptosis more effectively than free resveratrol and liposomes. This strategy has great potential for the treatment of osteosarcoma.

Further molecular mechanism analysis revealed the involvement of the JAK2/STAT3 signaling pathway in the antitumor effect of Cyy260.

Summary: Mammalian oocytes undergo morphological, structural, and molecular changes during a process called oocyte aging. The SUMO pathway, specifically SUMO-2/3 and the SUMO-specific protease SENP-2, are involved in spindle morphology changes and chromosome movements during oocyte aging. Decreased UBC9 and differential ubiquitination patterns also correlate with in vitro oocyte aging. These findings suggest that age-related changes in SUMOylation and the deSUMOylation of key target proteins may contribute to spindle and chromosome alignment defects during mammalian oocyte postovulatory aging, with implications for ART-related human oocyte aging and fertilization success.

Summary: In this study, an animal model of pulmonary fibrosis complicated with lung cancer was evaluated. It was found that exogenous recombinant human thymosin beta 4 (exo-rhT beta 4) can alleviate the impairment caused by pulmonary fibrosis, inhibit the growth of lung cancer tumor cells, and effectively inhibit the JAK2-STAT3 signaling pathway. The establishment of the IPF-LC animal model is helpful for drug development and exogenous rhT beta 4 shows potential for the treatment of IPF and LC.
