Generation of two gene edited iPSC-lines carrying a DOX-inducible NGN2 expression cassette with and without GFP in the AAVS1 locus

Benjamin Schmid, Bjorn Holst, Ulla Poulsen, Ida Jorring, Christian Clausen, M. Rasmussen, Ulrike A. Mau-Holzmann, Rachel Steeg, Hugh Nuthall, A. Ebneth, A. Cabrera-Socorro

Summary: Neurog2 is a gene encoding the neuronal transcription factor NGN2, converting stem cells into functional neurons efficiently. Two iPS cell lines, BIONi010-C-13 and BIONi010-C-15, were generated with DOX-inducible constructs of neurog2, allowing differentiation into neurons when DOX is added.

EBiSC best practice: How to ensure optimal generation, qualification, and distribution of iPSC lines

Rachel Steeg, Sabine C. Mueller, Nancy Mah, Bjorn Holst, Alfredo Cabrera-Socorro, Glyn N. Stacey, Paul A. De Sousa, Aidan Courtney, Heiko Zimmermann

Summary: The generation of disease-relevant human induced pluripotent stem cells requires adherence to ethical, legal, and quality standards, with best practices integrated into daily operations for maximizing their potential. Key experiences from operating the European Bank for induced Pluripotent Stem Cells (EBiSC) over 7 years are discussed, with recommendations on how to incorporate solutions into daily management framework.

Neubauer, Christian Clausen, Catherine Verfaillie, Andreas Ebneth, Mattias Hansson, Rachel Steeg, Heiko Zimmermann

Summary: Induced pluripotent stem cells (iPSCs), with their ability to differentiate into various cell types, have become an important tool in biomedical research. This article discusses the development of scalable methods for large-scale production of iPSCs using 3D suspension culture platforms. It also highlights the establishment of robust differentiation protocols for cardiac, neural, and hepatic cell types to meet the increasing demand for research tools.

Summary: In this study, an optimized protocol for generating hepatocyte-like cells (HLCs) and hepatic organoids from hiPSCs was presented. The protocol was further translated to scalable suspension-based bioreactors to meet the increasing demand of hepatic cells. Cryopreservation approaches were also analyzed to ensure the maintenance of organoid phenotype.

SOX9-induced Generation of Functional Astrocytes Supporting Neuronal Maturation in an All-human System

Katrien Neyrinck, Johanna Van den Daele, Tim Vervliet, Jonathan De Smedt, Keimpe Wierda, Melissa Nijs, Tom Vanbokhoven, Astrid D'hondt, Melanie Planque, Sarah-Maria Fendt, Pei-Yu Shih, Frederik Seibt, Juan Pita Almenar, Mohamed Kreir, Devesh Kumar, Vania Broccoli, Geert Bultynck, Andreas Ebneth, Alfredo Cabrera-Socorro, Catherine Verfaillie

Summary: By overexpressing SOX9 in pluripotent stem cells, functionally mature human astrocytes can be generated, displaying comparable functional properties to primary human astrocytes. The electrophysiological properties of neurons co-cultured with these astrocytes are similar to murine primary cultures. This method allows for scaled-up production for high-throughput analysis, aiding in drug development and reducing animal use in biomedical research.
