TheOstreid herpesvirus 1species affects shellfish, contributing significantly to high economic losses during production. To counteract the threat related to mortality, there is a need for the development of novel point-of-care testing (POCT) that can be implemented in aquaculture production to prevent disease outbreaks. In this study, a simple, rapid and specific colorimetric loop-mediated isothermal amplification (LAMP) assay has been developed for the detection ofOstreid herpesvirus1(OsHV-1) and its variants infectingCrassostrea gigas(C. gigas). The LAMP assay has been optimized to use hydroxynaphthol blue (HNB) for visual colorimetric distinction of positive and negative templates. The effect of an additional Tte UvrD helicase enzyme used in the reaction was also evaluated with an improved reaction time of 10 min. Additionally, this study provides a robust workflow for optimization of primers for uncultured viruses using designed target plasmid when DNA availability is limited.

The biosensor showed a low limit of detection and the ability to be reused multiple times. The experimental results demonstrated the high feasibility and accuracy of using this biosensor for detecting S. Typhi in clinical and food samples.

Inhibition of Listeria monocytogenes by Phage Lytic Enzymes Displayed on Tailored Bionanoparticles

Summary: The study demonstrated that tailored polyhydroxyalkanoate bionanoparticles (PHA BNPs) displaying recombinant listeriophage enzymes have the ability to lysine and inhibit the growth of Listeria monocytogenes. In laboratory conditions, BNPs displaying only the amidase domain of the phage endolysin were more effective than BNPs displaying the full-length endolysin. However, under conditions that better represent those found in food processing environments, BNPs displaying the full-length endolysin showed a greater inhibitory effect.

Food allergen recalls in the United Kingdom: A critical analysis of reported recalls from 2016 to 2021

Summary: This study analyzed allergen food recalls issued by food regulatory bodies in the UK from 2016 to 2021. The analysis included food type, allergenic food group, reasons for recall, and food expiry status. The findings revealed an increasing trend in allergen-related recalls until 2019, with a peak of 118 recalls, followed by a decrease to 82 and 84 recalls in 2020 and 2021 respectively. Milk was the most recalled allergenic food group, accounting for 25.2% of recalls. The omission of priority allergens from the list of ingredients was the main reason for recalls, and the supermarket Lidl issued the most recalls with 37 recalls involving 62 products. Some recalls with expiry dates posed microbiological risks to consumers, especially in cereal & bakery products.

A 3D paper microfluidic device for enzyme-linked assays: Application to DNA analysis

Summary: This paper presents a paper-based microfluidic device, called MEL-PAD, which can perform enzyme-linked assays. The MEL-PAD system utilizes a wash-free sandwich coupling and vertical flow to detect analytes. It achieves efficient washing and color change detection using nitrocellulose and a chromogenic substrate. The MEL-PAD demonstrates high sensitivity and versatility for quantifying various analytes. The article also shows the potential of MEL-PAD in detecting DNA from Staphylococcus epidermidis.

Single Nucleotide Variants (SNVs) of Angiotensin-Converting Enzymes (ACE1 and ACE2): A Plausible Explanation for the Global Variation in COVID-19 Prevalence

Summary: This study explored the prevalence of single nucleotide polymorphisms (SNPs) in ACE1 and ACE2 in different geographic locations and their association with COVID-19 prevalence. The DD genotype of ACE1 was found to be more prevalent in Europe and America, which had a higher impact of COVID-19 compared to Asia and Africa. The rs2285666 SNP of ACE2 varied in different locations. However, the association between ACE2 and COVID-19 remains inconclusive and further research is needed.

Detection of Ciguatoxins and Tetrodotoxins in Seafood with Biosensors and Other Smart Bioanalytical Systems

Summary: This article provides an overview of the detection of ciguatoxins (CTXs) and tetrodotoxins (TTXs) using biorecognition molecules and various assay configurations and transduction strategies. The advantages and limitations of systems based on cells, receptors, antibodies, and aptamers are described, along with identified challenges in marine toxin detection. The validation of these bioanalytical systems through sample analysis and comparison with other techniques is also discussed. These tools have shown promise in detecting and quantifying CTXs and TTXs, making them valuable for research and monitoring programs.
