Summary: PCGEM1 plays a crucial role in the progression of prostate cancer by sponging miR-129-5p as a ceRNA of CDT1. Both PCGEM1 and CDT1 are upregulated in prostate cancer, while miR-129-5p is downregulated and negatively correlated with PCGEM1 and CDT1. Downregulation of PCGEM1 or CDT1 inhibits the viability, promotes apoptosis and cycle arrest of prostate cancer cells in vitro, and controls tumor growth in vivo.

Summary: This study investigated the expression level and prognostic value of UCA1 in pancreatic cancer tissues, as well as its effects and mechanism in regulating cell proliferation, apoptosis, and metastasis. The results showed that high expression of UCA1 was associated with poor prognosis in pancreatic cancer. Downregulation of UCA1 inhibited cell proliferation, migration, invasion, and induced apoptosis. It was also found that UCA1 regulated the expression of BRCC3 by inhibiting miR-582-5p.

Summary: Endothelial dysfunction is crucial in vascular disease pathogenesis. Long noncoding RNA (lncRNA) and microRNA (miRNA) play significant roles in various cellular processes in vascular endothelial cells (VECs), including cell growth, migration, autophagy, and apoptosis. The present study investigated the role of plasmacytoma variant translocation 1 (PVT1) in regulating autophagy and apoptosis in human umbilical vein endothelial cells (HUVEC). It was found that PVT1 knockdown accelerates apoptosis and suppresses cellular autophagy. PVT1 functions as a competing endogenous RNA (ceRNA) of miR-15b-5p and miR-424-5p, promoting cellular autophagy and down-regulating apoptosis. This study suggests PVT1 as a potential therapeutic target for cardiovascular disease treatment.

Summary: In this study, bioinformatic analysis and experimental validation revealed that miR-26b-5p was downregulated in Helicobacter pylori-infected gastric cancer (GC) cells and GC tissues. Low expression of miR-26b-5p promoted GC proliferation in vitro and in vivo and was associated with poor prognosis in GC patients. Mechanistically, miR-26b-5p directly targeted PDE4B and CDK8, leading to decreased phosphorylation and nuclear translocation of STAT3, which regulated GC proliferation. Importantly, miR-26b-5p was transcriptionally suppressed by STAT3, forming a miR-26b-5p-PDE4B/CDK8-STAT3 positive feedback loop. This newly identified feedback loop may serve as a promising therapeutic target for inflammation-related GC progression.

Summary: Mounting evidence suggests that circular RNAs (circRNAs) play important roles in the development and progression of cancers. In this study, researchers identified a specific circRNA, circXPO1, that was significantly upregulated in glioblastomas (GBM). Functional studies demonstrated that circXPO1 promoted GBM cell proliferation, migration, and tumor formation. Further investigation revealed that circXPO1 directly bound to miR-7-5p, which negatively regulated the RAF1 gene expression. These findings suggest that circXPO1 may be a potential therapeutic target for GBM treatment.

Summary: This study assessed the prognostic performances of three pathological classifications in gastric cancer patients and developed a novel prognostic nomogram for individually predicting overall survival (OS). The results showed that the modified Lauren classification demonstrated superior predictive ability for OS, and the new prognostic nomogram incorporating this classification outperformed the American Joint Committee on Cancer 8th edition tumor-nodemetastasis classification in terms of discriminative ability, model fitting performance, and net benefits.

Summary: The emergence of CRISPR-based genome editing systems has revolutionized the field of molecular biology and holds great promise for the treatment of various diseases. However, the lack of suitable delivery vehicles for effective intracellular delivery of CRISPR components limits its practical applications. In this study, a nanometer-scaled vesicle platform (CRISPRsome) was developed to efficiently deliver the essential components of the CRISPR system, resulting in significant gene knockout efficacy in cancer cells. This study highlights the potential of CRISPRsome in facilitating the use of CRISPR technology in practical applications.
