Multi-slice representational learning of convolutional neural network for Alzheimer's disease classification using positron emission tomography

Background Alzheimer's Disease (AD) is a degenerative brain disorder that often occurs in people over 65 years old. As advanced AD is difficult to manage, accurate diagnosis of the disorder is critical. Previous studies have revealed effective deep learning methods of classification. However, deep learning methods require a large number of image datasets. Moreover, medical images are affected by various environmental factors. In the current study, we propose a deep learning-based method for diagnosis of Alzheimer's disease (AD) that is less sensitive to different datasets for external validation, based upon F-18 fluorodeoxyglucose positron emission tomography/computed tomography (FDG-PET/CT). Results The accuracy, sensitivity, and specificity of our proposed network were 86.09%, 80.00%, and 92.96% (respectively) using our dataset, and 91.02%, 87.93%, and 93.57% (respectively) using the Alzheimer's Disease Neuroimaging Initiative (ADNI) dataset. We observed that our model classified AD and normal cognitive (NC) cases based on the posterior cingulate cortex (PCC), where pathological changes occur in AD. The performance of the GAP layer was considered statistically significant compared to the fully connected layer in both datasets for accuracy, sensitivity, and specificity (p < 0.01). In addition, performance comparison between the ADNI dataset and our dataset showed no statistically significant differences in accuracy, sensitivity, and specificity (p > 0.05). Conclusions The proposed model demonstrated the effectiveness of AD classification using the GAP layer. Our model learned the AD features from PCC in both the ADNI and Severance datasets, which can be seen in the heatmap. Furthermore, we showed that there were no significant differences in performance using statistical analysis.

Summary: This study aims to compare the segmentation performance of current prominent deep learning algorithms with ground-truth segmentations and validate the reproducibility of manually created four cardiac chamber ground-truth annotations in 2D echocardiography. The results show that the three current prominent DL-based fully automated methods are able to reliably perform four-chamber segmentation and quantification of clinical indices, with some degree of inter-observer variability.

Summary: This study evaluated the prognostic value of metabolic parameters on F-18-FDG PET/CT and tumor dose (TD) on post-treatment Y-90 PET/CT in HCC patients undergoing Y-90 transarterial radioembolization (TARE). Impact of Exogenous Treatment with Histidine on Hepatocellular Carcinoma Cells

Summary: This study examined the use of exogenous histidine treatment to enhance tumor responses to sorafenib in patients with advanced hepatocellular carcinoma (HCC). Histidine treatment reduced the expression of tumor markers related to glycolysis, inflammation, angiogenesis, and stem cells in HCC cells. Combined treatment with sorafenib and histidine may be a novel therapeutic strategy to improve the sensitivity to sorafenib and enhance long-term survival in HCC.

Low C-11-ACE uptake in ODs is advantageous in overcoming the limitations of radiolabeled amino acid tracers.

Summary: Patients with low-glycolytic hepatocellular carcinoma (HCC) have better clinical outcomes compared to those with hypoxic and high-glycolytic HCC. The study found that SLC13A5/NaCT is an important solute carrier (SLC) in low-glycolytic HCCs. Extracellular citrate treatment in HCCs with high SLC13A5/NaCT expression led to reduced glucose uptake and tumor growth inhibition. This research suggests that extracellular nutrient administration related to SLCs can prevent hypoxic tumor progression in low-glycolytic HCC.

Cortical Thickness and Brain Glucose Metabolism in Healthy Aging

Summary: This study aimed to investigate the impact of demographic factors on cortical thickness and brain glucose metabolism in healthy aging individuals. A total of 71 subjects were assessed using neurological examination, 3-tesla magnetic resonance imaging, 18F-fluorodeoxyglucose positron-emission tomography, and neuropsychological tests. Cortical thickness and brain metabolism were measured using vertex- and voxelwise analyses, respectively. General linear models were used to determine the effects of age, sex, and education on cortical thickness and brain glucose metabolism. The study found that age and sex have differential effects on cortical thickness and brain metabolism, highlighting the importance of considering demographic factors in the analysis.

Patterns of striatal dopamine depletion and motor deficits in de novo Parkinson's disease

Summary: The study investigated whether different patterns of striatal dopamine depletion are associated with motor deficits in Parkinson's disease. A total of 404 drug-naive patients with early-stage PD were enrolled and underwent dopamine transporter imaging. Principal component analysis identified three patterns of dopamine depletion: overall deficiency, selective loss in the sensorimotor striatum, and symmetric loss in the striatum. Linear regression analysis revealed that overall deficiency and selective loss were associated with motor deficits, while symmetric loss was not. Mediation analysis showed that dopamine deficiency in the posterior putamen mediated the association between dopamine depletion patterns and motor deficits. These findings suggest that dopamine deficiency in the posterior putamen plays a crucial role in motor deficits in PD.
