There is enormous global anticipation for stem cell-based therapies that are safe and effective. Numerous pre-clinical studies present encouraging results on the therapeutic potential of different cell types including tissue derived stem cells. Emerging evidences in different fields of research suggest several cell types are safe, whereas their therapeutic application and effectiveness remain challenged. Multiple factors that in fluence treatment outcomes are proposed including immunocompatibility and potency, owing to variations in tissue origin, ex-vivo methodologies for preparation and handling of the cells. This communication gives an overview of literature data on the different types of cells that are potentially promising for regenerative therapy. As a case in point, the recent trends in research and development of the mesenchymal stem cells (MSCs) for cell therapy are considered in detail. MSCs can be isolated from a variety of tissues and organs in the human body including bone marrow, adipose, synovium, and perinatal tissues. However, MSC products from the different tissue sources exhibit unique or varied levels of regenerative abilities. The review finally focuses on adipose tissue-derived MSCs (ASCs), with the unique properties such as easier accessibility and abundance, excellent proliferation and differentiation capacities, low immunogenicity, immunomodulatory and many other trophic properties. The suitability and application of the ASCs, and strategies to improve the innate regenerative capacities of stem cells in general are highlighted among others. The study found that ADSC spheroids could be a promising treatment option for osteoarthritis, but further research on administration methods and their interaction with synovial fluid is necessary.

Summary: Mucoepidermoid carcinomas (MECs) are rare malignant tumors that were originally considered as benign. Diagnosis of MECs is challenging, especially for intraoral MECs. This case report highlights the importance of accurate histopathological evaluation for the diagnosis of MECs.

Summary: This report describes a rare case of chondroid syringoma (CS) of the lower lip, highlighting the importance of proper diagnosis, treatment, and prognostic evaluation. Surgical excision biopsy confirmed the diagnosis of benign CS, and no recurrence was observed during the 2-year follow-up period.

Management of temporomandibular joint diseases: a rare case report of coexisting calcium pyrophosphate crystal deposition and synovial chondromatosis

Summary: The coexistence of calcium pyrophosphate dihydrate crystal deposition (CPP) and synovial chondromatosis (SC) in the temporomandibular joint (TMJ) is rare. This report presents a case of a 61-year-old man with coexisting CPP and SC in the TMJ. The patient experienced pain and trismus in the right TMJ and was initially diagnosed with SC based on radiographic assessments. However, histopathology revealed deposits of rhomboid and rod-shaped crystals positive for birefringence, confirming the coexistence of CPP. Surgical removal of loose bodies and chalk-like lesions was performed and after a follow-up of 1.5 years, no recurrence was observed.

Regenerative medicine strategies for hair growth and regeneration: A narrative review of literature

Summary: Hair loss, associated with psychosocial and medical comorbidities, is a burdensome condition economically. Conventional medical interventions for alopecia have limitations, thus regenerative treatments like mesenchymal stem cell implantation, MSC-derived secretome treatment, and platelet-rich plasma therapies are being explored. This review summarizes the most promising regenerative treatments for alopecia.

Current regenerative medicine-based approaches for skin regeneration: A review of literature and a report on clinical applications in Japan

Summary: There is a growing interest among healthcare specialists and the public in using regenerative medicine-based approaches for skin regeneration. These approaches can be categorized into cell-based and cell-free therapies, and are reported to be safe and effective. This review provides an update on the different regenerative medicine-based therapies for skin regeneration and highlights the regulatory perspective on the clinical application of some of these therapies in Japan.

Exploring olfactory receptor family 7 subfamily C member 1 as a novel oral cancer stem cell target for immunotherapy

Summary: Despite advances in cancer therapies, the mortality rate of oral cancer has not improved in the past three decades. Oral cancers contain a subpopulation of cancer stem cells (CSCs) that contribute to cancer infiltration, recurrence, metastasis, and drug resistance. Cytotoxic CD8+ T lymphocytes (CTLs) play a crucial role in targeting CSCs and regulating cancer progression. This study investigates the potential of olfactory receptor family 7 subfamily C member 1 (OR7C1) as a target for CSC-targeted immunotherapy in oral cancer and finds that OR7C1 is expressed only in oral CSCs and is effectively recognized by CTLs.

Management of Axillary Contracture in Poland Syndrome: Differentiating Fibrous Band and Skin for Optimal Release

Summary: This report presents a case of a 10-year-old girl with Poland syndrome manifesting an axillary web containing a fibrous band, which was successfully corrected by a double-opposing Z-plasty. Histopathological findings indicated the presence of tendon-like tissue, highlighting the importance of appropriate treatment approaches for Poland syndrome. Addressing both tendon and skin components is essential for optimal contracture release.

Management of Rheumatoid Arthritis: Possibilities and Challenges of Mesenchymal Stromal/Stem Cell-Based Therapies

Summary: Rheumatoid arthritis is a chronic autoimmune disorder that primarily affects joints and muscles. Conventional treatments only address symptoms, and some patients experience adverse effects or poor responses. Transplanting multipotent adult stem cells, such as mesenchymal stromal/stem cells, has shown promise in controlling inflammation and promoting tissue regeneration in patients with refractory diseases, including rheumatoid arthritis. This review discusses the current status and challenges of MSC-based therapies for rheumatoid arthritis, offering opportunities for broader clinical application.

Summary: In this study, we evaluated the potential of oligo-tetra-PEG gel (Oligo gel) as a growth factor-releasing scaffold. Compared to tetra-PEG hydrogel (Tetra gel), Oligo gel underwent a higher degree of gel-gel phase separation, demonstrating better protein release and osteogenic potential, and achieving superior regeneration in a mouse calvarial defect model. Therefore, Oligo gel is considered a promising therapeutic biomaterial.

Development of triple-functionalized calcium phosphate nanoparticles as an advanced drug delivery system for bone tissue repair
