Summary: In this study, it was discovered that the long noncoding RNA (lncRNA) AFAP1-AS1 encodes a peptide called ATMLP, which is located on the mitochondria and promotes the malignancy of nonsmall cell lung cancer (NSCLC). The level of ATMLP increases as the tumor progresses and NSCLC patients with high levels of ATMLP have a poorer prognosis. ATMLP's translation is regulated by m(6)A methylation at a specific location of the AFAP1-AS1 gene. Mechanistically, ATMLP binds to NIPSNAP1 and inhibits its transport, which affects cell autolysosome formation. This study uncovers a complex regulatory mechanism of NSCLC malignancy orchestrated by a peptide encoded by a lncRNA. The potential application of ATMLP as an early diagnostic biomarker for NSCLC is also assessed comprehensively.
