Outer membrane vesicles (OMVs) produced by Gram-negative bacteria and their RNA content have recently received increasing attention as novel mediators of host-guest intercellular and interspecies communication. These kinds of vesicles allow not only the export of proteins, but also of DNA and other small RNAs. We previously reported that the production of bacterial vesicles could represent a way to communicate with the surrounding environment. This general mechanism, exploited by bacteria and viruses, allows conveying messages to the surrounding cells and tissues, thus leading to the manipulation of the host immune response. Many functions attributed to OMVs are a consequence of their high capability to survive in different environmental stressors and to enter into cells quite easily. As these properties can be exploited to advantage human health, in this review, we summarized the papers that reported the use of OMVs as modulators of cellular activities through the involvement of the RNAs contained within. We also discussed what has been done with OMVs and what remains to be discovered in order to prompt the investigations toward the complete elucidation of the role and functions of these vesicles.

The First Report of Bacteroides fragilis and Bacteroides thetaiotaomicron Relative Abundance in Obese Iranian Population

Vida Kazemi, Sara Ahmadi Badi, Azadeh Manayi, Fateme Ettehad Marvasti, Mahdi Bagheri, Arfa Moshiri, Mojtaba Mojtahedzadeh, Seyed Davar Siadat

Summary: This study investigated the abundance of two important Bacteroides strains in Iranian obese and normal-weight subjects for the first time. Although obese subjects had higher abundance of B. fragilis and B. thetaiotaomicron compared to normal-weight subjects, there was no significant difference in relative abundance between the two groups. The findings suggest that targeting the restoration of gut microbiota pattern could be valuable in controlling obesity.

Commensal and Pathogenic Bacterial-Derived Extracellular Vesicles in Host-Bacterial and Interbacterial Dialogues: Two Sides of the Same Coin

Samira Tarashi, Mohammad Saber Zamani, Mir Davood Omrani, Abolfazl Fateh, Arfa Moshiri, Ahmad Saedisomeolia, Seyed Davar Siadat, Stan Kubow

Summary: This review provides an overview of the protective and harmful roles of extracellular vesicles (EVs) derived from commensal and pathogenic bacteria in host-bacterial and interbacterial interactions. Bacterial EVs have important impacts on human health and metabolic homeostasis by regulating microbiota-host crosstalk and intestinal homeostasis.

Summary: The research on the relationship between gut microbiota and T2DM has exponentially grown in the past decade, revealing the anti-diabetic effects of interventions through alteration of gut microbiota. Studies suggest that obesity alongside T2DM may share similar pathways mediated by gut microbiota.

Nontuberculous Mycobacterial Resistance to Antibiotics and Disinfectants: Challenges Still Ahead

Samira Tarashi, Seyed Davar Siadat, Abolfazl Fateh

Summary: The mortality incidence from nontuberculous mycobacteria (NTM) infections has been increasing globally. Highly resistant NTM to antibiotics and disinfectants have emerged, making it difficult to control and treat infections. Further studies are crucial to uncover this association.

Downregulation of ACE, AGTR1, and ACE2 genes mediating SARS-CoV-2 pathogenesis by gut microbiota members and their postbiotics on Caco-2 cells

Summary: This study demonstrates the potential of A. muciniphila, F. prausnitzii, B. thetaiotaomicron, and B. fragilis and their postbiotics to reduce the expression of ACE, AGTR1, and ACE2 genes in COVID-19 patients through experimental and bioinformatic analyses. These findings suggest the importance of considering gut microbiota intervention when designing therapeutic strategies for COVID-19.

Summary: Probiotics such as A. muciniphila and its outer membrane vesicles (OMVs) play an important role in immunomodulation by regulating dendritic cell (DC) maturation and inducing tolerogenic DCs. These probiotics also affect the expression of microRNAs involved in inflammatory and anti-inflammatory pathways.

The effect of Bacteroides fragilis and its postbiotics on the expression of genes involved in the endocannabinoid system and intestinal epithelial integrity in Caco-2 cells

Amin Malek, Sara Ahmadi Badi, Gilda Karimi, Tania Bizouarn, Saeed Irian, Seyed Davar Siadat

Summary: This study investigated the effects of Bacteroides fragilis and its derivatives on the expression of genes related to the endocannabinoid system and the epithelial barrier permeability. The results showed that heat-inactivated B. fragilis induced the expression of cnr1, pparg, and tjp1 genes, while suppressing the faah gene expression. In contrast, live B. fragilis had the opposite effects. Outer membrane vesicles increased pparg and tjp1 gene expression by reducing the activity of the endocannabinoid system. Treatment with cell-free supernatants resulted in increased expression of pparg and ocln genes, and decreased expression of cnr1 gene. Overall, this study suggests that B. fragilis and its derivatives have potential beneficial effects on the permeability of the gut barrier.
