Summary: Polymyxin B (PMB) is a widely used polypeptide antibiotic for treating multidrug-resistant Gram-negative bacteria. However, its clinical use is limited due to nephrotoxicity. This study aimed to investigate the molecular mechanism of PMB-induced nephrotoxicity. The results showed that PMB induced oxidative stress damage by inhibiting the Nrf2/NQO1 pathway and promoting apoptosis in kidney tissues.

Summary: Our study investigated the cardioprotective effects of riboflavin in heart failure by activating SCAD and the DJ-1-Keap1-Nrf2 signaling pathway. In vivo, riboflavin improved cardiac function, reduced myocardial fibrosis, and inhibited oxidative stress and cardiomyocyte apoptosis. In vitro, riboflavin decreased reactive oxygen species and inhibited cell apoptosis. These effects were mediated through the restoration of FAD levels, SCAD expression, activation of DJ-1, and inhibition of the Keap1-Nrf2/HO1 pathway.

Summary: The study showed that Pgp3 promotes the translocation of Nrf2 and increases NQO1 expression to reduce oxidative stress induced by LPS, contributing to the survival of C. trachomatis. Inhibition of the Nrf2/NQO1 pathway or down-regulation of NQO1 weakened the oxidative stress resistance induced by Pgp3, indicating the important role of Pgp3 in the pathogenesis of C. trachomatis.

Summary: Doxorubicin, a drug with limited clinical application due to its cardiotoxicity, can be alleviated by Dimethyl fumarate (DMF) through reducing oxidative stress and apoptosis via Nrf2 pathway. The study found that DMF significantly improved cell viability and morphology, as well as alleviated DOX-induced cardiac injury, oxidative stress, and apoptosis. Moreover, DMF exerted its protective effects by promoting Nrf2 nuclear translocation and activating downstream antioxidant gene Hmox1. DMF may be a promising candidate to alleviate DOX-related cardiotoxicity in the future.

Summary: This study demonstrated that Sedanolide enhances cellular resistance to oxidative damage through the activation of the KEAP1-NRF2 pathway and upregulation of antioxidant enzymes. Additionally, Sedanolide attenuates cytosolic and mitochondrial ROS generation induced by H2O2, thereby exerting cytoprotective effects against H2O2-induced cell death.

Summary: This study investigates the role of Klotho in cardiac aging, revealing that Klotho deficiency causes cardiac aging through impairing the Nrf2-GR pathway. Supplement of exogenous secreted Klotho represents a promising therapeutic strategy for aging-associated cardiomyopathy and heart failure.

Summary: This study found that Pinot noir pomace extract contains malvidin-3-glucoside, a predominant anthocyanin. Anthocyanins can modify the interaction between Keap1 and Nrf2, without promoting Nrf2 transcription. In addition, the extract can regulate the expression of HO-1 and NQO1 genes, thus protecting cells against oxidative stress.

Summary: Sinomenine has been demonstrated to alleviate cardiac hypertrophy by activating the Nrf2/ARE signaling pathway, which leads to reduced cell area, decreased apoptosis rate, and regulation of gene and protein expression levels associated with cardiac hypertrophy. Further research is needed to confirm these findings in human subjects.
