An outlook on the trigeminovascular mechanisms of action and side effects concerns of some potential neuropeptidergic antimigraine therapies

Abimael Gonzalez-Hernandez, Miguel Condes-Lara, Enrique Garcia-Boll, Carlos M. Villalon

Summary: Neuropeptides play an important role in migraine pathophysiology, with molecules such as CGRP, PACAP, OT, and orexins showing potential as novel targets for treatment. However, targeting these neuropeptide systems may lead to both antimigraine effects and potential side effects due to their complex intracellular mechanisms. Pharmacokinetics issues may also limit the use of molecules targeting neuropeptidergic transmission.

Cortical Modulation of Nociception

Mohammed Gamal-Eltrabily, Guadalupe Martinez-Lorenzana, Abimael Gonzalez-Hernandez, Miguel Condes-Lara

Summary: Nociception modulation involves multiple cortical areas and mechanisms, including both descending modulation at spinal level and cortico-cortical or cortico-subcortical interactions. Understanding the involvement of these areas and the neurotransmitters/neuromodulators studied in each area is crucial for pain modulation.

CLARITY with neuronal tracing and immunofluorescence to study the somatosensory system in rats

Guadalupe Martinez-Lorenzana, Mohammed Gamal-Eltrabily, Irma Alejandra Tello-Garcia, Ataulfo Martinez-Torres, Marymar Becerra-Gonzalez, Abimael Gonzalez-Hernandez, Miguel Condes-Lara

Summary: The study showcases the application of CLARITY technique in visualizing neuronal connections and innervations in the nervous system in a transparent manner. Results indicate the reliability of CLARITY when combined with other techniques, providing valuable insights into neuroanatomical structures.

In Vivo Dissection of Two Intracellular Pathways Involved in the Spinal Oxytocin-Induced Antinociception in the Rat

Antonio Espinosa De los Monteros-Zuniga, Guadalupe Martinez-Lorenzana, Miguel Condes-Lara, Abimael Gonzalez-Hernandez

Summary: Behavioral and electrophysiological data demonstrate that oxytocin inhibits pain transmission at the spinal level by activating OTRs, which can mediate two distinct intracellular pathways - Gq (PLC/NOS/GC/K-ATP(+)) or Gi proteins, leading to differential effects on early and late nociceptive responses.

The impact of CGRPergic monoclonal antibodies on prophylactic antimigraine therapy and potential adverse events

Abimael Gonzalez-Hernandez, Bruno A. Marichal-Cancino, Carlos M. Villalon

Summary: Monoclonal antibodies act as prophylactic antimigraine agents by targeting CGRP or its receptors, revolutionizing the treatment of this disorder. Despite their long half-lives and hepato-friendly properties, potential cardiovascular side effects and immunogenicity induced by autoantibodies against mAbs present safety concerns for long-term use.

A critical review of the neurovascular nature of migraine and the main mechanisms of action of prophylactic antimigraine medications

Bruno A. Marichal-Cancino, Abimael Gonzalez-Hernandez, Raquel Guerrero-Alba, Roberto Medina-Santillan, Carlos M. Villalon

Summary: Migraine involves neurovascular, functional, and anatomical alterations, and there is no ideal preventive medication currently. However, some prophylactic medications may help decrease the frequency of migraine days. The mechanisms of action involved in antimigraine prophylaxis remain elusive.

The glial cell's role in antinociceptive differential effects of oxytocin upon female and male rats

Ana B. Salinas-Abarca, Francisco G. Vazquez-Cuevas, Adriana Gonzalez-Gallardo, Guadalupe Martinez-Lorenzana, Abimael Gonzalez-Hernandez, Miguel Condes-Lara

Summary: Sexual dimorphism plays a significant role in pain processing and response to analgesic drugs. Oxytocin showed a preference for preventing LPS-induced hypersensitivity in male rats compared to females, indicating a potential gender-specific analgesic effect. The results suggest that different strategies may be needed to treat pain in men and women, with oxytocin potentially being more effective for pain with an inflammatory component in men.

Ultrastructural Evidence for Oxytocin and Oxytocin Receptor at the Spinal Dorsal Horn: Mechanism of Nociception Modulation

Guadalupe Martinez-Lorenzana, Lourdes Palma-Tirado, Carmen Cifuentes-Diaz, Abimael Gonzalez-Hernandez, Miguel Condes-Lara

Summary: Oxytocin and its receptor play a role in analgesia at the spinal dorsal horn, possibly through a presynaptic mechanism or mediated by GABA interneurons. Additionally, the expression of oxytocin receptor in glial tissue at the spinal dorsal horn requires further investigation in nociception assays.

The differential in vivo contribution of spinal α2A- and α2C-adrenoceptors in tonic and acute evoked nociception in the rat

Gustavo Lopez-Cordoba, Guadalupe Martinez-Lorenzana, Jair Lozano-Cuenca, Miguel Condes-Lara, Abimael Gonzalez-Hernandez

Summary: Spinal alpha(2)-adrenoceptors induce analgesia by inhibiting primary afferent fibers. The alpha(2A) subtype is involved in acute or tonic nociception, while the alpha(2C) subtype has an opposing effect under tonic nociceptive input.

The role of the endocannabinoid 2-arachidonoylglycerol in the in vivo spinal oxytocin-induced antinociception in male rats

Ana B. Salinas-Abarca, Guadalupe Martinez-Lorenzana, Miguel Condes-Lara, Abimael Gonzalez-Hernandez

Summary: Activation of the oxytocin receptor (OTR) in the spinal cord can produce antinociception. It is suggested that central OTR activation enhances social interaction through an increase in endocannabinoids (eCB), but it's unclear if this also occurs at the spinal level, modulating pain transmission. This study aimed to investigate the role of the eCB system in spinal oxytocin-induced antinociception.

Serotonergic Modulation of Neurovascular Transmission: A Focus on Prejunctional 5-HT Receptors/Mechanisms

Abimael Gonzalez-Hernandez, Bruno A. Marichal-Cancino, Antoinette MaassenVanDenBrink, Carlos M. M. Villalon

Summary: 5-Hydroxytryptamine (5-HT), or serotonin, plays a crucial role as a neuromodulator and/or neurotransmitter in various nervous system functions. Its actions are complex and depend on multiple factors, including the type of effector or receptor activated. This review summarizes and discusses the impact of 5-HT on blood pressure regulation and vascular-related diseases.
