Ten lignans (1 - 10) were isolated from the hexane-ethyl acetate extract ofPhyllanthus amarusleaves. Three of them, cubebin dimethyl ether (3), urinatetralin (4), and lintetralin (7) are described for the first time in this species, while phyllanthin (1), niranthin (2), 5-demethoxyniranthin (5), isolintetralin (6), hypophyllanthin (8), nirtetralin (9), and phyltetralin (10) have been already reported fromP. amarus. Among the lignans tested againstTrypanosoma cruziintracellular amastigotes,2was the most active with an EC(50)of 35.28 mu M. Lignans2, 5, 7, and9showed inhibitory effects againstLeishmania amazonensispromastigotes with EC(50)of 56.34, 51.86, 23.57, and 43.27 mu M, respectively. Duringin vitroinfection assays,5reduced amastigotes by 91% at 103.68 mu M concentration, whereas7and9reduced amastigotes by approximately 84% at 47.5 and 86.04 mu M, respectively. Lignans5, 7, and9were more potent in intracellular amastigotes with EC(50)of 2.76, 8.30, and 15.83 mu M, respectively, than in promastigotes. CC(50)for all samples was > 100 mu g/mL, thus revealing low cytotoxicity against macrophages, and selectivity against the parasite.L. amazonensispromastigotes treated with compounds2and9showed decreased respiratory control of 38% and 25%, respectively, suggesting a change in mitochondrial membrane potential and lower ATP production.

Summary: This study aimed to find potential compounds inhibiting SARS-CoV-2 from extracts of Brazilian plants. Experimental validation showed that ethyl acetate fraction and F2, F4 sub-fractions of C. pluviosum extract exhibited good antiviral activity in infected cells. These results may be attributed to the synergistic effect of different hydrolyzable tannins present in the extract.

Potential of plant extracts in targeting SARS-CoV-2 main protease: an in vitro and in silico study

Summary: The deaths caused by covid-19 have decreased due to vaccination efforts, but severe cases still exist. Therefore, the search for antiviral drugs is crucial. Natural products, such as plant extracts, have shown potential in treating viral diseases. This study evaluated the antiviral activity of different plant extracts and found that Cytinus hypocistis extract had a high inhibition capacity against the SARS-CoV-2 main protease and viral infection in a cellular model. These results highlight the potential of C. hypocistis as a source of natural compounds with antiviral activity against covid-19.

Summary: The compound IVS320 was studied for its antifungal and antichagasic potential, but it has low aqueous solubility. To improve bioavailability, inclusion complexes with I3-cyclodextrin and methyl-I3-cyclodextrin were prepared using different methods. The inclusion complexes showed changes in physicochemical characteristics and decreased crystallinity compared to IVS320 alone. The IVS320 and IVS320-MI3CD/RE system exhibited anti-SARS-CoV-2 activity, and IVS320 showed interactions with the viral protein. The highest antichagasic activity was observed with IVS320-I3CD/KN and IVS320-MI3CD/PM.

Ecofriendly aminochalcogenation of alkenes: a green alternative to obtain compounds with potential anti-SARS-CoV-2 activity

Summary: Here, a solvent- and metal-free methodology for the aminochalcogenation of alkenes using molecular iodine as a catalyst, DMSO as a stoichiometric oxidant, and different nucleophiles under microwave irradiation is reported. The eco-friendly approach provided the desired products with good to excellent yields in just 20 minutes. Additionally, the antiviral activity of twenty compounds obtained by this methodology against the coronavirus SARS-CoV-2 was evaluated, and two compounds (4d and 5b) showed potent antiviral activity with low cytotoxicity, suggesting promising potential as antiviral agents.

Summary: Chalcones are chemical precursors of flavonoids that have various biological properties. In this study, novel copper(II)-2'-hydroxy-4-benzyloxychalcone complexes were synthesized and evaluated for their antileishmanial, antiproliferative, and antiviral activities. The results demonstrated that these complexes showed potential effects against tumor cells, leishmania parasites, and SARS-CoV-2 virus. The study also highlighted the influence of ligand structure on biological activity.

Synergism Between Essential Oils and Evaluation of Their Activities with a Focus on Malassezia furfur Control

Summary: This study evaluated the activity and anti-inflammatory potential of different essential oils against the pathogenic fungus of seborrheic dermatitis. A combination of Cymbopogon martini and Mentha x piperita essential oils showed synergistic effects against the fungus and exhibited activity against biofilm formation and eradication. The essential oil combination demonstrated no mutagenic potential and had no adverse effects on human cell viability. Therefore, it may serve as an alternative treatment for seborrheic dermatitis.

Biological characterization of Trypanosoma cruzi epimastigotes derived from trypomastigotes isolated from Brazilian chagasic patients

Summary: This study characterized the biochemical and mitochondrial bioenergetics of T. cruzi isolates obtained from Brazilian Chagas disease patients. The findings showed heterogeneity among the isolates in terms of growth characteristics, drug resistance, and energy source dependency. Mitochondrial bioenergetics analysis also revealed differences in the utilization of respiratory chain substrates among the isolates.
