Qualitative and quantitative determination of steroidal saponins in Trillium govanianum by UHPLC-QTOF-MS/MS and UHPLC-ELSD

Introduction Trillium govanianum (Nag Chhatri and Teen Patra) is traditionally used for curing joint pains, wounds, and sexual disorders. Steroidal saponins are the main active components of this species. However, only a small amount of information is available about steroidal saponins of this plant. Objective To develop an ultra-high-performance liquid chromatography-quadrupole time of flight tandem mass spectrometry (UHPLC-QTOF-MS/MS) and ultra-high-performance liquid chromatography-evaporative light scattering detector (UHPLC-ELSD) methods for the qualitative and quantitative determination of steroidal saponins in T. govanianum. Method The dried rhizomes of T. govanianum (100 mg) were extracted with ethanol-water (80:20, 10 mL) by ultrasonic treatment for 30 min at 40 degrees C. The prepared sample was analysed by UHPLC-QTOF-MS/MS and UHPLC-ELSD for the qualitative and quantitative determination of steroidal saponins. Result A total of 24 saponins were identified using UHPLC-QTOF-MS/MS; seven of them were characterised by comparing with standards. Furthermore, five saponins [govanoside B (2), protodioscin (6), pennogenin tetraglycosides (11), borassoside E (21) and borassoside D (24)] were quantified using UHPLC-ELSD method in different extracts and fractions of T. govanianum. The method showed good linearity (R-2 >= 0.993), limit of detection (0.92-4.09 mu g/mL), limit of quantification (3.1-13.5 mu g/mL), precision [intra-day relative standard deviations (RSDs) < 4.3% and inter-day RSDs < 5.5%], and accuracy (84.0-110.3%). This is the first report on the quantification of 2, 6, 11, 21 and 24 in T. govanianum. Conclusion The present study provides an efficient analytical method for the identification and quantification of steroidal saponins and will be helpful for the quality evaluation of T. govanianum.

Molecular docking and dynamic simulation approach to decipher steroidal sapogenins (genus Trillium) derived agonists for glucocorticoid receptor

Summary: In this study, we conducted in silico screening of steroidal sapogenins (SS) from the genus Trillium to identify potential agonists for the glucocorticoid receptor (GR). Our findings suggest that two SS molecules (SS 38 and SS 39) exhibit favorable binding to GR, as revealed by docking scores and MM-GBSA analysis. Molecular dynamics simulation studies further confirmed stable interactions between these SS molecules and GR. These findings indicate that SS 38 and SS 39 may have potential as agonists for GR-mediated diseases.

Comparative phytochemical analysis of Ferula assa-foetida with Ferula jaeschkeana and commercial oleo-gum resins using GC-MS and UHPLC-PDA-QTOF-IMS

Summary: This study conducted a detailed phytochemical analysis of F. assa-foetida in India and compared it with F. jaeschkeana and commercial samples of F. assa-foetida oleo-gum resin. Various compounds were identified and 141 metabolites were identified using the METLIN database. The nutritional analysis revealed major amino acids among the samples. Sulfurous compounds were identified in all samples except F. jaeschkeana. The study also showed significant differences in volatile constituents and revealed associations between biosynthetic pathways in these two species. This study explains why only F. assafoetida is used in culinary applications instead of F. jaeschkeana.

Multicomponent natural deep eutectic solvents: Super solvents for the efficient extraction of steviol glycosides (rebaudioside A) from Stevia rebaudiana

Summary: Using natural deep eutectic solvents (NADESs) as greener substitutes for organic solvents, the current study investigated the extraction of steviol glycosides from Stevia rebaudiana. The results showed that NADES 50 (lactic acid: glycerol: malic acid: glucose, 1:1:1:1) was the most efficient in extracting rebaudioside A and stevioside, while NADES 36 (choline chloride: lactic acid: oxalic acid: glycerol, 2:2:1:1) extracted all targeted analytes along with untargeted compounds. The study concluded that multicomponent NADESs have excellent extractability of steviol glycosides compared to binary NADESs.

Photoredox Minisci-Type Hydroxyfluoroalkylation of Isoquinolines with N-Trifluoroethoxyphthalimide

Summary: A photocatalytic approach has been used to introduce a trifluoroethanol unit onto isoquinolines by employing N-trifluoroethoxyphthalimide as a hydroxyfluoroalkyl radical precursor. The involvement of a free-radical pathway was confirmed by radical quenching experiments, and DFT calculations supported the intramolecular 1,2-HAT reactivity of the trifluoroethoxy radical with the trifluoroethanol radical. Fluorescence quenching studies revealed that isoquinoline was responsible for the quenching of Ir photocatalyst emission. A catalytic cycle involving the reaction of trifluoroethanol radical with isoquinolines was proposed.

Insecticidal potential of extracts, fractions, and molecules of Aconitum heterophyllum Wall ex. Royle against aphid Aphis craccivora Koch (Hemiptera: Aphididae)

Summary: In the current study, the residual toxicity of extracts, fractions, and isolated compounds of Aconitum heterophyllum were evaluated against Aphis craccivora. The ethanolic and aqueous methanolic extracts showed effective control against A. craccivora. Among the fractions, the n-butanol fraction of the aqueous methanolic extract was the most effective, followed by the ethyl acetate fraction of the ethanolic extract and the n-hexane fraction of both extracts. Aconitic acid was identified as the most effective pure molecule. The study also suggested that the action of the ethanoic extract might involve the inhibition of glutathione S-transferase in A. craccivora.

Experimental and Computational Studies on Ru-II-Catalyzed C7-Allylation of Indolines with Allyl Bromide

Summary: Selective C7-allylation of indolines with allyl bromide under ruthenium catalysis is demonstrated in this study. Good selectivity and yields were achieved for the C7-allylation of various indolines, including drug compounds, under established reaction conditions. Combined experimental and density functional theory (DFT) studies suggest that olefin insertion is the energetically favorable pathway among four possible routes. Furthermore, experimental and DFT studies reveal that C-H activation is a reversible rate-limiting step.

Govanosides C-F, unprecedented steroidal saponins with rare sugars from rhizomes of Trillium govanianum and their antagonistic effects on acetylcholinesterase

Summary: Four new compounds were isolated from the rhizomes of Trillium govanianum, including some rare sugar moieties. This is also the first report on the isolation of certain compounds from the Trillium genus, as well as feruloyl sucrose derivatives and (E)-4-hydroxy-dodec-2-enedioic acid. One of the compounds showed inhibitory activity against acetylcholinesterase.

NADES-based selective extraction of bioactive molecules: A case study with commercially important Himalayan medicinal plant Aconitum heterophyllum

Summary: In this study, natural deep eutectic solvents (NADESs) were used to extract bioactive compounds from the Himalayan medicinal herb Aconitum heterophyllum Wall. The most effective NADES-based solvent system was found to be NADES-15 [lactic acid: glycerol (1:1)], which showed higher extraction efficiency compared to conventional solvents. The efficiency of NADES-15 extraction was further improved by optimizing parameters such as solid: liquid ratio, extraction time, temperature, and water content. The recovered NADES-15 showed relatively lower extraction efficiency than the fresh counterpart, and molecular mechanism studies suggested that higher exergonic stability and intermolecular hydrogen bond interactions might contribute to the enhanced extraction efficiency of NADESs compared to conventional solvents.

Development and validation of an UHPLC-DAD method for simultaneous quantification of twenty-one diverse phenolics and its implementation in household food products

Summary: An ultra-high performance liquid chromatography coupled to a diode array detector (UHPLC-DAD) based method was developed for the simultaneous determination of 21 bioactive phenolic compounds in food products. The method was validated and showed ideal results, and was successfully applied for the quantitative analysis of phenolics in different targeted food samples.

Chemo selective C-H alkylation of isoquinolones with maleimides: A combined experimental and computational case study

Summary: A comparative study of C8-alkylation of isoquinolone with maleimides was conducted, and Ru and Rh catalysts were found to be excellent in providing desired products. Density functional theory (DFT) investigations and non-covalent interaction (NCI) analysis revealed the reaction mechanism and the selective formation of alkylated products. This catalytic methodology also showed a broad substrate scope and late-stage modification.

Flavonoids from Aerial Parts of Cissampelos pareira L. as Antimalarial Agents: Computational Validation of Ethnopharmacological Relevance

Summary: The diverse chemical scaffolds of Cissampelos pareira have pleiotropic therapeutical applications. The flavonoids in C. pareira have potential as antimalarial agents and showed good binding affinity with selected Plasmodium proteins. Molecular docking and molecular dynamics simulation revealed stable complexes between flavonoids and antimalarial targets. These findings suggest that C. pareira and its flavonoids could be developed as broad-spectrum antimalarial therapeutics.

Molecular links between metabolome and epigenome: AMPK-TET2 signalling pathway and their natural activators

Summary: Emerging evidence suggests that sustained diabetes-associated factors contribute to abnormal cell proliferation and neoplastic transformation. Epidemiological studies have also shown a link between diabetes and cancer, often associated with a sedentary lifestyle. However, our understanding of the molecular mechanisms connecting hyperglycemia and oncogenic transformation is limited. A recent study discovered that hyperglycemia inactivates AMPK, destabilizes TET2, and increases the risk of cancer in patients with diabetes. Utilizing natural products derived from plants may offer a pharmacological approach to managing hyperglycemia associated with cancer.
