Natural hydrogels R&D process: technical and regulatory aspects for industrial implementation

Since hydrogel therapies have been introduced into clinic treatment procedures, the biomedical industry has to face the technology transfer and the scale-up of the processes. This will be key in the roadmap of the new technology implementation. Transfer technology and scale-up are already known for some applications but other applications, such as 3D printing, are still challenging. Decellularized tissues offer a lot of advantages when compared to other natural gels, for example they display enhanced biological properties, due to their ability to preserve natural molecules. For this reason, even though their use as a source for bioinks represents a challenge for the scale-up process, it is very important to consider the advantages that originate with overcoming this challenge. Therefore, many aspects that influence the scaling of the industrial process should be considered, like the addition of drugs or cells to the hydrogel, also, the gelling process is important to determine the chemical and physical parameters that must be controlled in order to guarantee a successful process. Legal aspects are also crucial when carrying out the scale-up of the process since they determine the industrial implementation success from the regulatory point of view. In this context, the new law Regulation (EU) 2017/745 on biomedical devices will be considered. This review summarizes the different aspects, including the legal ones, that should be considered when scaling up hydrogels of natural origin, in order to balance these different aspects and to optimize the costs in terms of raw materials and engine.

Summary: A new high molecular weight aliphatic copolyester of PLLA with ABA triblock architecture was successfully synthesized via ring opening polymerization, with chain extension reaction performed to increase the polymer molecular weight. Annealing was found to be an efficient tool to control the crystalline phase and mechanical properties of the material.

Wide companions to M and L subdwarfs with Gaia and the Virtual Observatory

Summary: This study aims to identify wide common proper motion companions to a sample of M and L metal-poor dwarfs to investigate the impact of metallicity on binary fraction and to improve the determination of their metallicity. Through the use of Virtual Observatory tools and large-scale public surveys, a multiplicity rate for sdMs of 1.0±2.0% was found for projected physical separations up to 743,000 au. The study also found four M-L systems, with a frequency of wide systems for sdM5-9.5 of 0.60±1.17% for projected physical separations larger than 1,360 au.

Summary: Diseases like diabetes can lead to high sugar concentrations in the body, promoting the formation of glycoconjugates such as glycated collagen. Glucose and galactose produce similar changes in collagen, while ribose is faster in glycation and has qualitatively different effects on collagen structure.

A Gut-Ex-Vivo System to Study Gut Inflammation Associated to Inflammatory Bowel Disease (IBD)

Summary: Inflammatory Bowel Disease (IBD) is a complex, chronic, and dysregulated inflammatory condition with largely unknown etiology. The prognosis and disease progression are highly variable and unpredictable. IBD comprises various inflammatory conditions from Ulcerative Colitis (UC) to Crohn's Disease (CD). Effective clinical treatments for these pathologies are still lacking, emphasizing the urgent need for deeper understanding of the molecular mechanisms underlying IBD development and progression. The proposed ex vivo model using colon tissues from mice in a dynamic culture condition shows promise in efficiently studying the pathogenesis of IBD in a time- and cost-effective manner.

Regenerative Potential of A Bovine ECM-Derived Hydrogel for Biomedical Applications

Summary: Recent advancements in regenerative medicine have enhanced the development of biomaterials as multi-functional dressings. In this study, a bovine pericardium decellularized extracellular matrix (dECM) hydrogel was characterized and evaluated for its potential in wound healing. The results showed that the dECM hydrogel has a complex composition, is biocompatible, and has the ability to stimulate angiogenesis, drive cell polarization, promote fibroblast migration, and facilitate wound closure, demonstrating its great potential for regenerative applications.

Reaching the boundary between stellar kinematic groups and very wide binaries IV. The widest Washington Double Star systems with ρ ≥ 1000 arcsec in Gaia DR3

Summary: Using the latest Gaia DR3 data, this study analyzes the widest pairs in the Washington Double Star (WDS) catalogue with angular separations greater than 1000 arcsec. Based on common proper motions, parallaxes, and radial velocities, the study confirms the membership of these pairs to stellar systems. Additionally, the study searches for additional closer companions to these ultrawide pairs and determines masses for each star, calculating the gravitational potential energy of the systems. The results show that out of the 155,159 pairs catalogued by WDS, only 94 remain as bona fide ultrawide pairs in the galactic field.

Cardiac Differentiation Promotes Focal Adhesions Assembly through Vinculin Recruitment

Summary: Cells of the cardiovascular system are exposed to various mechanical forces that are crucial for cardiac development and functions. Focal adhesion (FA) complexes and actomyosin networks play vital roles in regulating cellular behaviors related to cytoskeleton dynamics, cell adhesion, migration, differentiation, and tissue organization. This study investigates the involvement of FA in cardiomyocyte differentiation, focusing on vinculin and focal adhesion kinase (FAK) family. The results show that differentiation conditions upregulate FAK-Tyr397 and vinculin, leading to their translocation to the cell membrane. Mechanical stretching also affects the cell shape and the length and number of vinculin-positive FAs, indicating the significant role of mechanotransductive pathways in cardiomyocyte differentiation and cytoskeleton rearrangement.

Summary: This study demonstrates, for the first time, the presence and functionality of a type 2 bitter receptor hTAS2R46 in human skeletal muscle cells. This finding suggests that bitter receptors may play an important role in the development, maintenance, and protection of muscle tissue functions.

Summary: The clinical demand for tissue-engineered vascular grafts is increasing, and there are many challenges to obtain functional small-diameter grafts. Advances in cell culture, biomaterials, manufacturing techniques, and tissue engineering methods have led to promising solutions for vascular graft production. The use of natural materials as scaffolds for vascular tissue engineering has gained attention and new strategies have been developed to overcome the drawbacks.
