Effects of Vibrio parahaemolyticus infection on physiological response, histopathology and transcriptome changes in the mud crab (Scylla paramamosain)

Mud crab (Scylla paramamosain) is an important economic species in China. Vibrio parahaemolyticus infection have caused a great economic loss in mud crab farming. The mechanism involved in the immune responses of mud crab to V. parahaemolyticus is unclear. In this study, the physiological and immune response to V. parahaemolyticus infection were investigated in S. paramamosain. The results showed that V. parahaemolyticus infection decreased total hemocyte counts, led to cytological damage, and caused high mortality. Transcriptome analysis showed that 1327 differentially expressed genes (DEGs), including 809 up-regulated and 518 downregulated ones, were obtained after V. parahaemolyticus challenge. These DEGs were mainly involved in the immune response and infectious disease. Additionally, transcriptome analysis revealed that Toll, immune deficiency (IMD), and prophenoloxidase signalling pathways played essential roles in antibacterial immunity against V. parahaemolyticus infection in mud crab.

Characterization of phosphofructokinase (PFK) from mud crab Scylla paramamosain and its role in mud crab dicistrovirus-1 proliferation

Summary: In this study, a PFK gene from the mud crab Scylla paramamosain, named SpPFK, was cloned and characterized. Knocking down SpPFK using RNA interference significantly reduced lactate dehydrogenase expression after MCDV-1 infection and increased the survival rate of mud crabs. These results suggest that SpPFK plays an important role in the proliferation of MCDV-1 in mud crabs.

Nutrients, temperature, and oxygen mediate microbial antibiotic resistance in sea bass (Lateolabrax maculatus) ponds

Summary: This study investigated the pollution of antibiotic resistance genes (ARGs) in sea bass ponds and its relationship with bacterial community composition and environmental parameters. The results showed that sea bass ponds are reservoirs of ARGs, and the changes are mainly influenced by water temperature, dissolved oxygen, and NO3-. Regulation of these environmental factors can reduce drug resistance risk in aquaculture ponds.

Federated Learning Over Multihop Wireless Networks With In-Network Aggregation

Summary: This paper investigates the problem of federated learning over wireless mesh networks and proposes a framework for in-network model aggregation to reduce outgoing data traffic and improve model aggregation under limited communication resources. The optimization problem is formulated by considering model aggregation, routing, and spectrum allocation, and a solution approach based on mixed-integer linear programming is developed. Simulation results demonstrate the effectiveness of the proposed solution and the superiority of the in-network aggregation scheme.

Summary: In this study, vaccines against Ranavirus-type grouper iridovirus (GIV-R) and/or Vibrio harveyi were developed and evaluated. The results showed that these vaccines provided effective protection against both pathogens. Additionally, booster immunization prolonged the duration of protection against V. harveyi infection.

Summary: In this study, a novel full-length of Glutaredoxin 5 (Sp-Grx 5) was cloned and characterized in the mud crab. Sp-Grx 5 was widely expressed in various tissues with high-level expression in hepatopancreas. It was found to be involved in innate immunity and detoxification, playing important roles in redox homeostasis and innate immune response in crustaceans.

Task Offloading in Multi-Hop Relay-Aided Multi-Access Edge Computing

Summary: As the demand for multi-access edge computing (MEC) increases exponentially, resource limitations at individual edge servers (ESs) become bottlenecks. This paper presents a cooperative MEC framework that enables network-wide cooperation of ESs through multi-hop task relaying. The framework balances the workload for both spot communication and computing, maximizing overall throughput. An optimization problem is formulated and matching theory is applied to find a low-complexity task offloading scheme. Numerical analysis demonstrates the effectiveness of the proposed scheme and the superiority of the multi-hop relay-assisted MEC systems.

Hypoxia triggers the proliferation of antibiotic resistance genes in a marine aquaculture system

Summary: The concentration of dissolved oxygen affects the spread of antibiotic resistance genes (ARGs). Low oxygen treatment increases the accumulation of ammonium and nitrite and the abundance of ARGs and mobile genetic elements (MGEs), especially ARGs resistant to drugs such as sulfonamide, chloramphenicol, and macrolides. This study highlights the importance of oxygen concentration in controlling the spread of ARGs, especially through horizontal gene transfer.

The global role of Lrp in Vibrio alginolyticus and its response to diverse physicochemical factors

Essential role of the Cytochrome P450 2 (CYP2) in the mud crab Scylla paramamosain antioxidant defense and immune responses

Summary: A novel cytochrome P450 enzyme, Sp-CYP2, was cloned and characterized in the mud crab. Sp-CYP2 was found to be widely expressed in various tissues, with the highest expression in the heart and hepatopancreas. Its expression was induced by ammonia exposure and infection of Vibrio parahaemolyticus. Knocking down Sp-CYP2 increased oxidative stress and mortality in mud crabs, suggesting its crucial role in defense against environmental stress and pathogen infection in crustaceans.

First insight into how stress exposure triggers Vibrio harveyi recipient successful conjugation

Summary: This study systematically analyzed the effect of environmental stress on the conjugation ability of the marine fish pathogen Vibrio harveyi. The results showed that certain stress exposure can induce the fertility of V. harveyi and promote the transfer of shuttle plasmids from Escherichia coli. Ethanol and heat stress exhibited the strongest induction effect. These findings will contribute to the development of gene knockout technology and advance the molecular theoretical research on V. harveyi, as well as support the establishment of disease prevention and control strategies based on environmental regulation of gene transfer.

Prevalence and distribution of antibiotic resistance in the water environment of sea bass (Lateolabrax maculatus) breeding area in spring in South China

Summary: This study investigated the abundance and distribution of bacterial antibiotic resistance associated with farmed fish in Zhuhai, China. The results showed high resistance to erythromycin and sulfamethoxazole with trimethoprim, while resistance to other antibiotics was generally low. Acinetobacter sp. and Pseudomonas sp. were the dominant antibiotic resistant bacteria, and most cultivable resistant bacteria were opportunistic pathogens. The abundance of sul family genes was higher than other tested resistance genes, and the presence of antibiotics in the water positively correlated with antibiotic resistance genes and bacteria.
