Oral cancer, a universal malady, has become a stumbling block over the years due to its significant morbidity and mortality rates. The greater morbidity associated with this deadly disease is attributed to delay in its diagnosis / its presentation in advanced stage. Being multifactorial, Oral squamous cell carcinoma (OSCC) is the outcome of genetic and epigenetic instability. However, in many instances, oral cancer is preceded by precursor lesions named as oral potentially malignant disorders (OPMDs), the early detection of which makes it beneficial for patients with the possible increase in the productive longevity. Many diagnostic tools / aids have been explored with the aim of early detection of oral precancer and cancer. The basic chair-side procedures or relatively advanced aids come with a set of limitations along with subjectivity as one of the setbacks. The advent and exploitation of molecular techniques in the field of health diagnostics, is demanding the molecular typing of the OPMDs and also of oral cancer. The saga of various diagnostic aids for OSCC has witnessed the so-called latest trends such as lab-on-chip, microfluidics, nano diagnostics, liquid biopsy, omics technology and synthetic biology in early detection of oral precancer and cancer. Oral cancer being multifactorial in origin with the chief participation of altered genetics and epigenetics would demand high-end diagnostics for designing personalized therapy. Hence, the present paper highlights the role of various advanced diagnostic aids including 'omics' technology and synthetic biology in oral precancer and cancer. (C) 2020 Elsevier Inc. Summary: Oral immune-mediated disorders are a group of oral diseases or conditions that may result from an abnormality in the individual's normal immune response. Some of these disorders are associated with malignancy. This article provides an overview of several oral diseases, such as oral lichen planus, primary Sjogren's syndrome, systemic lupus erythematosus, dermatitis herpetiformis, and linear immunoglobulin A bullous dermatosis, which are caused by irregularities in the immune system and have the potential to undergo malignant transformation, thereby increasing morbidity and mortality rates.

The effect of clear aligners on speech: a systematic review

Summary: A systematic review of the literature revealed that clear aligners can have an impact on patients' speech, causing pronunciation errors and speech difficulties. However, these issues were temporary and most patients adapted to the aligners and recovered their normal speech.

Summary: This study investigated the binding affinity and inhibitory activity of resveratrol and its two natural analogues against the Rhizopus oryzae specific RdRp through computer simulations. The results suggest that these compounds could be potential candidates for the treatment of mucormycosis.

Sulfate-Reducing Bacteria in Patients Undergoing Fixed Orthodontic Treatment

Summary: This study aimed to examine the prevalence of sulfate-reducing bacteria (SRB) in orthodontic patients undergoing fixed appliance treatment. The results showed that the prevalence of SRB was significantly higher in orthodontic patients compared to non-orthodontic participants. This may be attributed to the presence of stainless steel in the oral environment facilitating the colonization of SRB.

Red-Complex Bacterial Levels in Pregnant Women With Preeclampsia and Chronic Periodontitis

Summary: This study aimed to explore the levels of periodontal red-complex bacteria (RCB) in pregnant women with periodontal disease and its association with preeclampsia. The results showed that preeclamptic women with periodontitis had higher periodontal indices, blood pressure, and urine protein content. Therefore, reducing the periodontal bacterial load in pregnant women through good oral hygiene maintenance can further decrease the risk of adverse pregnancy outcomes, including preeclampsia.

Lysosome signaling in cell survival and programmed cell death for cellular homeostasis

Summary: Recent studies have shown that lysosomes, traditionally thought of as static garbage disposals, play a dynamic and multi-functional role in maintaining cellular homeostasis. Lysosomes sense cellular metabolic status and regulate the balance between catabolism and anabolism through signaling pathways, proteins, and transcription factors. Lysosomes also interact with other organelles and their positioning and motility are critical for cellular adaptation. Furthermore, lysosomes can orchestrate different cell death mechanisms to maintain cellular homeostasis, highlighting their central role as a signaling hub.

Summary: This study aimed to compare the effectiveness of AmnioGuard and BioMesh in combination with NovaBone putty in intrabony defects. The results showed that at 6 months post-operatively, Group B had a statistically significant change in both clinical and radiographic measures (33% bone gain) compared to Group A (16% bone gain).

Summary: Patients with prolonged edentulism face challenges in replacing missing teeth. The use of non-resorbable and resorbable membranes in guided bone regeneration showed similar efficacy. However, further research is needed to draw conclusive results due to the low quality of evidence.

CLU (clusterin) promotes mitophagic degradation of MSX2 through an AKT-DNM1L/Drp1 axis to maintain SOX2-mediated stemness in oral cancer stem cells

Summary: This study found that cisplatin treatment activated higher mitophagy in oral cancer stem cells by regulating CLU levels. CLU regulated mitochondrial fission by activating AKT kinase, leading to mitochondrial fission. Furthermore, CLU-mediated mitophagy positively regulated oral cancer stem cells by degrading MSX2 and preventing its nuclear translocation, which suppressed SOX2 activity and inhibited cancer stemness and self-renewal ability.

Efficacy of Two Different Hydrodynamic Sinus Lift Systems for Atraumatic Elevation in Immediate Implant Placement

Summary: The study aimed to compare the efficacy of two hydrodynamic sinus lifting procedures in placing immediate implants in maxillary posterior sites with compromised native bone. The MIAMBE group had lower rates of sinus membrane perforations and nasal bleeding, as well as lower VAS scores, compared to the DIHSFE group. Post-operative sinusitis rates did not differ significantly between the two groups. Therefore, MIAMBE appears to have superior patient outcomes and fewer complications compared to DIHSFE.

Estimation of GCF and Salivary Levels of NT-proBNP in Systemically Healthy Subjects with Severe Chronic Periodontitis Before and After Periodontal Flap Surgery

Summary: The purpose of this study was to estimate and compare the levels of NT-proBNP in saliva and GCF in systemically healthy subjects with severe chronic periodontitis before and after periodontal flap surgery. The results showed that NT-proBNP levels were higher in the periodontitis group compared to the controls, and they decreased following surgical periodontal therapy. The findings suggest that NT-proBNP could serve as a potential biomarker for periodontitis in saliva and GCF.

Efficacy of artificial intelligence in the detection of periodontal bone loss and classification of periodontal diseases A systematic review

Summary: This systematic review evaluated the efficacy of AI models in detecting radiographic periodontal bone loss (PBL) and accuracy in classifying lesions. The results suggest that AI has the potential to aid in the detection of PBL and classification of periodontal diseases, but further research is needed to standardize AI algorithms and validate their clinical usefulness.

Exosomes in cancer therapy: Advances and current challenges

Recent research also reveals the potential role of exosomes in anticancer drug delivery and their future orientation in theranostics perspectives.
