Summary: In this study, we examined the changes in chromatin accessibility and transcriptional programs during the differentiation of cochlear hair cells from postmitotic supporting cells using organoids. We identified Lgr5+ cells as the progenitors for hair cells and revealed the gene regulatory networks involved in their development. The analysis also predicted the involvement of additional regulatory factors in hair cell regeneration. This study provides important insights into the signals for hair cell regeneration from mammalian supporting cells.

The review concludes by exploring interventions that may restore EPCs expression and function in frail individuals.

Summary: Alveolar epithelial type 1 (AT1) cells are necessary for gas exchange, while alveolar epithelial type 2 (AT2) cells play a role in lung development and repair. In this study, deleting a specific subunit of the mitochondrial electron transport chain complex I in lung epithelial cells led to abnormal alveolar development and death. Restoring NAD(+) regeneration was able to correct the developmental defects and prevent lethality. These findings highlight the importance of mitochondrial complex I-dependent NAD(+) regeneration in directing cell fate during lung development.

Summary: For the first time, this study explores the hematopoietic and nonhematopoietic roles of p66Shc in regulating HSPC traffic and ischemic responses. Hematopoietic deletion of p66Shc is sufficient to rescue HSPC mobilization and homing in diabetes after ischemia and improves blood flow recovery. Inhibiting p66Shc in blood cells may be a novel strategy to counter PAD in diabetes.

Summary: Bone fractures are a major health burden worldwide, especially among the elderly population. The risk and frequency of bone fractures increase significantly with age, primarily due to the loss of skeletal integrity and reduced bone regeneration potential. Skeletal stem/progenitor cells (SSPCs) play a central role in these processes, but their exact identity and functions during aging remain unclear. This review discusses recent findings on SSPCs identification, their involvement in normal bone regeneration, and how aging affects their regeneration potential, leading to age-associated bone fractures and impaired repair. Novel perspectives based on the authors' experience are also provided.
